    Proliferative analysis of trophoblastic cells in cattle

    The aim of the present study was to evaluate the cell proliferative activity, by AgNORs number, in different regions of bovine placenta throughout gestation. A total of 28 bovine placentas were separated into four groups: group I (60 to 120 days), group II (121 to 170 days), group III (171 to 220 days), and group IV (221 to 290 days). It was found a greater number of AgNORs in giant trophoblastic cells (GTC) when compared with mononuclear trophoblastic cells (MTC) (p<0,001) in all regions and gestational groups analyzed, that confirms their intensive synthesis activity in trophoblast epithelium. The central region of the placentome begins an intense proliferative activity in group II, observed by clusters, while placentomes edges showed a higher number of clusters on group III. These data suggest that the central region of the placentomes began an intense proliferative activity prior to its edge, both declines at the end of pregnancy. Interplacentomal area showed a higher number of AgNORs in the group IV, suggesting a higher proliferative activity of these cells at the end of pregnancy. The results of this study indicate that the proliferative activity, as determined by the amount of intranuclear AgNORs, exhibits patterns that are not only specific to each type of trophoblastic cells, but also for each specific region of bovine placenta throughout pregnancy.Este estudo teve como objetivo analisar atividade proliferativa das células trofoblásticas, através da quantificação de AgNORs, em diferentes regiões da placenta bovina ao longo da gestação. Foram utilizados 28 úteros, sendo estes agrupados de acordo com as idades gestacionais: grupo I (60-120 dias); II (121- 170 dias); III (171-220 dias) e IV (221-290 dias). Foi encontrado um número significativamente maior de AgNORs nas células trofoblásticas gigantes (CTG) em relação às mononucleadas (CTM) (p<0,001) em todas as regiões e grupos gestacionais analisados, o que confirma sua intensa atividade de síntese no epitélio trofoblástico. A região central do placentônio inicia uma atividade proliferativa mais intensa já no grupo II, observada pelo número de clusters, enquanto que a margem do placentônio apresenta uma maior quantidade de clusters no grupo III. Estes dados sugerem que a região central do placentônio inicia uma intensa atividade proliferativa anteriormente a sua margem, ambas declinando no final da gestação. A área interplacentomal apresentou um maior número de AgNORs no último grupo gestacional, sugerindo uma maior atividade proliferativa dessas células no final da prenhez. Os resultados deste estudo indicam que a atividade proliferativa, determinada pela quantidade de AgNORs intranucleares, exibe padrões que são específicos não somente para cada tipo de célula trofoblástica, mas também para cada região específica da placenta bovina ao longo da gestação

    Kids Club Afterschool es un servicio dirigido a niños de primaria (de seis a once años de edad) que estructura su tiempo combinando la realización de tareas acompañados de profesores especializados con el desarrollo de actividades lúdicas que complementarán su formación integral en un ambiente seguro y agradable. Los factores diferenciadores que caracterizan a Kids Club Afterschool son asesoría de tareas, variedad de actividades y horario flexible. Las dos primeras resuelven la problemática y el horario flexible aporta un valor extra al dar la facilidad a los padres de recoger a sus hijos cuando terminen de trabajar. La capacidad proyectada por local es de 186 niños, que para el caso del primer local representa la captación de un 5% de los usuarios potenciales. Para la apertura de nuevos locales se apuntará como máximo al 25% de los usuarios potenciales por ubicación. Para efectos del plan de negocios presentado se está considerando hasta 12%. Este porcentaje garantiza la viabilidad de captación y apertura de los ocho locales planteados al año 4. Se estableció contacto con la red de colegios Pamer con más de treinta años en el negocio de educación en el Perú. Este plan de negocio contempla una alianza con esta cadena. Lo interesante de Pamer es que la localización de sus locales está alineada con lo que se busca para Kids Club Afterschool tanto a corto como a mediano plazo en las proyecciones de expansión. El despliegue del negocio se efectuará por fases. Se contará con un solo local en la primera fase y en la fase cuatro se llegará a tener ocho locales. El primer local servirá como prueba para validar el modelo de negocio, luego de ello se abrirán los siguientes locales. La inversión requerida es de S/. 164 mil, con una estructura deuda-capital de 50%-50%. La proyección de ventas para el año 1 es de S/. 477 mil y S/. 29 millones acumulados para el año 6. El valor presente neto de los seis primeros años de un local es de S/. 346 mil y para todo el proyecto es de S/. 979 mil, considerando una tasa de descuento del 19,75% anual

