    The effect of mark enhancement techniques on the subsequent detection of saliva

    There appears to be a limited but growing body of research on the sequential analysis/treatment of multiple types of evidence. The development of an integrated forensic approach is necessary to maximise evidence recovery and to ensure that a particular treatment is not detrimental to other types of evidence. This study aims to assess the effect of latent and blood mark enhancement techniques (e.g. fluorescence, ninhydrin, acid violet 17, black iron-oxide powder suspension) on the subsequent detection of saliva. Saliva detection was performed by means of a presumptive test (PhadebasÂź) in addition to analysis by a rapid stain identification (RSID) kit test and confirmatory DNA testing. Additional variables included a saliva depletion series and a number of different substrates with varying porosities as well as different ageing periods. Examination and photography under white light and fluorescence was carried out prior to and after chemical enhancement All enhancement techniques (except BluestarÂź Forensic Magnum luminol) employed in this study resulted in an improved visualisation of the saliva stains, although the inherent fluorescence of saliva was sometimes blocked after chemical treatment. The use of protein stains was, in general, detrimental to the detection of saliva. Positive results were less pronounced after the use of black iron-oxide powder suspension, cyanoacrylate fuming followed by BY40 and ninhydrin when compared to the respective positive controls. The application of BluestarÂź Forensic Magnum luminol and black magnetic powder proved to be the least detrimental, with no significant difference between the test results and the positive controls. The use of non-destructive fluorescence examination provided good visualisation; however, only the first few marks in the depletion were observed. Of the samples selected for DNA analysis only depletion 1 samples contained sufficient DNA quantity for further processing using standard methodology. The 28 day delay between sample deposition and collection resulted in a 5-fold reduction in the amount of useable DNA. When sufficient DNA quantities were recovered, enhancement techniques did not have a detrimental effect on the ability to generate DNA profiles. This study aims to contribute to a strategy for maximising evidence recovery and efficiency for the detection of latent marks and saliva. The results demonstrate that most of the enhancement techniques employed in this study were not detrimental to the subsequent detection of saliva by means of presumptive, confirmative and DNA tests

We describe a distinctive pattern of individually variable white ‘islands’ and dots on the ventral surfaces of the throat and forearm regions of male and female Phyllomedusa trinitatis. Crucially, we report from captive-reared individuals that these patterns are ontogenetically stable. We have used these patterns to recognize individual frogs in populations of 60 and more. Examination of the ventral surfaces of other Phyllomedusa species in museum specimens and from published accounts suggests that use of these patterns provides a generally useful, minimally invasive recognition method in this genus. We fnd that species previously classed as Phyllomedusa but now regarded as belonging to different genera lack these patterns

    Modern innovative solutions to improve outcomes in severe asthma : Protocol for a mixed methods observational comparison of clinical outcomes in mission versus current care delivery

    Background: Asthma that is poorly controlled and undertreated can progress to more severe disease that is associated with high levels of unscheduled care that requires high-cost therapy, leading to a significant health economic burden. The identification and appropriate referral to a specialist asthma service is also often delayed by several months or years because of poor recognition and understanding of symptom severity. Current severe asthma services may take several months to provide a comprehensive multidisciplinary assessment, often necessitating multiple hospital visits and costing up to ÂŁ5000 per patient. Objective: This study aims to evaluate whether a new service model could identify poorly controlled and potentially severe asthma much earlier in the patient pathway, and then compare clinical outcomes between this new care model with standard care. Methods: Modern Innovative Solutions to Improve Outcomes in (MISSION) Severe Asthma is a novel service model developed by asthma specialists from Portsmouth and Southampton severe asthma services. MISSION Severe Asthma identified patients with poorly controlled disease from general practice databases who had not been under secondary outpatient care in the last 12 months or who were not known to secondary care. In 1- or 2-stop assessments, a thorough review of diagnosis, disease phenotype, and control is undertaken, and clinical outcomes collected at baseline. Results: A variety of clinical outcomes will be collected to assess the service model. The results will be reported in February 2020. Conclusions: This protocol outlines a mixed methods study to assess the impact on disease control, unscheduled health care usage, and quality of life in patients seen in the MISSION clinic compared with a closely matched cohort who declined to attend

