    Purification of a lectin with antibacterial activity from Bothrops leucurus snake venom

    A novel lectin was isolated from Bothrops leucurus snake venom using a combination of affinity and gel filtration chromatographies. the lectin (BIL) agglutinated glutaraldehyde-treated rabbit and human erythrocytes with preference for rabbit erythrocytes. Galactose, raffinose, lactose, fetal bovine serum and casein inhibited lectin-induced rabbit erythrocyte agglutination. BIL, with a molecular mass of 30 kDa and composed of two subunits of 15 kDa, showed dependence on calcium. BIL is an acidic protein with highest activity over the pH range of 4.0-7.0 and stable under heating to 70 degrees C. Fluorescence emission spectra showed tryptophan residues partially buried within the lectin structure. the percentages of secondary structure revealed by circular dichroism were 1% alpha-helix, 44% beta-sheet, 24% beta-turn and 31% unordered. BIL showed effective antibacterial activity against Gram-positive bacteria Staphylococcus aureus, Enterococcus faecalis and Bacillus subtilis with minimal inhibitory concentrations of 31.25, 62.25 and 125 mu g/mL, respectively. in conclusion, B. leucurus snake venom contains a galactoside-binding lectin with antibacterial activity.     Characterization of protease, protease inhibitors and a storage protein (cratilin) from Cratylin mollis seeds

    Uma atividade endopeptidasica (Cm-endopepbdase), uma atividade aminopeptidasica (Cm-aminopeptidase), isoinibidores de tripsina (CmTIl e CmTI2) e uma proteina tipo vicilina (cratilina) foram isoladas e parcialmente caracterizadas de sementes de Cratylia moiiis. Cm-endopepbdase hidrolisa Bz-Arg-p-Nan e Ac-Phe-Arg-p-Nan, e termosenslvel, seu pH otimo de acao situa-se entre 7,5 e 8,0 e massa molecular estimada e 80.000. Eletroforese em gel de poliacrilamida, em presenca de DTT, revelou uma banda de massa molecular 45.000. Em cromatografia de fase reversa, em coluna de C-18, Cm-endopepddase apresentou um tempo de retencao de 24 minutos. A atividade em presenca de agentes quelantes, inibidores e reagentes grupos -SH indica que a enzima e uma serinoproteinase que pode depender de metal e talvez tenha um grupo SH importante para a sua estrutura. A hidrolise de Bz-Arg-p-Nan foi estudada em presenca de metais e foi apenas parcialmente inibida por ions de zinco. A preparacao de Cm-endopepbdase apresentou atividade proteolitica sobre caserna e hidrolisou a ligacao Phe-Ser da bradicinina, Lys-bradicinina e Met-Lys-bradicinina; TPCK inibiu parcialmente a hidroiise de bradicinina. Cm-amjnopepddase hidrolisa preferencialmente Leu-b-Na e, semelhante a endopeptidase, e termosensivel, o pH otimo de acao situa-se na faixa de pH alcalino, e ocorre com massas moleculares de 80.000 e 45.000, na ausencia e na presenca de agente redutor, respectivamente. A cromatografia de fase reversa en coluna de C-18 revelou que a enzima e mais hidrofobica que Cm-endopepbdase, sendo eluida aos 30 minutos. A atividade aminopeptidasica foi parcialmente inibida por bestatina e puromicina e foi sensivel a metais, sendo totalmente abolida em presenca de zinco e parcialmente inibida com calcio, cobalto, magnesio e manganes. CmTI isolado por diferentes procedimentos, inclusive cromatografia em tripsina-Sepharose, e estavel na faixa de pH de 2,0 a 10,0 e em diferentes temperaturas. Da cromatografla de gel filtracao foi isolado o dimero do inibidor, com massa molecular 21.000. A eletroforese em gel de poliacrilamida revelou que o monomero tem massa molecular 11.000. Da cromatografia de fase reversa foram isoladas duas formas do inibidor e cujas estruturas primarias apresentaram alta homologia entre si mesmas e com inibidores da familia Bowman-Birk. Animals (Nine-week-old male albino Wistar rats, Rattus norvegicus) were treated with intraperitoneal injection of Cramoll 1,4 (235 mu g ml(-1) single dose) and, 7 days later, spleen lymphocytes were isolated and analyzed for intracellular ROS, cytosolic Ca(2+), and IL-6, IL-10, and IL-1 mRNAs. Cell viability was investigated by annexin V-FITC and 7-amino-actinomycin D staining. The data showed that in lymphocytes activated by Cramoll 1,4 the increase in cytosolic and mitochondrial ROS was related to higher cytosolic Ca(2+) levels. Apoptosis and necrosis were not detected in statistically significant values and thus the lectin effector activities did not induce lymphocyte death. In vivo Cramoll 1,4 treatment led to a significant increase in IL-1 beta but IL-6 and -10 levels did not change.     Hemagglutinating activity has been associated to presence of lectin, carbohydrate-binding proteins. in this work Crataeva tapia bark lectin (CrataBL) was purified in milligram quantities (28 mg per g of bark) by ion exchange chromatography. the lectin was thermo-stable, ion-independent and N-terminal sequence analysis demonstrated similarity with miraculin and miraculin-like proteins (plant defensive proteins). Glycosylated nature of CrataBL was revealed using glycoprotein staining (periodic acid-Schiff's reagent), positive for polypeptides of apparent molecular masses 21 and 40 kDa on SOS-PAGE. Gel diffusion assay showed that glucose/mannose isolectins from Cratylia mollis recognized CrataBL glycan moiety. CrataBL hemagglutinating activity was inhibited by glycoproteins and CrataBL immobilized on cyanogen bromide-activated sepharose 4B (1 mL) bound 0.54 mg of glycoprotein (casein, fetuin and ovalbumin) per cycle. CrataBL was an insecticide agent against Nasutitermes corniger workers (termite that attack woods) with LC50 of 0.475 mg mL(-1) for 6 days. (C) 2011 Elsevier Ireland Ltd.     Structural characterization of coagulant Moringa oleifera Lectin and its effect on hemostatic parameters

