    The article presents the experience of a training and orientation carried out in a lower secondary school. The overall objective of the project was carried out to assist and support the students of the third class in their choice for secondary school degree, within a structured “action path” for the orientation can evaluate and support the individual’s ability to choose, his motivations and aspirations, as well as to develop its transversal skills. The path of the results were evaluated through analysis and quantitative tools, but also with the support of qualitative analysis methods

    Adenosine A2A Receptors Modulate Acute Injury and Neuroinflammation in Brain Ischemia.

    The extracellular concentration of adenosine in the brain increases dramatically during ischemia. Adenosine A2A receptor is expressed in neurons and glial cells and in inflammatory cells (lymphocytes and granulocytes). Recently, adenosine A2A receptor emerged as a potential therapeutic attractive target in ischemia. Ischemia is a multifactorial pathology characterized by different events evolving in the time. After ischemia the early massive increase of extracellular glutamate is followed by activation of resident immune cells, that is, microglia, and production or activation of inflammation mediators. Proinflammatory cytokines, which upregulate cell adhesion molecules, exert an important role in promoting recruitment of leukocytes that in turn promote expansion of the inflammatory response in ischemic tissue. Protracted neuroinflammation is now recognized as the predominant mechanism of secondary brain injury progression. A2A receptors present on central cells and on blood cells account for important effects depending on the time-related evolution of the pathological condition. Evidence suggests that A2A receptor antagonists provide early protection via centrally mediated control of excessive excitotoxicity, while A2A receptor agonists provide protracted protection by controlling massive blood cell infiltration in the hours and days after ischemia.     The neuron-astrocyte-microglia triad in a rat model of chronic cerebral hypoperfusion: Protective effect of dipyridamole

    Chronic cerebral hypoperfusion during aging may cause progressive neurodegeneration as ischemic conditions persist. Proper functioning of the interplay between neurons and glia is fundamental for the functional organization of the brain. The aim of our research was to study the pathophysiological mechanisms, and particularly the derangement of the interplay between neurons and astrocytes-microglia with the formation of triads, in a model of chronic cerebral hypoperfusion induced by the 2-vessel occlusion (2VO) in adult Wistar rats (n=15). The protective effect of dipyridamole given during the early phases after 2VO (4 mg/kg/day i.v., the first 7 days after 2VO) was verified (n=15). Sham-operated rats (n=15) were used as controls. Immunofluorescent triple staining of neurons (NeuN), astrocytes (GFAP) and microglia (IBA1) was performed 90 days after 2VO. We found significantly higher amount of ectopic neurons, neuronal debris and apoptotic neurons in CA1 Str. Radiatum and Str. Pyramidale of 2VO rats. In CA1 Str. Radiatum of 2VO rats the amount of astrocytes (cells/mm2) did not increase. In some instances several astrocytes surrounded ectopic neurons and formed a micro scar around them. Astrocyte branches could infiltrate the cell body of ectopic neurons, and, together with activated microglia cells formed the triads. In the triad, significantly more numerous in CA1 Str. Radiatum of 2VO than in sham rats, astrocytes and microglia cooperated in the phagocytosis of ectopic neurons. These events might be common mechanisms underlying many neurodegenerative processes. The frequency to which they appear might depend upon, or might be the cause of, the burden and severity of neurodegeneration. Dypiridamole significantly reverted all the above described events. The protective effect of chronic administration of dipyridamole might be a consequence of its vasodilatory, antioxidant and anti-inflammatory role during the early phases after 2VO

    A2B Adenosine Receptors and Sphingosine 1-Phosphate Signaling Cross-Talk in Oligodendrogliogenesis

    Oligodendrocyte-formed myelin sheaths allow fast synaptic transmission in the brain. Impairments in the process of myelination, or demyelinating insults, might cause chronic diseases such as multiple sclerosis (MS). Under physiological conditions, remyelination is an ongoing process throughout adult life consisting in the differentiation of oligodendrocyte progenitor cells (OPCs) into mature oligodendrocytes (OLs). During pathological events, this process fails due to unfavorable environment. Adenosine and sphingosine kinase/sphingosine 1-phosphate signaling axes (SphK/S1P) play important roles in remyelination processes. Remarkably, fingolimod (FTY720), a sphingosine analog recently approved for MS treatment, plays important roles in OPC maturation. We recently demonstrated that the selective stimulation of A(2)(B) adenosine receptors (A(2)(B)Rs) inhibit OPC differentiation in vitro and reduce voltage-dependent outward K(+) currents (I(K)) necessary to OPC maturation, whereas specific SphK1 or SphK2 inhibition exerts the opposite effect. During OPC differentiation A(2)(B)R expression increases, this effect being prevented by SphK1/2 blockade. Furthermore, selective silencing of A(2)(B)R in OPC cultures prompts maturation and, intriguingly, enhances the expression of S1P lyase, the enzyme responsible for irreversible S1P catabolism. Finally, the existence of an interplay between SphK1/S1P pathway and A(2)(B)Rs in OPCs was confirmed since acute stimulation of A(2)(B)Rs activates SphK1 by increasing its phosphorylation. Here the role of A(2)(B)R and SphK/S1P signaling during oligodendrogenesis is reviewed in detail, with the purpose to shed new light on the interaction between A(2)(B)Rs and S1P signaling, as eventual innovative targets for the treatment of demyelinating disorders

    Urban trees facilitate the establishement of non-native forest insects

    Research ArticleCities, due to the presence of ports and airports and the high diversity of trees in streets, parks, and gardens, may play an important role for the introduction of invasive forest pests. We hypothesize that areas of urban forest facilitate the establishment of non-native forest pests. Based on scientific literature and a pan-European database on non-native species feeding on woody plants, we analysed where the first detections occurred in European countries. We collected site data for 137 first detections in Europe and 508 first European country-specific records. We also estimated the percentage of tree cover and suitable habitat (green areas with trees) in buffers around detection points. The large majority of first records (89% for first record in Europe and 88% for first records in a European country) were found in cities or suburban areas. Only 7% of the cases were in forests far from cities. The probability of occurrence decreased sharply with distance from the city. The probability to be detected in urban areas was higher for sap feeders, gall makers, and seed or fruit feeders (>90%) than for bark and wood borers (81%). Detection sites in cities were highly diverse, including public parks, street trees, university campus, arboreta, zoos, and botanical gardens. The average proportion of suitable habitat was less than 10% in urban areas where the species were detected. Further, more than 72% of the cases occurred in sites with less than 20% of tree cover. Hotspots of first detection were identified along the coastal regions of the Mediterranean and Atlantic, and near industrial areas of central Europe. We conclude that urban trees are main facilitators for the establishment of non-native forest pests, and that cities should thus be intensely surveyed.