    In-situ characterization of the thermal state of resonant optical interferometers via tracking of their higher-order mode resonances

    Thermal lensing in resonant optical interferometers such as those used for gravitational wave detection is a concern due to the negative impact on control signals and instrument sensitivity. In this paper we describe a method for monitoring the thermal state of such interferometers by probing the higher-order spatial mode resonances of the cavities within them. We demonstrate the use of this technique to measure changes in the Advanced LIGO input mode cleaner cavity geometry as a function of input power, and subsequently infer the optical absorption at the mirror surfaces at the level of 1 ppm per mirror. We also demonstrate the generation of a useful error signal for thermal state of the Advanced LIGO power recycling cavity by continuously tracking the first order spatial mode resonance frequency. Such an error signal could be used as an input to thermal compensation systems to maintain the interferometer cavity geometries in the presence of transients in circulating light power levels, thereby maintaining optimal sensitivity and maximizing the duty-cycle of the detectors

    Chronic amphetamine enhances visual input to and suppresses visual output from the superior colliculus in withdrawal

    Heightened distractibility is a core symptom of Attention DeïŹcit Hyperactivity Disorder (ADHD). Effective treatment is normally with chronic orally administered psychostimulants including amphetamine.Treatment prevents worsening of symptoms but the site of therapeutic processes, and their nature, is unknown. Mounting evidence suggests that the superior colliculus (SC) is a key substrate in distractibility and a therapeutic target, so we assessed whether therapeutically-relevant changes are induced in this structure by chronic oral amphetamine. We hypothesized that amphetamine would alter visual responses and morphological measures. Six-week old healthy male rats were treated with oral amphetamine (2, 5 or 10 mg/kg) or a vehicle for one month after which local ïŹeld potential and multiunit recordings were made from the superïŹcial layers of the SC in response to whole-ïŹeld light ïŹ‚ashes in withdrawal. Rapid Golgi staining was also used to assess dendritic spines, and synaptophysin staining was used to assess synaptic integrity. Chronic amphetamine increased local ïŹeld potential responses at higher doses, and increased synaptophysin expression, suggesting enhanced visual input involving presynaptic remodelling. No comparable increases in multiunit activity were found suggesting a mphetamine suppresses collicular output activity, counterbalancing the increased input. We also report,for the ïŹrst time, ïŹve different dendritic spine types in the superïŹcial layers and show these to be unaffected by amphetamine, indicating that suppression does not involve gross postsynaptic structural alterations. In conclusion, we suggest that amphetamine produces changes at the collicular level that potentially stabilise the structure and may prevent the worsening of symptoms in disorders like ADHD

    The effects of tryptophan loading on Attention Deficit Hyperactivity in adults:A remote double blind randomised controlled trial

    BackgroundDespite the impact and prevalence of Attention Deficit Hyperactivity Disorder (ADHD), current treatment options remain limited and there is a drive for alternative approaches, including those building on evidence of a role for tryptophan (TRP) and serotonin (5-HT). This study aimed to evaluate the effect of acute TRP loading on attention and impulsivity in adults with ADHD.Trial design and methods We conducted a remote double blind randomised controlled trial (RCT) using TRP loading to examine the effects of a balanced amino acid load in comparison to low and high TRP loading in individuals with ADHD (medicated, N = 48, and unmedicated, N = 46) and controls (N = 50). Participants were randomised into one of three TRP treatment groups using stratified randomisation considering participant group and gender using a 1:1:1 ratio. Baseline testing of attention and impulsivity using the Test of Variables of Attention Task, Delay Discounting Task, and Iowa Gambling Task was followed by consumption of a protein drink (BAL, LOW, or HIGH TRP) before repeated testing. Results and ConclusionsNo effects of TRP were observed for any of the measures. In the present study, TRP loading did not impact on any measure of attention or impulsivity in those with ADHD or Controls. The findings need to be confirmed in another trial with a larger number of patients that also considers additional measures of dietary protein, plasma TRP and aggression.     Cardiovascular disease in transgender individuals

