    Endometrial assessment in cases of primary infertility in women: a prospective study

    Background: Worldwide Infertility rate prevails around 8-12% of all couples; approximately 80 million are infertile. In India 10-15% of couples are infertile. Investigating for infertility is to intervene into the modifiable/treatable causes. Therefore, the present study has been undertaken to investigate the morphological patterns of endometrial tissue in women with primary infertility.Methods: The study included 60 cases with complaints of infertility (primary). The premenstrual D and C was done to obtain endometrial biopsy. The present prospective study was a descriptive study and the values are mentioned in percentages.Results: In present study a total of 60 cases were studied. Out of which 36 cases (60%) belonged to the young adult age group. The predominant morphological pattern was that of secretory endometrium seen in 22 cases (36.66%) as opposed to proliferative, endometritic or tubercular pattern. The predominant menstrual pattern recorded was regular; seen in 41 cases (68.33%) as opposed to irregular or mennorhagic patterns.Conclusions: The endometrial biopsy has a great role in screening the cases of infertility as it helps to assess the information about ovulation, ripening of the endometrial tissue and other abnormal endometrial reaction, hormonal imbalance. This is the only method to label the diagnosis of endometrial tuberculosis in an apparently healthy female

    Chitosan mediated nanoparticles from Saccharomyces cerevisiae and its Mosquito Larvicidal Activity

    Mosquitoes are one of the most medically significant groups of vectors, having an ability to transmit parasites and pathogens that can have devastating impacts on humans. In this study, chitosan nanoparticles were synthesized from chitosan polymer by ionic gelation method. The chitin was first extracted from Saccharomyces cerevisiae and then deacetylated to chitosan. Silver nanopartcals were also prepared and the presence and characterization was investigated by scanning electron microscopy (SEM). The comparative study of the larvicidal activity of chitosan nanoparticles and silvers was also studied which shows chitosan nanoparticles started  mortality at higher concentrations, it showed uniform rise in mortality of mosquito larvae than silver nanoparticles.

    SUBSTRATE CARRIERS FOR C-1(2)-DEHYDROGENATION OF 6-METHYLENE ANDROSTENEDIONE TO EXEMESTANE BY GROWING AND IMMOBILIZED ARTHROBACTER SIMPLEX NCIM 2449

    Objective: Permeability of hydrophobic steroid substrates across cell membrane is a critical factor during microbial bioconversion. To increase substrate intake, the feasibility of some organic solvents and emulsifiers as substrate carrier on the bioconversion of 6-methylene androstenedione to exemestane was assessed.Methods: Androstenedione, a commonly available steroid precursor, was chemically converted 6-methylene androstenedione. The time course of exemestane accumulation was estimated after addition of 6-methylene androstenedione dissolved in some organic solvents or dispersed with emulsifiers by growing and immobilized cells of Arthrobacter simplex NCIM 2449 in shake flask cultures.  Results: The use of substrate carriers for addition of 6-methylene androstenedione enhanced the bioconversion several folds. With growing bacterium in triplicate flasks, a peak mol % bioconversion recorded was- ethanol (67.25, 72 h); soybean oil + tween 80 (50.37, 48 h); acetone (38.84, 48 h); soybean oil (38.36, 48 h); lecithin (32.73, 48 h), methanol (32.71, 48 h) and tween 80 (10.37, 48 h). As compared to the growing cells, the bioconversion with Ca-alginate immobilized cells was delayed and peak mol % bioconversion was recorded as ethanol (60.78, 120 h); soybean oil + tween 80 (42.98, 120 h);  methanol (40.50,  72 h);  soybean oil (38.36, 48 h);  acetone (31.18, 72h ) and lecithin (33.67, 120 h); tween 80 (13.87, 120 h).Conclusion: The use of substrate carriers for addition of 6-methylene androstenedione increased the permeability of substrate and may be used to increase the yield of exemestane and reduce incubation time

