Efeito da profundidade de colocação do tubo de rega gota-a-gota na uniformidade de rega e na eficiência do uso da água em tomate de indústria
RESUMO
A influência da profundidade da colocação dos gotejadores na uniformidade de rega e na eficiência do uso da água foi avaliada, durante dois anos, num ensaio em “split-plot”, com quatro repetições, sendo o tratamento principal a profundidade de colocação do tubo de rega: à superfície do solo (P0), a 20 cm (PI) e a 40 cm (PII) e o secundário a cultivar: Brigade e H3044. Nos dois anos, o débito médio dos gotejadores foi semelhante nos diferentes tratamentos. Os coeficientes de uniformidade (CU) e de variação (CV) e a uniformidade de distribuição (UD), determinados após a colheita da cultura, não foram afectados pela profundidade de colocação do tubo, tendo variado respectivamente, entre 96,5 e 98,2%, 1,91 e 4,15% e 94,5 e 97,4%. A rega gota-a-gota subsuperficial, comparativamente com a superficial, contribuiu para o aumento da eficiência do uso da água (Produção comercial/ETa) em 14%, fundamentalmente devido a uma diminuição da ETa, na fase inicial da cultura
Structural alterations in the seminiferous tubules of rats treated with immunosuppressor tacrolimus
<p>Abstract</p> <p>Background</p> <p>Tacrolimus (FK-506) is an immunosuppressant that binds to a specific immunophilin, resulting in the suppression of the cellular immune response during transplant rejection. Except for some alterations in the spermatozoa, testicular morphological alterations have not been described in rats treated with tacrolimus. In the present study, we purpose to evaluate if the treatment with tacrolimus at long term of follow-up interferes in the integrity of the seminiferous tubules.</p> <p>Methods</p> <p>Rats aging 42-day-old received daily subcutaneous injections of 1 mg/kg/day of tacrolimus during 30 (T-30) and 60 (T-60) days; the rats from control groups (C-30 and C-60) received saline solution. The left testes were fixed in 4% formaldehyde and embedded in glycol methacrylate for morphological and morphometric analyses while right testes were fixed in Bouin's liquid and embedded in paraffin for detection of cell death by the TUNEL method. The epithelial and total tubular areas as well as the stages of the seminiferous epithelium and the number of spermatocytes, spermatids and Sertoli cells (SC) per tubule were obtained.</p> <p>Results</p> <p>In the treated groups, seminiferous tubules irregularly outlined showed disarranged cellular layers and loss of germ cells probably due to cell death, which was revealed by TUNEL method. In addition to germ cells, structural alterations in the SC and folding of the peritubular tissue were usually observed. The morphometric results revealed significant decrease in the number of SC, spermatocytes, spermatids and significant reduction in the epithelial and total tubular areas.</p> <p>Conclusion</p> <p>Tacrolimus induces significant histopathological disorders in the seminiferous tubules, resulting in spermatogenic damage and reduction in the number of Sertoli cells. A careful evaluation of the peritubular components will be necessary to clarify if these alterations are related to the effect of FK-506 on the peritubular tissue.</p
NF-kB overexpression and decreased immunoexpression of AR in the muscular layer is related to structural damages and apoptosis in cimetidine-treated rat vas deferens
Background: Cimetidine, histamine H-2 receptors antagonist, has caused adverse effects on the male hormones and reproductive tract due to its antiandrogenic effect. in the testes, peritubular myoid cells and muscle vascular cells death has been associated to seminiferous tubules and testicular microvascularization damages, respectively. Either androgen or histamine H-2 receptors have been detected in the mucosa and smooth muscular layer of vas deferens. Thus, the effect of cimetidine on this androgen and histamine-dependent muscular duct was morphologically evaluated.Methods: the animals from cimetidine group (CMTG; n=5) received intraperitoneal injections of 100 mg/kg b.w. of cimetidine for 50 days; the control group (CG) received saline solution. the distal portions of vas deferens were fixed in formaldehyde and embedded in paraffin. Masson's trichrome-stained sections were subjected to morphological and the following morphometrical analyzes: epithelial perimeter and area of the smooth muscular layer. TUNEL (Terminal deoxynucleotidyl-transferase mediated dUTP Nick End Labeling) method, NF-kB (nuclear factor kappa B) and AR (androgen receptors) immunohistochemical detection were also carried out. the birefringent collagen of the muscular layer was quantified in picrosirius red-stained sections under polarized light. the muscular layer was also evaluated under Transmission Electron Microscopy (TEM).Results: in CMTG, the mucosa of vas deferens was intensely folded; the epithelial cells showed numerous pyknotic nuclei and the epithelial perimeter and the area of the muscular layer decreased significantly. Numerous TUNEL-labeled nuclei were found either in the epithelial cells, mainly basal cells, or in the smooth muscle cells which also showed typical features of apoptosis under TEM. Variabilidade espacial de atributos do solo e produtividade da cultura de cana-de-açúcar em relação a localização topográfica