    Nutritive value of rations containing sugar cane, poultry litter and corn

    Realizou-se, no campus da Faculdade de Ciências Agrárias e Veterinárias da UNESP, em Jaboticabal, SP, um estudo com o objetivo de determinar a ingestão voluntária e a digestibilidade aparente de rações contendo diferentes relações de cana-de-açúcar: cama-de-frango (CC1 = 65,50 : 34,50%; CC2 = 58,00 : 42,00%; e CC3 - 52,00 : 48,00% combinadas com três níveis de milho (M1 = 0 kg; M2 = 0,75 kg e M3 = 1,50 kg), através do delineamento em blocos casualizados em esquema fatorial 3 com três repetições. Utilizaram-se nove bezerros pesando, em média, 334 kg. Através da análise de regressão, detectou-se efeito linear positivo (P < 0,01) entre os níveis de milho e a digestibilidade aparente da matéria seca, da matéria orgânica, do extrato etéreo, do extrato não nitrogenado e dos teores de nutrientes digestíveis totais. As proporções de cana: cama-de-frango com menores quantidades de cana proporcionaram aumentos na digestibilidade da proteína e da fibra bruta (P < 0,01). Não houve interação significativa entre os níveis de milho e as proporções de cana: cama-de-frango. O valor nutritivo e a ingestão de matéria seca total foram significativamente aumentadas nas rações com os níveis crescentes de milho (P < 0,01).This work was conduced at Faculdade de Ciências Agrárias e Veterinárias, Campus de Jaboticabal-UNESP, São Paulo, Brazil, with the objective of finding out the intake and apparent digestibility of the nutrients in rations with different proportions of sugar cane (SC) and poultry litter (PL): 65.50 : 34.50%; 58.00 : 42.00% and 52.00 : 48.00% combined with three levels of corn: 0; 0.15 kg an 1,50 kg. Treatments were studied according to a completely randomized block design 32 with three replications. Nine steers with an initial average weight of 334 kg were used. Linear regression studies showed significant effects (P < 0.01) of the different levels of corn on apparent digestibility of the dry matter, organic matter, ether extract, nitrogen free extract and total digestible nutrients. Proportions of SC/PL with lower sugarcane ration increased crude fiber and protein digestibility (P < 0.01). There was no significant interaction of the levels of corn and different proportions of sugarcane to poultry litter. Nutritive value and total intake of dry matter increased with the levels of corn in the ration (P < 0.01)

    Validating the predictive ability of the 2MACE score for major adverse cardiovascular events in patients with atrial fibrillation:results from phase II/III of the GLORIA-AF registry

    The 2MACE score was specifically developed as a risk-stratification tool in atrial fibrillation (AF) to predict cardiovascular outcomes. We evaluated the predictive ability of the 2MACE score in the GLORIA-AF registry. All eligible patients from phase II/III of the prospective global GLORIA-AF registry were included. Major adverse cardiac events (MACEs) were defined as the composite outcome of stroke, myocardial infarction and cardiovascular death. Cox proportional hazards were used to examine the relationship between the 2MACE score and study outcomes. Predictive capability of the 2MACE score was investigated using receiver-operating characteristic curves. A total of 25,696 patients were included (mean age 71 years, female 44.9%). Over 3 years, 1583 MACEs were recorded. Patients who had MACE were older, with more cardiovascular risk factors and were less likely to be managed using a rhythm-control strategy. The median 2MACE score in the MACE and non-MACE groups were 2 (IQR 1–3) and 1 (IQR 0–2), respectively (p &lt; 0.001). The 2MACE score was positively associated with an increase in the risk of MACE, with a score of ≥ 2 providing the best combination of sensitivity (69.6%) and specificity (51.6%), HR 2.47 (95% CI, 2.21–2.77). The 2MACE score had modest predictive performance for MACE in patients with AF (AUC 0.655 (95% CI, 0.641–0.669)). Our analysis in this prospective global registry demonstrates that the 2MACE score can adequately predict the risk of MACE (defined as myocardial infarction, CV death and stroke) in patients with AF.     Temporal single-cell regeneration studies: the greatest thing since sliced pancreas?

    The application of single-cell analytic techniques to the study of stem/progenitor cell niches supports the emerging view that pancreatic cell lineages are in a state of flux between differentiation stages. For all their value, however, such analyses merely offer a snapshot of the cellular palette of the tissue at any given time point. Conclusions about potential developmental/regeneration paths are solely based on bioinformatics inferences. In this context, the advent of new techniques for the long-term culture and lineage tracing of human pancreatic slices offers a virtual window into the native organ and presents the field with a unique opportunity to serially resolve pancreatic regeneration dynamics at the single-cell level. Single-cell RNA-seq (scRNA-seq) is now widely available due to the advent of microfluidics methods that enable the study of thousands of cells.Many reports have dissected the human pancreas’ composition, revealing heterogeneity within subpopulations hitherto believed to be homogeneous. These observations have been interpreted as a reflection of underlying developmental/regenerative plasticity.However, all scRNA-seq analyses are only snapshots of the cell composition at the time of collection. Inferring time-dependent trajectories is akin to figuring out a movie’s storyline from a single frame. New bioinformatics tools have been used to gather latent temporal information from data sets obtained at single time points, but their accuracy has not been validated.Long-term organotypic cultures of the human pancreas may offer the means to conduct true temporal analysis of regeneration at the single-cell level