    QUANTIFICATION OF EXEMESTANE ACCUMULATION DURING MICROBIAL BIOCONVERSION BY TLC IMAGE ANALYSIS

    Objective: The present study was aimed at developing a rapid, cost effective and accurate method for quantification of exemestane using thin layer chromatography (TLC) separation followed by image analysis and to test it for monitoring the accumulation of exemestane during microbial bioconversion.Methods: After microbial bioconversion and TLC separation of products formed, exemestane was quantified using ImageQuant TL v2003 image analysis software and the results were compared with high performance liquid chromatography (HPLC) analysis.Results: The percentage error between TLC and HPLC analyses was ranged from (-) 5.18 to (+) 5.51. Bacterial strains Arthrobacter simplex IAM 1660, Nocardia sp. MTCC 1534, Pseudomonas putida MTCC 1194 and Rhodococcus rhodochorus MTCC 291 respectively yielded 79.7 (72 h), 63.9 (72 h), 69.8 (96 h) and 83.2 (96 h) mole percent bioconversion of 6-methylene androstenedione to exemestane. Conclusion: Rhodococcus rhodochorus MTCC 291 was found to be the most suitable organism for the bioconversion and may be used to develop an eco-friendly route to replace chemical synthesis that eliminates the use of toxic chemicals and side products

    Assessment of parasitic load in goat through the examination of faecal matter

    Parasitic infection do not show heavy rate of mortality, however there occurrence being chronic, most of the time leads to serious production losses, this led to study about severity of parasitic load and type of parasitic infection in goats. Parasitic infection most of the time leads to serious production losses. Gastrointestinal nematodes are ubiquitous parasites of grazing ruminants and cause decreases in survival, live weight gain, wool and milk production and reproduction performance. Parasitic problems are a serious problem in goat. Total 60 goat faecal samples were analyzed. These results would serve as a baseline for future studies. The majority of the faecal samples (70%) of both zone I and zone II had heavy parasitic load (>3000 epg) followed by 60 per cent samples of zone III. This indicates that majority of the goats of the study area had severe parasitic infection. Chi-square analysis revealed non-significant relation between parasitic load and categories of zones. Majority of samples (48.33%) were infected with the combination of Strongyles, Strongyloides and Coccidiosis. It can be concluded that faecal egg count level was severe in majority of the samples examined

Unfortunately, the natural non linear characteristic of sensor itself and also the dynamic nature of the environment, aging effect, inherent sensor’s noise and data loss due to transients or intermittent faults affects the sensor characteristics non linearly. As the transfer characteristic of most sensors is nonlinear in nature, obtaining data from such a nonlinear sensor, by using an optimized device, has always been a design challenge. Linearization of nonlinear sensor characteristic in digital environment, is a vital step in the instrument signal conditioning process.     COMPARATIVE STUDY OF EFFECT OF SWERTIA CHIRATA LEAF EXTRACT ON INDINAVIR TREATED RATS

    Background: Indinavir is widely used for the treatment of human immunodeficiency virus (HIV) infection. It is known to cause hyperglycemia or insulin resistance and hyperlipidemia.Aim and Objectives: To study the effect of Swertia chirata leaf extract with metformin and pioglitazone on indinavir treated rats.Methods: Swiss albino rats were divided into five Groups of six animals each. All the groups (except control) were treated with indinavir 216 mg/kg (oral) for 15 days. Group I (control) received normal saline (oral) from day 8 to day 15, Group II received indinavir 216 mg/kg (oral), Group III received S. chirata plant extract 500 mg/kg (oral) from day 8 to day 15, Group IV received pioglitazone 4 mg/kg (oral) from day 8 to day 15, and Group V received metformin 36 mg/kg (oral) from day 8 to day 15. The biochemical parameters such as serum glucose, insulin, and lipid levels were measured on day 15. Results were analyzed using one-way analysis of variance followed by Bonferroni's multiple comparison test.Results: Indinavir (216 mg/kg) treated rats showed a significant (p<0.05) increase in glucose and insulin levels and also altered lipid levels. This indicates indinavir produces diabetic-like state in rats. S. chirata extract (500 mg/kg) decreases glucose and insulin levels and also improves lipid levels the effect is almost similar to metformin and pioglitazone.Conclusion: Indinavir causes elevated glucose, insulin and lipid levels, so care must be taken while prescribing indinavir for HIV patients. Treatment with S. chirata extract improved the altered glucose, insulin, and lipid profile in indinavir treated rats.Key words: Indinavir, Insulin resistance, Diabetes dyslipidemia, Glucose intolerance
