Genome-wide and Mendelian randomisation studies of liver MRI yield insights into the pathogenesis of steatohepatitis
Background
A non-invasive method to grade the severity of steatohepatitis and liver fibrosis is magnetic resonance imaging (MRI) based corrected T1 (cT1). We aimed to identify genetic variants influencing liver cT1 and use genetics to understand mechanisms underlying liver fibroinflammatory disease and its link with other metabolic traits and diseases.
Methods
First, we performed a genome-wide association study (GWAS) in 14,440 Europeans in UK Biobank with liver cT1 measures. Second, we explored the effects of the cT1 variants on liver blood tests, and a range of metabolic traits and diseases. Third, we used Mendelian randomisation to test the causal effects of 24 predominantly metabolic traits on liver cT1 measures.
Results
We identified six independent genetic variants associated with liver cT1 that reached GWAS significance threshold (p<5x10-8). Four of the variants (rs75935921 in SLC30A10, rs13107325 in SLC39A8, rs58542926 in TM6SF2, rs738409 in PNPLA3) were also associated with elevated transaminases and had variable effects on liver fat and other metabolic traits. Insulin resistance, type 2 diabetes, non-alcoholic fatty liver and BMI were causally associated with elevated cT1 whilst favourable adiposity (instrumented by variants associated with higher adiposity but lower risk of cardiometabolic disease and lower liver fat) was found to be protective.
Conclusion
The association between two metal ion transporters and cT1 indicates an important new mechanism in steatohepatitis. Future studies are needed to determine whether interventions targeting the identified transporters might prevent liver disease in at risk individuals

TutorialThe tutorial outlines the basic concepts and offers some tips on witness testing of API 610 centrifugal pumps and API 611 steam turbines. The section on API 610 pump testing discusses performance, NPSH and mechanical run tests of API 610 centrifugal pumps that are often specified with witness testing. Various parameters that are measured during testing and the related instrumentation are introduced. This section references related paragraphs and tables from ANSI/API Standard 610 (2010). The section on API 611 steam turbine testing covers the mechanical run test and cites the related paragraphs of API Standard 611 (2008). The same section also recommends a few additional tests that could be performed on general purpose steam turbines, if feasible and applicable
Density functional theory of spin-polarized disordered quantum dots
Using density functional theory, we investigate fluctuations of the ground
state energy of spin-polarized, disordered quantum dots in the metallic regime.
To compare to experiment, we evaluate the distribution of addition energies and
find a convolution of the Wigner-Dyson distribution, expected for noniteracting
electrons, with a narrower Gaussian distribution due to interactions. The tird
moment of the total distribution is independent of interactions, and so is
predicted to decrease by a factor of 0.405 upon application of a magnetic field
which transforms from the Gaussian orthogonal to the Gaussian unitary ensemble.Comment: 13 pages, 2 figure
Quantum dots in magnetic fields: thermal response of broken symmetry phases
We investigate the thermal properties of circular semiconductor quantum dots
in high magnetic fields using finite temperature Hartree-Fock techniques. We
demonstrate that for a given magnetic field strength quantum dots undergo
various shape phase transitions as a function of temperature, and we outline
possible observable consequences.Comment: In Press, Phys. Rev. B (2001
Haloperidol and Ziprasidone for Treatment of Delirium in Critical Illness
BACKGROUND:
There are conflicting data on the effects of antipsychotic medications on delirium in patients in the intensive care unit (ICU).
METHODS:
In a randomized, double-blind, placebo-controlled trial, we assigned patients with acute respiratory failure or shock and hypoactive or hyperactive delirium to receive intravenous boluses of haloperidol (maximum dose, 20 mg daily), ziprasidone (maximum dose, 40 mg daily), or placebo. The volume and dose of a trial drug or placebo was halved or doubled at 12-hour intervals on the basis of the presence or absence of delirium, as detected with the use of the Confusion Assessment Method for the ICU, and of side effects of the intervention. The primary end point was the number of days alive without delirium or coma during the 14-day intervention period. Secondary end points included 30-day and 90-day survival, time to freedom from mechanical ventilation, and time to ICU and hospital discharge. Safety end points included extrapyramidal symptoms and excessive sedation.
RESULTS:
Written informed consent was obtained from 1183 patients or their authorized representatives. Delirium developed in 566 patients (48%), of whom 89% had hypoactive delirium and 11% had hyperactive delirium. Of the 566 patients, 184 were randomly assigned to receive placebo, 192 to receive haloperidol, and 190 to receive ziprasidone. The median duration of exposure to a trial drug or placebo was 4 days (interquartile range, 3 to 7). The median number of days alive without delirium or coma was 8.5 (95% confidence interval [CI], 5.6 to 9.9) in the placebo group, 7.9 (95% CI, 4.4 to 9.6) in the haloperidol group, and 8.7 (95% CI, 5.9 to 10.0) in the ziprasidone group (P=0.26 for overall effect across trial groups). The use of haloperidol or ziprasidone, as compared with placebo, had no significant effect on the primary end point (odds ratios, 0.88 [95% CI, 0.64 to 1.21] and 1.04 [95% CI, 0.73 to 1.48], respectively). There were no significant between-group differences with respect to the secondary end points or the frequency of extrapyramidal symptoms.
CONCLUSIONS:
The use of haloperidol or ziprasidone, as compared with placebo, in patients with acute respiratory failure or shock and hypoactive or hyperactive delirium in the ICU did not significantly alter the duration of delirium. Resonant transmission through an open quantum dot
We have measured the low-temperature transport properties of a quantum dot
formed in a one-dimensional channel. In zero magnetic field this device shows
quantized ballistic conductance plateaus with resonant tunneling peaks in each
transition region between plateaus. Studies of this structure as a function of
applied perpendicular magnetic field and source-drain bias indicate that
resonant structure deriving from tightly bound states is split by Coulomb
charging at zero magnetic field.Comment: To be published in Phys. Rev. B (1997). 8 LaTex pages with 5 figure