    The population of people identifying as transgender has grown rapidly in recent years, resulting in a substantive increase in individuals obtaining gender-affirming medical care to align their secondary sex characteristics with their gender identity. This has established benefits for patients including improvements in gender dysphoria and psychosocial functioning, while reducing adverse mental health outcomes. Despite these potential advantages, recent evidence has suggested that gender-affirming hormone therapy (GAHT) may increase the risk of cardiovascular disease. However, owing to a paucity of research, the mechanisms underpinning these increased risks are poorly understood. Moreover, previous research has been limited by heterogenous methodologies, being underpowered, and lacking appropriate control populations. Consequently, the need for evidence regarding cardiovascular health in LGBTQ + individuals has been recognised as a critical area for future research to facilitate better healthcare and guidance. Recent research investigating the effect of transmasculine (testosterone) GAHT on cardiovascular disease risk points to testosterone effecting the nitric oxide pathway, triggering inflammation, and promoting endothelial dysfunction. Equivalent studies focussing on transfeminine (oestrogen) GAHT are required, representing a crucial area of future research. Furthermore, when examining the effects of GAHT on the vasculature, it cannot be ignored that there are multiple factors that may increase the burden of cardiovascular disease in the transgender population. Such stressors include major psychological stress; increased adverse health behaviours, such as smoking; discrimination; and lowered socioeconomic status; all of which undoubtedly impact upon cardiovascular disease risk and offers the opportunity for intervention

    Physiographic Controls on Landfast Ice Variability from 20 Years of Maximum Extents across the Northwest Canadian Arctic

    Landfast ice is a defining feature among Arctic coasts, providing a critical transport route for communities and exerting control over the exposure of Arctic coasts to marine erosion processes. Despite its significance, there remains a paucity of data on the spatial variability of landfast ice and limited understanding of the environmental processes’ controls since the beginning of the 21st century. We present a new high spatiotemporal record (2000–2019) across the Northwest Canadian Arctic, using MODIS Terra satellite imagery to determine maximum landfast ice extent (MLIE) at the start of each melt season. Average MLIE across the Northwest Canadian Arctic declined by 73% in a direct comparison between the first and last year of the study period, but this was highly variable across regional to community scales, ranging from 14% around North Banks Island to 81% in the Amundsen Gulf. The variability was largely a reflection of 5–8-year cycles between landfast ice rich and poor periods with no discernible trend in MLIE. Interannual variability over the 20-year record of MLIE extent was more constrained across open, relatively uniform, and shallower sloping coastlines such as West Banks Island, in contrast with a more varied pattern across the numerous bays, headlands, and straits enclosed within the deep Amundsen Gulf. Static physiographic controls (namely, topography and bathymetry) were found to influence MLIE change across regional sites, but no association was found with dynamic environmental controls (storm duration, mean air temperature, and freezing and thawing degree day occurrence). For example, despite an exponential increase in storm duration from 2014 to 2019 (from 30 h to 140 h or a 350% increase) across the Mackenzie Delta, MLIE extents remained relatively consistent. Mean air temperatures and freezing and thawing degree day occurrences (over 1, 3, and 12-month periods) also reflected progressive northwards warming influences over the last two decades, but none showed a statistically significant relationship with MLIE interannual variability. These results indicate inferences of landfast ice variations commonly taken from wider sea ice trends may misrepresent more complex and variable sensitivity to process controls. The influences of different physiographic coastal settings need to be considered at process level scales to adequately account for community impacts and decision making or coastal erosion exposure
