    Bringing power and progress to Africa in a financially and environmentally sustainable manner

    EXECUTIVE SUMMARY: The future of electricity supply and delivery on the continent of Africa represents one of the thorniest challenges facing professionals in the global energy, economics, finance, environmental, and philanthropic communities. Roughly 600 million people in Africa lack any access to electricity. If this deficiency is not solved, extreme poverty for many Africans is virtually assured for the foreseeable future, as it is widely recognized that economic advancement cannot be achieved in the 21st Century without good electricity supply. Yet, if Africa were to electrify in the same manner pursued in developed economies around the world during the 20th Century, the planet’s global carbon budget would be vastly exceeded, greatly exacerbating the worldwide damages from climate change. Moreover, due to low purchasing power in most African economies and fiscal insolvency of most African utilities, it is unclear exactly how the necessary infrastructure investments can be deployed to bring ample quantities of power – especially zero-carbon power – to all Africans, both those who currently are unconnected to any grid as well as those who are now served by expensive, high-emitting, limited and unreliable electricity supply. With the current population of 1.3 billion people expected to double by 2050, the above-noted challenges associated with the African electricity sector may well get substantially worse than they already are – unless new approaches to infrastructure planning, development, finance and operation can be mobilized and propagated across the continent. This paper presents a summary of the present state and possible futures for the African electricity sector. A synthesis of an ever-growing body of research on electricity in Africa, this paper aims to provide the reader a thorough and balanced context as well as general conclusions and recommendations to better inform and guide decision-making and action.     Retinal Fundus Image Enhancement Using the Normalized Convolution and Noise Removing

    Retinal fundus image plays an important role in the diagnosis of retinal related diseases. The detailed information of the retinal fundus image such as small vessels, microaneurysms, and exudates may be in low contrast, and retinal image enhancement usually gives help to analyze diseases related to retinal fundus image. Current image enhancement methods may lead to artificial boundaries, abrupt changes in color levels, and the loss of image detail. In order to avoid these side effects, a new retinal fundus image enhancement method is proposed. First, the original retinal fundus image was processed by the normalized convolution algorithm with a domain transform to obtain an image with the basic information of the background. Then, the image with the basic information of the background was fused with the original retinal fundus image to obtain an enhanced fundus image. Lastly, the fused image was denoised by a two-stage denoising method including the fourth order PDEs and the relaxed median filter. The retinal image databases, including the DRIVE database, the STARE database, and the DIARETDB1 database, were used to evaluate image enhancement effects. The results show that the method can enhance the retinal fundus image prominently. And, different from some other fundus image enhancement methods, the proposed method can directly enhance color images

    Microscopy-Based Automated Live Cell Screening for Small Molecules That Affect Ciliation

    The primary monocilium, or cilium, is a single antenna-like organelle that protrudes from the surface of most mammalian cell types, and serves as a signaling hub. Mutations of cilia-associated genes result in severe genetic disorders termed ciliopathies. Among these, the most common is autosomal dominant polycystic kidney disease (ADPKD); less common genetic diseases include Bardet–Biedl syndrome, Joubert syndrome, nephronophthisis, and others. Important signaling cascades with receptor systems localized exclusively or in part at cilia include Sonic Hedgehog (SHH), platelet derived growth factor alpha (PDGFRα), WNTs, polycystins, and others. Changes in ciliation during development or in pathological conditions such as cancer impacts signaling by these proteins. Notably, ciliation status of cells is coupled closely to the cell cycle, with cilia protruding in quiescent (G0) or early G1 cells, declining in S/G2, and absent in M phase, and has been proposed to contribute to cell cycle regulation. Because of this complex biology, the elaborate machinery regulating ciliary assembly and disassembly receives input from many cellular proteins relevant to cell cycle control, development, and oncogenic transformation, making study of genetic factors and drugs influencing ciliation of high interest. One of the most effective tools to investigate the dynamics of the cilia under different conditions is the imaging of live cells. However, developing assays to observe the primary cilium in real time can be challenging, and requires a consideration of multiple details related to the cilia biology. With the dual goals of identifying small molecules that may have beneficial activity through action on human diseases, and of identifying ciliary activities of existing agents that are in common use or development, we here describe creation and evaluation of three autofluorescent cell lines derived from the immortalized retinal pigmented epithelium parental cell line hTERT-RPE1. These cell lines stably express the ciliary-targeted fluorescent proteins L13-Arl13bGFP, pEGFP-mSmo, and tdTomato-MCHR1-N-10. We then describe methods for use of these cell lines in high throughput screening of libraries of small molecule compounds to identify positive and negative regulators of ciliary disassembly