The first multi-wavelength campaign of AXP 4U 0142+61 from radio to hard X-rays
For the first time a quasi-simultaneous multi-wavelength campaign has been
performed on an Anomalous X-ray Pulsar from the radio to the hard X-ray band.
4U 0142+61 was an INTEGRAL target for 1 Ms in July 2005. During these
observations it was also observed in the X-ray band with Swift and RXTE, in the
optical and NIR with Gemini North and in the radio with the WSRT. In this paper
we present the source-energy distribution. The spectral results obtained in the
individual wave bands do not connect smoothly; apparently components of
different origin contribute to the total spectrum. Remarkable is that the
INTEGRAL hard X-ray spectrum (power-law index 0.79 +/- 0.10) is now measured up
to an energy of ~230 keV with no indication of a spectral break. Extrapolation
of the INTEGRAL power-law spectrum to lower energies passes orders of magnitude
underneath the NIR and optical fluxes, as well as the low ~30 microJy (2 sigma)
upper limit in the radio band.Comment: 6 pages, 1 figure. To be published in the proceedings of the

Magnetic Moments of Heavy Baryons Using Effective Mass and Screened Charge Scheme
Magnetic moments of heavy charmed baryons with are predicted
employing the concept of effective quark mass and screened charge of quark. We
also extend our scheme to predict the transition magnetic
moments. A comparison of our results with the predictions obtained in recent
models is presented.Comment: 19 pages, Accepted for publication in EPJ-
Plane-symmetric inhomogeneous magnetized viscous fluid universe with a variable
The behavior of magnetic field in plane symmetric inhomogeneous cosmological
models for bulk viscous distribution is investigated. The coefficient of bulk
viscosity is assumed to be a power function of mass density . The values of cosmological constant for these models are
found to be small and positive which are supported by the results from recent
supernovae Ia observations. Some physical and geometric aspects of the models
are also discussed.Comment: 18 pages, LaTex, no figur
