Differential Astrometry over 15 degrees
We observed the pair of radio sources 1150+812 and 1803+784 in November 1993
with a VLBI array, simultaneously recording at 8.4 and 2.3 GHz. We determined
the angular separation between the two sources with submilliarcsecond accuracy
by using differential techniques. This result demonstrates the feasibility of
high precision differential astrometry for radio sources separated in the sky
by almost 15 degrees, and opens the avenue to its application to larger samples

Retromer deficiency in amyotrophic lateral sclerosis
The retromer is a protein complex whose function is to mediate the recycling of proteins from the endosome to either the plasma membrane or the trans-Golgi network. A deficit in retromer function has been associated with multiple neurodegenerative disorders, including Alzheimer’s disease (AD) and Parkinson’s disease (PD). In both AD and PD, deficiencies have been found in retromer expression both in patient tissues and in animal models of disease. Furthermore, mutations in the retromer and in retromer-associated genes have been strongly linked with both diseases. Despite ample evidence of the link between the retromer and neurodegeneration, little is known about the retromer in the context of amyotrophic lateral sclerosis (ALS), another common neurodegenerative disorder. ALS is an adult-onset neurodegenerative disorder of the upper and lower motor neurons (MNs) characterized by muscle wasting and weakness leading to death within 3-5 years after diagnosis. To date, the most commonly used model of ALS is a transgenic (Tg) mouse that overexpresses an ALS-causing G93A mutation in the human superoxide dismutase 1 (SOD1) gene. In this study, I first establish a link between the retromer and ALS by showing that cells from ALS patients as well as tissues and cells from SOD1G93A-Tg mice express lower protein levels of the retromer core components—vacuolar protein sorting 35 (Vps35), Vps26a, and Vps29. I then establish that deficiencies in retromer core proteins have functional consequences in an in vitro model of ALS. Having found significant deficiencies in the retromer in SOD1G93A-Tg mice, I then followed the model of studies performed in mouse models of other neurodegenerative disorders by investigating whether repletion of retromer levels, either virally or pharmacologically, in SOD1G93A-Tg mice confers a therapeutic benefit. Surprisingly, I find that rather than ameliorating disease, repletion of retromer levels in SOD1G93A-Tg mice exacerbates it, resulting in a faster decline in motor performance, earlier mortality, and a decrease in MNs in the spinal cord. Finally, since retromer repletion causes deleterious effects on SOD1G93A-Tg mouse disease progression, I study the effect of a single allele deletion of Vps35 in SOD1G93A-Tg mice and find that this depletion of the retromer results in amelioration of disease, including delayed onset of symptomatology, slower decline of motor deficits, delayed mortality, and an increase in MNs in the spinal cord. Altogether, the findings reported herein, support the notion that a mild defect in retromer develops over the course of the disease, which, rather than being deleterious may be therapeutic in mutant SOD1-induced MN degeneration. Perhaps this unexpected outcome may be explained by the fact that the observed mild nature of the defect is not sufficient to kill MNs but enough to alter the trafficking of specific cargos such as AMPA receptors, allowing MNs to better withstand the neurodegenerative process
Multi-wavelength differential astrometry of the S5 polar cap sample
We report on the status of our S5 polar cap astrometry program. Since 1997 we
have observed all the 13 radio sources of the complete S5 polar cap sample at
the wavelengths of 3.6 cm, 2 cm and 7 mm. Images of the radio sources at 3.6
and 2 cm have already been published reporting morphological changes.
Preliminary astrometric analyses have been carried out at three frequencies
with precisions in the relative position determination ranging from 80 to 20
microarcseconds. We report also on the combination of our phase-delay global
astrometry results with the microarcsecond-precise optical astrometry that will

Resource Provisioning Exploiting Cost and Performance Diversity within IaaS Cloud Providers
IaaS platforms such as Amazon EC2 allow clients access to massive computational power in the form of instances. Amazon hosts three different instance purchasing options, each with its own SLA covering pricing and availability. Amazon also offers access to a number of geographical regions, zones, and instance types to select from. In this thesis, the problem of utilizing Spot and On-Demand instances is analyzed and two approaches are presented in order to exploit the cost and performance diversity among different instance types and availability zones, and among the Spot markets they represent. We first develop RAMP, a framework designed to calculate the expected profit of using a specific Spot or On-Demand instance through an evaluation of instance reliability. RAMP is extended to develop RAMC-DC, a framework designed to allocate the most cost effective instance through strategies that facilitate interchangeability of instances among short jobs, reliability of instances among long jobs, and a comparison of the estimated costs of possible allocations. RAMC-DC achieves fault tolerance through comparisons of the price dynamics across instance types and availability zones, and through an examination of three basic checkpointing methods. Evaluations demonstrate that both frameworks take a large step toward low-volatility, high cost-efficiency resource provisioning. While achieving early-termination rates as low as 2.2%, RAMP can completely offset the total cost when charging the user just 17.5% of the On-Demand price. Moreover, the increases in profit resulting from relatively small additional charges to users are notably high, i.e., 100% profit compared to the resource provisioning cost with 35% of the equivalent On-Demand price. RAMC-DC can maintain deadline breaches below 1.8% of all jobs, achieve both early-termination and deadline breach rates as low as 0.5% of all jobs, and lowers total costs by between 80% and 87% compared to using only On-Demand instances
Reproductive Structures and Early Life History of the Gulf Toadfish, Opsanus beta, in the Tecolutla Estuary, Veracruz, Mexico
Although the Gulf toadfish, Opsanus beta, is an abundant member of the nearshore Gulf of Mexico ichthyofaunal assemblage, little information exists regarding the ecology of the species, especially for southern Gulf of Mexico populations. We added to the existing knowledge of this species by describing the reproductive structures and examining the early life history of this species in the Tecolutla estuary, Mexico. Macro- and microscopic examination of 7 males showed spermatogenesis to be similar to other teleost species except for the occurrence of biflagellate spermatozoa. Histological examination of the male accessory gland showed 3 tissue layers, but their functions are still undetermined. We found asynchronous development of oocytes in the ovaries of 16 females, which may indicate multiple spawning over the long spawning season noted in this study. Batch fecundity estimates among females ranged from 79 to 518 mature ova with a mean ovum diameter of 3.5 mm. The above-mentioned factors along with large size at hatching, attached larval forms, and paternal care may account, in part, for the abundance of this species in highly dynamic systems