    Measles Resurgence Associated with Continued Circulation of Genotype H1 Viruses in China, 2005

    Measles morbidity and mortality decreased significantly after measles vaccine was introduced into China in 1965. From 1995 to 2004, average annual measles incidence decreased to 5.6 cases per 100,000 population following the establishment of a national two-dose regimen. Molecular characterization of wild-type measles viruses demonstrated that genotype H1 was endemic and widely distributed throughout the country in China during 1995-2004. A total of 124,865 cases and 55 deaths were reported from the National Notifiable Diseases Reporting System (NNDRS) in 2005, which represented a 69.05% increase compared with 2004. Over 16,000 serum samples obtained from 914 measles outbreaks and the measles IgM positive rate was 81%. 213 wild-type measles viruses were isolated from 18 of 31 provinces in China during 2005, and all of the isolates belonged to genotype H1. The ranges of the nucleotide sequence and predicted amino acid sequence homologies of the 213 genotype H1 strains were 93.4%-100% and 90.0%-100%, respectively. H1-associated cases and outbreaks caused the measles resurgence in China in 2005. H1 genotype has the most inner variation within genotype, it could be divided into 2 clusters, and cluster 1 viruses were predominant in China throughout 2005

    Genetic characterization of wild-type measles viruses isolated in China, 2006-2007

    Molecular characterization of wild-type measles viruses in China during 1995-2004 demonstrated that genotype H1 was endemic and widely distributed throughout the country. H1-associated cases and outbreaks caused a resurgence of measles beginning in 2005. A total of 210,094 measles cases and 101 deaths were reported by National Notifiable Diseases Reporting System (NNDRS) and Chinese Measles Laboratory Network (LabNet) from 2006 to 2007, and the incidences of measles were 6.8/100,000 population and 7.2/100,000 population in 2006 and 2007, respectively. Five hundred and sixty-five wild-type measles viruses were isolated from 24 of 31 provinces in mainland China during 2006 and 2007, and all of the wild type virus isolates belonged to cluster 1 of genotype H1. These results indicated that H1-cluster 1 viruses were the predominant viruses circulating in China from 2006 to 2007. This study contributes to previous efforts to generate critical baseline data about circulating wild-type measles viruses in China that will allow molecular epidemiologic studies to help measure the progress made toward China's goal of measles elimination by 2012

    Single Endemic Genotype of Measles Virus Continuously Circulating in China for at Least 16 Years

    The incidence of measles in China from 1991 to 2008 was reviewed, and the nucleotide sequences from 1507 measles viruses (MeV) isolated during 1993 to 2008 were phylogenetically analyzed. The results showed that measles epidemics peaked approximately every 3 to 5 years with the range of measles cases detected between 56,850 and 140,048 per year. The Chinese MeV strains represented three genotypes; 1501 H1, 1 H2 and 5 A. Genotype H1 was the predominant genotype throughout China continuously circulating for at least 16 years. Genotype H1 sequences could be divided into two distinct clusters, H1a and H1b. A 4.2% average nucleotide divergence was found between the H1a and H1b clusters, and the nucleotide sequence and predicted amino acid homologies of H1a viruses were 92.3%–100% and 84.7%–100%, H1b were 97.1%–100% and 95.3%–100%, respectively. Viruses from both clusters were distributed throughout China with no apparent geographic restriction and multiple co-circulating lineages were present in many provinces. Cluster H1a and H1b viruses were co-circulating during 1993 to 2005, while no H1b viruses were detected after 2005 and the transmission of that cluster has presumably been interrupted. Analysis of the nucleotide and predicted amino acid changes in the N proteins of H1a and H1b viruses showed no evidence of selective pressure. This study investigated the genotype and cluster distribution of MeV in China over a 16-year period to establish a genetic baseline before MeV elimination in Western Pacific Region (WPR). Continuous and extensive MeV surveillance and the ability to quickly identify imported cases of measles will become more critical as measles elimination goals are achieved in China in the near future. This is the first report that a single endemic genotype of measles virus has been found to be continuously circulating in one country for at least 16 years

    Impact of opioid-free analgesia on pain severity and patient satisfaction after discharge from surgery: multispecialty, prospective cohort study in 25 countries