    Lectins are carbohydrate recognition proteins. cMoL, a coagulant Moringa oleifera Lectin, was isolated from seeds of the plant. Structural studies revealed a heat-stable and pH resistant protein with 101 amino acids, 11.67 theoretical pI and 81% similarity with a M. oleifera flocculent protein. Secondary structure content was estimated as 46% alpha-helix, 12% beta-sheets, 17% beta-turns and 25% unordered structures belonging to the alpha/beta tertiary structure class. cMoL significantly prolonged the time required for blood coagulation, activated partial thromboplastin (aPTF) and prothrombin times (PT), but was not so effective in prolonging aPTT in asialofetuin presence. cMoL acted as an anticoagulant protein on in vitro blood coagulation parameters and at least on aPTT, the lectin interacted through the carbohydrate recognition domain.     Application of Omics Technologies for evaluation of antibacterial mechanisms of action of plant-derived products

    In the face of increasing bacterial resistance to antibiotics currently in use, the search for new antimicrobial agents has received a boost in recent years, with natural products playing an important role in this field. In fact, several methods have been proposed to investigate the antibacterial activities of natural products. However, given that the ultimate aim is future therapeutic use as novel drugs, it is extremely necessary to elucidate their modes of action, stating the molecular effects in detail and identifying their targets in the bacterial cell. This review analyzes the application of omics technologies to understand the antibacterial mechanisms of bioactive natural products, to stimulate research interest in this area and promote scientific collaborations. Some studies have been specifically highlighted herein by examining their procedures and results (targeted proteins and metabolic pathways). These approaches have the potential to provide new insights into our comprehension of antimicrobial resistance/susceptibility, creating new perspectives for the struggle against bacteria, and leading to the development of novel products in the future

    Crystal Structure of Crataeva tapia Bark Protein (CrataBL) and Its Effect in Human Prostate Cancer Cell Lines

    A protein isolated from the bark of Crataeva tapia (CrataBL) is both a Kunitz-type plant protease inhibitor and a lectin. We have determined the amino acid sequence and three-dimensional structure of CrataBL, as well as characterized its selected biochemical and biological properties. We found two different isoforms of CrataBL isolated from the original source, differing in positions 31 (Pro/Leu); 92 (Ser/Leu); 93 (Ile/Thr); 95 (Arg/Gly) and 97 (Leu/Ser). CrataBL showed relatively weak inhibitory activity against trypsin (K-iapp = 43 mu M) and was more potent against Factor Xa (K-iapp = 8.6 mu M), but was not active against a number of other proteases. We have confirmed that CrataBL contains two glycosylation sites and forms a dimer at high concentration. the high-resolution crystal structures of two different crystal forms of isoform II verified the beta-trefoil fold of CrataBL and have shown the presence of dimers consisting of two almost identical molecules making extensive contacts (similar to 645 angstrom(2)). the structure differs from those of the most closely related proteins by the lack of the N-terminal beta-hairpin. in experiments aimed at investigating the biological properties of CrataBL, we have shown that addition of 40 mM of the protein for 48 h caused maximum growth inhibition in MTT assay (47% of DU145 cells and 43% of PC3 cells). the apoptosis of DU145 and PC3 cell lines was confirmed by flow cytometry using Annexin V/FITC and propidium iodide staining. Treatment with CrataBL resulted in the release of mitochondrial cytochrome c and in the activation of caspase-3 in DU145 and PC3 cells

    Tecoma stans (yellow elder) has shown medicinal properties and antimicrobial activity. Previous reports on antifungal activity of T. stans preparations and presence of trypsin inhibitor activity from T. stans leaves stimulated the investigation reported here. In this work, we proceeded to the purification and characterization of a trypsin inhibitor (TesTI), which was investigated for anti-Candida activity. Finally, in order to determine the potential of TesTI as a new natural chemotherapeutic product, its cytotoxicity to human peripheral blood mononuclear cells (PBMCs) was evaluated. TesTI was isolated from saline extract by ammonium sulphate fractionation followed by ion exchange and gel filtration chromatographies. Antifungal activity was evaluated by determining the minimal inhibitory (MIC) and fungicide (MFC) concentrations using fungal cultures containing only yeast form or both yeast and hyphal forms. Candida cells treated with TesTI were evaluated for intracellular ATP levels and lipid peroxidation. Cytotoxicity of TesTI to PBMCs was evaluated by MTT assay. TesTI (39.8 kDa, pI 3.41, Ki 43 nM) inhibited similarly the growth of both C. albicans and C. krusei culture types at MIC of 100 µg/mL. The MFCs were 200 µg/mL for C. albicans and C. krusei. Time-response curves revealed that TesTI (at MIC) was more effective at inhibiting the replication of C. albicans cells. At MIC, TesTI promoted reduction of ATP levels and lipid peroxidation in the Candida cells, being not cytotoxic to PBMCs. In conclusion, TesTI is an antifungal agent against C. albicans and C. krusei, without toxicity to human cells