Soils submitted to the same management system in places with little variation of landscape, manifest differentiated spatial variability of their attributes and crop yield. The aim of this work was to investigate the correlation between spatial variability of the soil attributes and sugarcane yield as a result of soil topography. To achieve this objective, a test area of 42 ha located at the São João Sugar Mill, in Araras, in the State of São Paulo, Brazil, was selected. Sugarcane yield was measured with a yield monitor fitted in a sugarcane harvester and GPS signal. A total of 170 soil samples were taken at regular 50 m grid, at a depth of 0 - 0.2 m. The area under study was divided into two sites based on topography. The following soil attributes were analysed: organic matter (OM) content, exchangeable potassium (K), calcium (Ca) and magnesium (Mg), their base saturation percentage (%BS), cation exchange capacity (CEC), pH, clay, silt, total sand and density. The use of landscape and geostatistics enable defining areas with different spatial variability in soil attributes and crop yield, providing the visualization and definition of homogeneous management zones. The largest spatial variability of soil attributes and sugarcane yield was in the lowest part of the field.Solos submetidos ao mesmo sistema de manejo em locais com pequena variação de relevo manifestam variabilidade espacial diferenciada de atributos do solo e das plantas. Objetivou-se com este trabalho estudar a correlação entre a variabilidade espacial de atributos físicos e químicos do solo com a produtividade da cana-de-açúcar em duas posições do relevo, de acordo com a topografia da área. O estudo foi realizado no município de Araras, SP, em uma área de 42 ha, pertencente à Usina São João. Para o mapeamento da produtividade da cultura da cana-de-açúcar utilizou-se um monitor de produtividade acoplado à colhedora e ao sinal de GPS. Coletaram-se 170 amostras do solo, em uma malha regular de 50 m entre pontos, na profundidade de 0,0 - 0,2 m. O talhão de estudo foi dividido em duas áreas menores, segundo a topografia. Os atributos do solo avaliados foram: teor de matéria orgânica (MO), potássio (K), cálcio (Ca), magnésio (Mg), saturação por bases (V%), capacidade de troca catiônica (CTC), pH, argila, silte, areia total e densidade. O uso do relevo e da geoestatística possibilitou definir áreas com diferentes variabilidades espaciais para atributos do solo e produtividade da cultura de cana-de-açúcar, proporcionando a visualização e definição de zonas homogêneas de manejo de acordo com o relevo. A maior variabilidade espacial de atributos do solo e a produtividade da cultura de cana-de-açúcar ocorreram na parte mais baixa do talhão.1250125
Tissue Response and Immunoexpression of Interleukin 6 Promoted by Tricalcium Silicate-based Repair Materials after Subcutaneous Implantation in Rats

Previous issue date: 2018-03-01Introduction: The aim of the present study was to evaluate the inflammatory response induced by experimental tricalcium silicate cement with 20% zirconium oxide (TSC) and MTA Plus (MTAP; Avalon Biomed Inc, Bradenton, FL) in rat subcutaneous tissues. Methods: Polyethylene tubes were filled with TSC (n = 20) and MTAP (n = 20) and implanted in the dorsal subcutaneous tissues of 32 rats. Empty tubes were used as the control (control group [CG], n = 20). After 7, 15, 30, and 60 days, the tubes with connective tissue were removed, and the inflammatory cells and immunolabeled cells for interleukin 6 (IL-6) were counted. Data were statistically analyzed using analysis of variance and the Tukey test (P ≤.05). Results: An increased number of inflammatory and immunolabeled cells for IL-6 were observed at 7 days. The number of inflammatory cells was higher for TSC and MTAP than the CG (P <.001) at 7 days; after 30 and 60 days, no significant differences were observed among the TSC, MTAP, and CG (P =.955). The number of immunolabeled cells for IL-6 was similar for TSC, MTAP, and CG at all evaluated periods. A gradual and significant decrease was observed in the number of inflammatory cells and IL-6–immunopositive cells. At 60 days, the capsules adjacent to TSC and MTAP exhibited fibroblasts and bundles of collagen fibers. System of synchronism between sugar cane harvest machine and infield wagon
One of the problems found in mechanical harvest of sugar cane is the lack of synchronism between the harvest machine and the infield wagon, causing crop losses as well as operational capacity. The objective of the present research was to design a system capable of helping to synchronize the sugar cane harvest machine with the wagon. The communication between tractor and harvest machine is wireless. Two ultrasound sensors coupled to the elevator and a microprocessor manage such information, generating a correct synchronization among the machines. The system was tested in laboratory and on field performing its function adequately, maintaining the two machines in synchronization, indicating and alerting the operators their relative positions. The developed system reduced the sugar cane lost in 60 kg ha-1 comparing to the harvest with the system turned off.Um dos problemas encontrados na colheita mecanizada da cana-de-açúcar é a falta de sincronismo entre a colhedora e o transbordo, ocasionando perdas tanto de material como de capacidade operacional. A presente pesquisa teve como objetivo desenvolver um sistema capaz de auxiliar no sincronismo entre a colhedora e o veículo de transbordo por meio de comunicação sem fio. Dois sensores de ultra-som acoplados ao elevador e um microprocessador gerenciam tais informações, gerando correta sincronia entre as máquinas. O sistema foi testado em laboratório e em campo, cumprindo corretamente a função de manter as máquinas em sincronia, indicando e alertando aos operadores as suas posições relativas. Morphological evidences indicate that the interference of cimetidine on the peritubular components is responsible for detachment and apoptosis of Sertoli cells