    Background: Balancing opioid stewardship and the need for adequate analgesia following discharge after surgery is challenging. This study aimed to compare the outcomes for patients discharged with opioid versus opioid-free analgesia after common surgical procedures.Methods: This international, multicentre, prospective cohort study collected data from patients undergoing common acute and elective general surgical, urological, gynaecological, and orthopaedic procedures. The primary outcomes were patient-reported time in severe pain measured on a numerical analogue scale from 0 to 100% and patient-reported satisfaction with pain relief during the first week following discharge. Data were collected by in-hospital chart review and patient telephone interview 1 week after discharge.Results: The study recruited 4273 patients from 144 centres in 25 countries; 1311 patients (30.7%) were prescribed opioid analgesia at discharge. Patients reported being in severe pain for 10 (i.q.r. 1-30)% of the first week after discharge and rated satisfaction with analgesia as 90 (i.q.r. 80-100) of 100. After adjustment for confounders, opioid analgesia on discharge was independently associated with increased pain severity (risk ratio 1.52, 95% c.i. 1.31 to 1.76; P < 0.001) and re-presentation to healthcare providers owing to side-effects of medication (OR 2.38, 95% c.i. 1.36 to 4.17; P = 0.004), but not with satisfaction with analgesia (beta coefficient 0.92, 95% c.i. -1.52 to 3.36; P = 0.468) compared with opioid-free analgesia. Although opioid prescribing varied greatly between high-income and low- and middle-income countries, patient-reported outcomes did not.Conclusion: Opioid analgesia prescription on surgical discharge is associated with a higher risk of re-presentation owing to side-effects of medication and increased patient-reported pain, but not with changes in patient-reported satisfaction.     Regeneration For Young Generation: Promote livability for young graduates through urban regeneration in Shenzhen

    Young graduates are facing housing difficulties in the Chinese metropolis. The Chinese government published new policies to encourage regeneration of the land stock resources as affordable rental housing targeting young people. This study aims to explore how urban regeneration can benefit young graduates’ livability. Specifically, it investigates the possibility to regenerate urban villages as affordable rental housing provisions. In this context, affordable rental housing tackles spatial inequities and should be located in opportunity-rich areas.To test the assumption that urban villages can be regenerated as affordable rental housing, spatial analysis on multiple scales was conducted and four well-located urban villages were chosen as cases to study. The villages were evaluated based on the indicators from young graduates’ demand analysis, and different governance models were analyzed to explore the possibilities for regeneration. The results shows that the conflict between the private interest in densification and public interest in open space is the main challenge for regeneration, and not only the spatial intervention but also the operation mechanism need to be proposed.These results suggest that encouraging spontaneous regeneration by combining top-down and bottom-up strategies may be an approach for future regeneration as well as a chance to legalize the informal village settlement. This requires not only the efforts of the village collective but also the support and active participation of the public sector. By improving the public space network and doing some experiments on strategic projects, the regeneration strategy can be tested and applied to other well-located urban villages in Shenzhen, providing more affordable housing options for young graduates.Architecture, Urbanism and Building Sciences | Complex Citie

    Males with low risk-taking propensity overestimate risk under acute psychological stress

    Arousing research has investigated stressed individuals&#39; decision biases, but whether and how stress and individual traits interact to impact the underlying decision-making process is unknown. Here, we aim to explore the effect of acute stress on the interaction between the objective level of risk and subjective risk preference (i.e. risk-taking propensity). Eighty-three healthy males participated in the study. We adopted the Trier Social Stress Test (TSST) to induce acute psychological stress and categorized participants into the high or low risk-taking propensity (HRP/LRP) group according to their traits in daily life. The Balloon Analogue Risk Task (BART) was applied to measure their feedback processing in a risky decision task, while behavioral indexes and EEG signals were recorded. The results showed that stressful participants pumped fewer times than the controls, especially for the LRP under stress, indicating that they were more willing to avoid taking risks. Compared with the stressed HRP group, the stressed LRP showed higher salivary cortisol responses and a more positive FRN following positive feedback in higher risk levels. It implies that acute psychological stress leads the LRP to overestimate the risk probability and become more cautious in the sequential processing of risk. These findings highlight the role of the feedback process and individual traits in risky decision-making under stress.</p

    The complete chloroplast genome of Petunia exserta (Solanaceae: Petunioideae), an endangered ornamental species

    Petunia exserta is an ornamental species on the brink of extinction in the wild. We report here the complete chloroplast genome of P. exserta, which is 156,598 bp in size consisting of a large single-copy region (87,095 bp), a small single-copy region (18,643 bp), and a pair of inverted repeats (25,430 bp for each). The chloroplast (used ‘cp’ hereafter) genome contains 132 genes, including 8 rRNA genes, 37 tRNA genes, and 87 protein-coding genes. Phylogenetic analysis demonstrated that P. exserta was most closely related to P. hybrida, and they together were closer to Calibrachoa hybrida than other taxa in the Solanaceae family
