    Clinical Training in Explicitly Christian Doctoral Programs: Introduction to the Special Issue

    This article serves as a brief introduction to this special journal issue on clinical training in doctoral psychology programs described as explicitly Christian. Seven doctoral programs are described by their respective Directors of Clinical Training (DCT) in this special issue. In this introductory paper, we identify four common themes found across the program descriptions: 1) the enthusiasm for respective training missions, 2) the transformation observed in students, 3) the intentional progression of training, and 4) the varied meanings of the term integration

    The importance of research training at explicitly Christian doctoral programs is suggested on the basis of four reasons: the need to hold science and practice together, the need for skills to evaluate interventions, the need to serve a society with a growing interest in religion and spirituality, and the need to assess the effectiveness of explicitly Christian doctoral programs. A discussion of these issues serves as an introduction to the rest of this special issue that focuses on research training at seven explicitly Christian programs

    Satisfaction with Research Training in Christian Psychology Doctoral Programs: Survey Findings and Implications

    Perceptions of research training at seven explicitly Christian doctoral programs in clinical psychology were assessed with a satisfaction survey. A total of 283 students, 98 alumni, and 51 faculty completed the online questionnaire. Perceived strengths include faculty-student collaboration, encouraging students to present at national meetings, respect for faculty, and effective curricula. Areas for future growth include enhanced research funding, making research mentoring available to all students, and decreasing the perceived dichotomy between clinical and research competence. Self-reported faculty and student publication and presentation rates are also presented and discussed

    Murray Valley Encephalitis Virus Envelope Protein Antigenic Variants with Altered Hemagglutination Properties and Reduced Neuroinvasiveness in Mice

    AbstractNeutralization escape variants of Murray Valley encephalitis virus were selected using a type-specific, neutralizing, and passively protective anti-envelope protein (E) monoclonal antibody (4B6C-2) which defines epitope E-1c. Nucleotide sequence analysis revealed single nucleotide changes in the E genes of 15 variants resulting in nonconservative amino acid substitutions in all cases. One variant had a three-nucleotide deletion in the E gene which resulted in loss of serine at residue 277. Changes were clustered into two separate regions of the E polypeptide (residues 126-128 and 274-277), indicating that E-1c is a discontinuous epitope. One variant (BHv1), altered at residue 277 (Ser → Ile), failed to hemagglutinate across the pH range 55-7.5, in contrast to parental virus and the other escape variants which hemagglutinated at an optimal pH of 6.6. BHv1 was also of reduced neuroinvasiveness in 21-day-old mice following intraperitoneal inoculation compared to the other viruses. Parental virus and the neutralization escape variants grew equally well in both vertebrate and invertebrate cell cultures, indicating that the reduced neuroinvasiveness of BHv1 was not due to a major abnormality of replication

    Spiritual Formation among Doctoral Psychology Students in Explicitly Christian Programs

    How does training in an explicitly Christian doctoral program in clinical psychology affect students\u27 faith development? Two studies are reported that consider students\u27 locus of control, spiritual perceptions, and religious behaviors over the course of training. The first study involved 157 students from 5 doctoral programs who completed questionnaires at the beginning and end of an academic year. A number of changes were reported from the beginning to the end of the year, including increased internal locus of control, decreased awareness of God. decreased church attendance, and decreased ratings on the importance of religion. A number of differences between cohorts were also observed, with flrst-year students affirming more spiritual attributions, religious problem-solving, and religious behaviors than students in other cohorts. The second study included 140 first- and second-year students from 4 doctoral programs. Changes were reported over the academic year, including increased disappointment with God and fatigue, and decreased church attendance, personal prayer, and importance of religion. No differences between first and second-year students were observed. Various possible explanations are offered for these findings, including eroding of faith, enhanced self-efficacy, rearraging faith, and fatigue

    Mapping genetic determinants of the cell-culture growth phenotype of enterovirus 71

    Enterovirus 71 (EV71) is a member of the species Human enterovirus A within the family Picornaviridae and is a major causative agent of epidemics of hand, foot and mouth disease associated with severe neurological disease. Three EV71 genogroups, designated A, B and C, have been identified, with 75–84 % nucleotide sequence similarity between them. Two strains, EV71-26M (genogroup B) and EV71-6F (genogroup C), were found to have distinct cell-culture growth (26M, rapid; 6F, slow) and plaque-formation (26M, large; 6F, small) phenotypes. To identify the genome regions responsible for the growth phenotypes of the two strains, a series of chimeric viruses was constructed by exchanging the 5′ untranslated region (UTR), P1 structural protein or P2/P3 non-structural protein gene regions plus the 3′UTR using infectious cDNA clones of both virus strains. Analysis of reciprocal virus chimeras revealed that the 5′UTRs of both strains were compatible, but not responsible for the observed phenotypes. Introduction of the EV71-6F P1 region into the EV71-26M clone resulted in a small-plaque and slow-growth phenotype similar to that of EV71-6F, whereas the reciprocal chimera displayed intermediate-growth and intermediate-sized plaque phenotypes. Introduction of the EV71-26M P2–P3–3′UTR regions into the EV71-6F clone resulted in a large-plaque and rapid-growth phenotype identical to that of strain EV71-26M, whereas the reciprocal chimera retained the background strain large-plaque phenotype. These results indicate that, although both the P1 and P2–P3–3′UTR genome regions influence the EV71 growth phenotype in cell culture, phenotype expression is dependent on specific genome-segment combinations and is not reciprocal

