    The Grounding of an Ice Shelf in the Central Arctic Ocean: A Modeling Experiment

    A numerical ice sheet model was used in a first test towards evaluating the hypothesis that, during a period of large-scale glaciation, an ice shelf emanating from the Barents/Kara Seas grounded across parts of the Lomonosov Ridge to a depth of around 1000 m below present sea level (Jakobsson, 1999; Polyak et al., 2001). Despite that we not include complex ice shelf physics or grounding line mechanics in our model and treat the process of marine melting in a simple manner, our experiments are the necessary first steps toward providing a comprehensive reconstruction of the former ice-sheet/ice-shelf system in the Arctic Ocean. A series of model runs was performed where ice shelf mass balance and ice shelf strain per unit time (strain rate) were adjusted. The mass balance and shelf ice strain rate are the key model parameters that govern the flux of ice into the Arctic Ocean. Grounding on the Lomonosov Ridge was not modeled when the ice shelf strain rate was 0.005 year-1 (i.e. a free flowing ice shelf). Even with low rates (\u3c10 cm/year) of basal melting, the ice shelf thickness was always less than 100 m over the central part of the ridge. Our experiment suggests that grounding on the Lomonosov Ridge by a free-flowing ice shelf is not possible. When the strain rate in the shelf ice was reduced to zero, however, the shelf thickness increased substantially. Such conditions are likely only to have occurred during periods of large-scale glaciation if substantial stagnant and thickened sea ice was present in the ocean, buttressing the ice shelf flowing from the Barents Sea. A comprehensive study using a coupled icesheet/ shelf/sea-ice model would build on these preliminary results and have the potential to further constrain the history of circum-Arctic Ocean ice sheets

    A Modeling Experiment on the Grounding of an Ice Shelf in the Central Arctic Ocean During MIS 6

    High-resolution chirp sonar subbottom profiles from the Lomonosov Ridge in the central Arctic Ocean, acquired from the Swedish icebreaker Oden in 1996, revealed large-scale erosion of the ridge crest down to depths of 1000 m below present sea level [Jakobsson, 1999]. Subsequent acoustic mapping during the SCICEX nuclear submarine expedition in 1999 showed glacial fluting at the deepest eroded areas and subparallel ice scours from 950 m water depth to the shallowest parts of the ridge crest [Polyak et al., 2001]. The directions of the mapped glaciogenic bed-forms and the redeposition of eroded material on the Amerasian side of the ridge indicate ice flow from the Barents-Kara Sea area. Core studies revealed that sediment drape the eroded areas from Marine Isotope Stage (MIS) 5.5 and, thus, it was proposed that the major erosional event took place during Marine Isotope Stage (MIS) 6 [Jakobsson et al., 2001]. Glacial geological evidence suggests strongly that the Late Saalian (MIS 6) ice sheet margin reached the shelf break of the Barents-Kara Sea [Svendsen et al. in press] and this gives us two possible ways to explain the ice erosional features on the Lomonosov Ridge. One is the grounding of a floating ice shelf and the other is the scouring from large deep tabular iceberg. Here we apply numerical ice sheet modeling to test the hypothesis that an ice shelf emanating from the Barents/Kara seas grounded across part of the Lomonsov Ridge and caused the extensive erosion down to a depth of around 1000 m below present sea level. A series of model experiments was undertaken in which the ice shelf mass balance (surface accumulation and basal melting) and ice shelf strain rates were adjusted. Grounding of the Lomonosov Ridge was not achieved when the ice shelf strain rate was 0.005 yr-1 (i.e. a free flowing ice shelf). However this model produced two interesting findings. First, with basal melt rates of up to 50 cm yr-1 an ice shelf grew from the St. Anna Trough ice stream across the section of the ridge where there is evidence for grounding. Second, even with ultra low rates of basal melting, the ice shelf thickness was always less than 200 m over the ridge. We conclude that grounding of the Lomonosov Ridge by a free-flowing ice shelf is not possible. When the strain rate was reduced to zero, however, the shelf thickness increased substantially. Such conditions are likely only to have occurred during periods of large-scale glaciation across the Eurasian Arctic such as in the Saalian, and if a substantial stagnant thickened sea ice was present in the ocean, buttressing the shelf flowing from the Barents Sea. Our results are interpreted using new techniques for dynamic 3Dvisualization

    The impact of pneumolysin on the macrophage response to Streptococcus pneumoniae is strain-dependent

