    Geological and geophysical characterization of the Brindisi di Montagna Scalo landslide (Basilicata, Southern Italy)

    The Brindisi di Montagna Scalo Landslide in Southern Italy is an active complex mass movement, which affects the left slope of the Basento River. In the last few decades, this landslide has been continuously monitored, as it directly threatened some of the most important communication routes in the Basilicata Region. Nevertheless, little progresses have been made to prevent further landslide advancement, and continuous maintenance is required. With the aims of better understanding, the main factors controlling the evolution of this landslide, and suggesting the most appropriate countermeasures, a multidisciplinary study, based on the integration of direct and indirect techniques, was carried out. Direct techniques included multi-temporal geomorphological analysis of the slope, alongside geological and structural field observations. Indirect techniques consisted of electrical resistivity tomography acquisition. The combined analyses of the geological and geophysical data showed that Quaternary tectonic processes played a fundamental role as a predisposing factor, whereas seasonal rainfall, and the perpetual undercutting by erosional processes caused by the Basento River at the toe of the landslide are the main triggering mechanisms. The Brindisi di Montagna Scalo Landslide represents an outstanding case-study, concerning the interaction between a flow-like complex landslide and essential linear infrastructure, such as motorways and railways

    Two new rhamnocitrin glycosides (1 and 2) were isolated from the aerial parts of Astragalus vogelii, along with one known rhamnocitrin glycoside (3). Two known flavonol glycosides (4 and 5) and four known flavone derivatives (6-9) were isolated from the aerial parts of Astragalus eremophilus. Their structures were elucidated by extensive spectroscopic methods including 1D- (1H, 13C and TOCSY) and 2D-NMR (DQF-COSY, HSQC, HMBC) experiments, as well as ESIMS analysis

    Refractory vasculitic ulcer of the toe in adolescent suffering from Systemic Lupus Erythematosus treated successfully with hyperbaric oxygen therapy

    Skin ulcers are a dangerous and uncommon complication of vasculitis. We describe the case of a teenager suffering from Systemic Lupus Erythematosus with digital ulcer resistant to conventional therapy, treated successfully with Hyperbaric Oxygen Therapy. The application of hyperbaric oxygen, which is used for the treatment of ischemic ulcers, is an effective and safe therapeutic option in patients with ischemic vasculitic ulcers in combination with immunosuppressive drugs. Further studies are needed to evaluate its role as primary therapy for this group of patients

    A Prototype System for Time-Lapse Electrical Resistivity Tomographies

    A prototype system for time-lapse acquisition of 2D electrical resistivity tomography (ERT) and time domain reflectometry (TDR) measurements was installed in a test site affected by a landslide in Basilicata region (southern Italy). The aim of the system is to monitor in real-time the rainwater infiltration into the soil and obtain information about the variation of the water content in the first layers of the subsoil and the possible influence of this variation on landslide activity. A rain gauge placed in the test site gives information on the rainfall intensity and frequency and suggests the acquisition time interval. The installed system and the preliminary results are presented in this paper

    Cystatin B Involvement in Synapse Physiology of Rodent Brains and Human Cerebral Organoids

    Cystatin B (CSTB) is a ubiquitous protein belonging to a superfamily of protease inhibitors. CSTB may play a critical role in brain physiology because its mutations cause progressive myoclonic epilepsy-1A (EPM1A), the most common form of progressive myoclonic epilepsy. However, the molecular mechanisms underlying the role of CSTB in the central nervous system (CNS) are largely unknown. To investigate the possible involvement of CSTB in the synaptic plasticity, we analyzed its expression in synaptosomes as a model system in studying the physiology of the synaptic regions of the CNS. We found that CSTB is not only present in the synaptosomes isolated from rat and mouse brain cortex, but also secreted into the medium in a depolarization-controlled manner. In addition, using biorthogonal noncanonical amino acid tagging (BONCAT) procedure, we demonstrated, for the first time, that CSTB is locally synthesized in the synaptosomes. The synaptic localization of CSTB was confirmed in a human 3D model of cortical development, namely cerebral organoids. Altogether, these results suggest that CSTB may play a role in the brain plasticity and open a new perspective in studying the involvement of CSTB deregulation in neurodegenerative and neuropsychiatric diseases