    Human Placental-Specific Epipolymorphism and its Association with Adverse Pregnancy Outcomes

    Interindividual variation in DNA-methylation level is widespread in the human genome, despite its critical role in regulating gene expression. The nature of this variation, including its tissue-specific nature, and the role it may play in human phenotypic variation and disease is still poorly characterized. The placenta plays a critical role in regulating fetal growth and development in ways that have lifelong effects on health. To identify genes with a high degree of interindividual DNA methylation variation in the human placenta, we surveyed the human genome using the Illumina GoldenGate Methylation Cancer panel targeting 1505 CpG sites of 807 genes. While many sites show a continuous pattern of methylation levels, WNT2, TUSC3 and EPHB4 were identified to have a polymorphic “on-or-off” pattern of DNA methylation variation at their promoter region which was confirmed by pyrosequencing. Methylation of these genes can be found in 7%–25% of over 100 placentas tested. The methylation state at the promoter of these genes is concordant with mRNA allelic expression. In three informative cases TUSC3 was observed to be methylated on the maternal allele, and it is thus possible this represents a polymorphically imprinted gene. Furthermore, TUSC3 promoter methylation showed evidence for association with preeclampsia. A biological significance of these methylation allelic polymorphisms (MAPs) to human placental diversity is further implied by their placental specificity and absence in mouse. An extended study of blood suggests that MAPs may also be found in other tissues, implicating their utility for tissue-specific association with complex disorders. The identification of such “epipolymorphism” in other tissues and their use in association studies, should improve our understanding of interindividual phenotypic variability and complex disease susceptibility

    The Molecular Epidemiology and Evolution of Murray Valley Encephalitis Virus: Recent Emergence of Distinct Sub-lineages of the Dominant Genotype 1

Background: Recent increased activity of the mosquito-borne Murray Valley encephalitis virus (MVEV) in Australia has renewed concerns regarding its potential to spread and cause disease. Methodology/Principal Findings: To better understand the genetic relationships between earlier and more recent circulating strains, patterns of virus movement, as well as the molecular basis of MVEV evolution, complete pre-membrane (prM) and Envelope (Env) genes were sequenced from sixty-six MVEV strains from different regions of the Australasian region, isolated over a sixty year period (1951–2011). Phylogenetic analyses indicated that, of the four recognized genotypes, only G1 and G2 are contemporary. G1 viruses were dominant over the sampling period and found across the known geographic range of MVEV. Two distinct sub-lineages of G1 were observed (1A and 1B). Although G1B strains have been isolated from across mainland Australia, Australian G1A strains have not been detected outside northwest Australia. Similarly, G2 is comprised of only Western Australian isolates from mosquitoes, suggesting G1B and G2 viruses have geographic or ecological restrictions. No evidence of recombination was found and a single amino acid substitution in the Env protein (S332G) was found to be under positive selection, while several others were found to be under directional evolution. Evolutionary analyses indicated that extant genotypes of MVEV began to diverge from a common ancestor approximately 200 years ago. G2 was the first genotype to diverge, followed by G3 and G4, and finally G1, from which subtypes G1A and G1B diverged between 1964 and 1994. Conclusions/Significance: The results of this study provides new insights into the genetic diversity and evolution of MVEV. The demonstration of co-circulation of all contemporary genetic lineages of MVEV in northwestern Australia, supports the contention that this region is the enzootic focus for this virus

    Evaluating the Effects of SARS-CoV-2 Spike Mutation D614G on Transmissibility and Pathogenicity.

    Global dispersal and increasing frequency of the SARS-CoV-2 spike protein variant D614G are suggestive of a selective advantage but may also be due to a random founder effect. We investigate the hypothesis for positive selection of spike D614G in the United Kingdom using more than 25,000 whole genome SARS-CoV-2 sequences. Despite the availability of a large dataset, well represented by both spike 614 variants, not all approaches showed a conclusive signal of positive selection. Population genetic analysis indicates that 614G increases in frequency relative to 614D in a manner consistent with a selective advantage. We do not find any indication that patients infected with the spike 614G variant have higher COVID-19 mortality or clinical severity, but 614G is associated with higher viral load and younger age of patients. Significant differences in growth and size of 614G phylogenetic clusters indicate a need for continued study of this variant