Cimetidine, referred as antiandrogenic agent, has caused alterations in the seminiferous tubules, including alterations in the peritubular tissue and death of myoid cells by apoptosis. Regarding the structural and functional importance of the peritubular tissue for the maintenance of Sertoli cells (SC), we purpose to investigate the SC-basement membrane interface, focusing the morphological features of SC and their interaction with the basement membrane in the affected tubules by cimetidine. Ten animals were distributed into two groups, control (CG) and cimetidine (CmG) which received saline solution and 50 mg of cimetidine per kg of body weight, respectively, for 52 days. The testes were fixed, dehydrated and embedded for analyses under light and transmission electron microscopy. Paraffin sections were submitted to the TUNEL method; sections of testes embedded in glycol methacrylate were submitted to PAS method and stained by H&E for morphological and quantitative analyses of Sertoli Cells. In the CmG, the SC nuclei were positive to the TUNEL method and showed typical morphological alterations of cell death by apoptosis (from early to advanced stages). A significant reduction in the number of Sertoli Cells was probably due to death of these cells by apoptosis. A close relationship between SC nuclear alterations (including a high frequency of dislocated nuclei from the basal portion) and damage in the peritubular tissue was observed. The ultrastructural analysis showed a parallelism between the gradual advancement of apoptotic process in SC and detachment of the anchoring sites (hemidesmosomes) of SC plasma membrane from the lamina densa. The presence of portions of lamina densa underlying the detached hemidesmosomes indicates a continuous deposition of lamina densa, resulting in the thickening of the basal lamina. The results indicate a possible disarrangement of the SC cytoskeleton, including the focal adhesion structure. These alterations are related to SC apoptosis and probably result from disturbs induced by cimetidine on the peritubular tissue
Magnetic resonance imaging-guided occult breast lesion localization and simultaneous sentinel lymph node mapping
Background: Radio-guided occult lesion localization is a valid technique for the diagnosis of suspicious non-palpable lesions. Here we determine the feasibility of pre-operative localization of occult suspect non-palpable breast lesions using radio-guided occult lesion localization, as well as for identifying the sentinel lymph node.Methods: This is a descriptive study of data collected retrospectively. Pre-operative mapping of 34 breast lesions in 25 patients suspected of being malignant was performed using conventional imaging methods with a magnetic resonance imaging-guided radiopharmaceutical injection.Results: the mean time required to perform the localization was 25 minutes. After resection of the lesions using a gamma probe, malignancy was confirmed in fifteen patients (60.0%), with nine invasive ductal carcinomas, two invasive lobular carcinomas, and four in situ ductal carcinomas the resection was confirmed by the complete removal of the radioactive material. the pathologic results and images were concordant in all but two cases, which were submitted for new magnetic resonance imaging examinations and surgery that confirmed the malignancies. of the 15 patients with confirmed malignancies, 10 had sentinel lymph node resection. of these, eight were negative for metastases, one had micro-metastases and one had confirmed metastases. Three patients had full axillary node dissection, with metastases found in only one. Jaw and Long Bone Marrows Have a Different Osteoclastogenic Potential
Osteoclasts, the multinucleated bone-resorbing cells, arise through fusion of precursors from the myeloid lineage. However, not all osteoclasts are alike; osteoclasts at different bone sites appear to differ in numerous respects. We investigated whether bone marrow cells obtained from jaw and long bone differed in their osteoclastogenic potential. Bone marrow cells from murine mandible and tibiae were isolated and cultured for 4 and 6 days on plastic or 6 and 10 days on dentin. Osteoclastogenesis was assessed by counting the number of TRAP+ multinucleated cells. Bone marrow cell composition was analyzed by FACS. The expression of osteoclast- and osteoclastogenesis-related genes was studied by qPCR. TRAP activity and resorptive activity of osteoclasts were measured by absorbance and morphometric analyses, respectively. At day 4 more osteoclasts were formed in long bone cultures than in jaw cultures. At day 6 the difference in number was no longer observed. The jaw cultures, however, contained more large osteoclasts on plastic and on dentin. Long bone marrow contained more osteoclast precursors, in particular the myeloid blasts, and qPCR revealed that the RANKL:OPG ratio was higher in long bone cultures. TRAP expression was higher for the long bone cultures on dentin. Although jaw osteoclasts were larger than long bone osteoclasts, no differences were found between their resorptive activities. In conclusion, bone marrow cells from different skeletal locations (jaw and long bone) have different dynamics of osteoclastogenesis. We propose that this is primarily due to differences in the cellular composition of the bone site-specific marrow