    Spray-dried mucoadhesives for intravesical drug delivery using N-acetylcysteine- and glutathione-glycol chitosan conjugates

    This work describes N-acetylcysteine (NAC)- and glutathione (GSH)-glycol chitosan (GC) polymer conjugates engineered as potential platform useful to formulate micro-(MP) and nano-(NP) particles via spray-drying techniques. These conjugates are mucoadhesive over the range of urine pH, 5.0-7.0, which makes them advantageous for intravesical drug delivery and treatment of local bladder diseases. NAC- and GSH-GC conjugates were generated with a synthetic approach optimizing reaction times and purification in order to minimize the oxidation of thiol groups. In this way, the resulting amount of free thiol groups immobilized per gram of NAC-and GSH-GC conjugates was 6.3 and 3.6 mmol, respectively. These polymers were completely characterized by molecular weight, surface sulfur content, solubility at different pH values, substitution and swelling degree. Mucoadhesion properties were evaluated in artificial urine by turbidimetric and zeta (zeta)-potential measurements demonstrating good mucoadhesion properties, in particular for NAC-GC at pH 5.0. Starting from the thiolated polymers, MP and NP were prepared using both the Bidchi B-191 and Nano Buchi B-90 spray dryers, respectively. The resulting two formulations were evaluated for yield, size, oxidation of thiol groups and ex-vivo mucoadhesion. The new spray drying technique provided NP of suitable size (<1 mu m) for catheter administration, low degree of oxidation, and sufficient mucoadhesion property with 9% and 18% of GSH- and NAC-GC based NP retained on pig mucosa bladder after 3 h of exposure, respectively

    A new joint application of non-invasive remote sensing techniques for structural health monitoring

    This paper aims at analysing the potentialities of a new technological approach for the dynamic monitoring of civil infrastructures. The proposed approach is based on the joint use of a high-frequency thermal camera and a microwave radar interferometer to measure the oscillations due to traffic excitations of the Sihlhochstrasse Bridge, Switzerland, which was selected as test bed site in the ISTIMES project (EU - Seventh Framework Programme). The good quality of the results encourages the use of the proposed approach for the static and dynamic investigation of structures and infrastructures. Moreover, the remote sensing character of the two applied techniques makes them particularly suitable to study structures located in areas affected by natural hazard phenomena, and also to monitor cultural heritage buildings for which some conventional techniques are considered invasive. Obviously, their reliability needs further experiments and comparisons with standard contact sensors

    A Novel PET Imaging Probe for the Detection and Monitoring of Translocator Protein 18 kDa Expression in Pathological Disorders

    A new fluorine-substituted ligand, compound 1 (CB251), with a very high affinity (Ki = 0.27 ± 0.09 nM) and selectivity for the 18-kDa translocator protein (TSPO), is presented as an attractive biomarker for the diagnosis of neuroinflammation, neurodegeneration and tumour progression. To test compound 1 as a TSPO PET imaging agent in vivo, 2-(2-(4-(2-[18F]fluoroethoxy)phenyl)-6,8-dichloroimidazo[1,2-a]pyridin-3-yl)-N,N-dipropylacetamide ([18F]1; [18F]CB251) was synthesized by nucleophilic aliphatic substitution in a single-step radiolabelling procedure with a 11.1 ± 3.5% (n = 14, decay corrected) radiochemical yield and over 99% radiochemical purity. In animal PET imaging studies, [18F]CB251 provided a clearly visible image of the inflammatory lesion with the binding potential of the specifically bound radioligand relative to the non-displaceable radioligand in tissue (BPND 1.83 ± 0.18), in a neuroinflammation rat model based on the unilateral stereotaxic injection of lipopolysaccharide (LPS), comparable to that of [11C]PBR28 (BPND 1.55 ± 0.41). [18F]CB251 showed moderate tumour uptake (1.96 ± 0.11%ID/g at 1 h post injection) in human glioblastoma U87-MG xenografts. These results suggest that [18F]CB251 is a promising TSPO PET imaging agent for neuroinflammation and TSPO-rich cancers