    Streptococcus pneumoniae is the world's leading cause of pneumonia, bacteremia, meningitis and otitis media. A major pneumococcal virulence factor is the cholesterol-dependent cytolysin, which has the defining property of forming pores in cholesterol-containing membranes. In recent times a clinically significant and internationally successful serotype 1 ST306 clone has been found to express a non-cytolytic variant of Ply (Ply306). However, while the pneumococcus is a naturally transformable organism, strains of the ST306 clonal group have to date been virtually impossible to transform, severely restricting efforts to understand the role of non-cytolytic Ply in the success of this clone. In this study isogenic Ply mutants were constructed in the D39 background and for the first time in the ST306 background (A0229467) to enable direct comparisons between Ply variants for their impact on the immune response in a macrophage-like cell line. Strains that expressed cytolytic Ply were found to induce a significant increase in IL-1β release from macrophage-like cells compared to the non-cytolytic and Ply-deficient strains in a background-independent manner, confirming the requirement for pore formation in the Ply-dependent activation of the NLRP3 inflammasome. However, cytolytic activity in the D39 background was found to induce increased expression of the genes encoding GM-CSF (CSF2), p19 subunit of IL-23 (IL23A) and IFNβ (IFNB1) compared to non-cytolytic and Ply-deficient D39 mutants, but had no effect in the A0229467 background. The impact of Ply on the immune response to the pneumococcus is highly dependent on the strain background, thus emphasising the importance of the interaction between specific virulence factors and other components of the genetic background of this organism

    Isolation site influences virulence phenotype of serotype 14 Streptococcus pneumoniae strains belonging to multilocus sequence type 15

    Streptococcus pneumoniae is a diverse species causing invasive as well as localized infections that result in massive global morbidity and mortality. Strains vary markedly in pathogenic potential, but the molecular basis is obscured by the diversity and plasticity of the pneumococcal genome. We have previously reported that S. pneumoniae serotype 3 isolates belonging to the same multilocus sequence type (MLST) differed markedly in in vitro and in vivo phenotypes, in accordance with the clinical site of isolation, suggesting stable niche adaptation within a clonal lineage. In the present study, we have extended our analysis to serotype 14 clinical isolates from cases of sepsis or otitis media that belong to the same MLST (ST15). In a murine intranasal challenge model, five ST15 isolates (three from blood and two from ears) colonized the nasopharynx to similar extents. However, blood and ear isolates exhibited significant differences in bacterial loads in other host niches (lungs, ear, and brain) at both 24 and 72 h postchallenge. In spite of these differences, blood and ear isolates were present in the lungs at similar levels at 6 h postchallenge, suggesting that early immune responses may underpin the distinct virulence phenotypes. Transcriptional analysis of lung tissue from mice infected for 6 h with blood isolates versus ear isolates revealed 8 differentially expressed genes. Two of these were exclusively expressed in response to infection with the ear isolate. These results suggest a link between the differential capacities to elicit early innate immune responses and the distinct virulence phenotypes of clonally related S. pneumoniae strains

    An object-oriented approach has been applied to the different stages involved in developing a knowledge base about insulin metabolism. At an early stage the separation of terminological and assertional knowledge was made. The terminological component was developed by medical experts and represented in CORE. An object-oriented knowledge acquisition process was applied to the assertional knowledge. A frame description is proposed which includes features like states and events, inheritance and collaboration. States and events are formalized with qualitative calculus. The terminological knowledge was very useful in the development of the assertional component. It assisteed in understanding the problem domain, and in the implementation stage, it assisted in building good inheritance hierarchies

    Social development for the Cape Flats: an alternative planning structure

    An initial analysis into the present problems which will affect the future growth of the Western Cape resulted in the conclusion that the situation of the Coloured population is the factor most urgently requiring positive planning resolutions. Understanding the area's problems leads one to conclude that the main thrusts must be directed towards increased opportunities for the residents to fulfil their potential, and towards a greater involvement in the decisions made about their future. The arrangement of the thesis was thus developed around the three interrelated notions of administrative structure, human resources and social development, physical and economic resources. An unbound 'process diagram’ is provided in the back cover to facilitate an understanding of the sequence adopted. The administrative and planning activities of various local authorities, particularly the Cape Town City Council, were investigated with respect to the relationship between their actions and the causes of problems on the Flats. The complexity of social problems necessitated a consideration of the concept and the theory of social development. This was defined as the process whereby the individual and/or group is enabled to increasingly participate in and contribute to the growing flow of social transactions over time. By adopting Social Development as the goal for the Cape Flats and developing objectives for its six primary subsystems (housing, education, employment, recreation, community and civic), the subsequent form of the thesis was established
