    Traditional decoction and PUAE aqueous extracts of pomegranate peels as potential low-cost anti-tyrosinase ingredients

    open6The aim of the study is to evaluate the anti-tyrosinase activity of dierent aqueous extracts obtained from pomegranate juice processing by-products. External pomegranate peels of two certified cultivars (Akko and Wonderful), were extracted using only water as the extraction solvent. A traditional decoction and a pulsed ultrasound-assisted extraction (PUAE), both 10 min long, were performed and compared. All the aqueous extracts proved to be rich in bioactive compounds. In particular, the total phenolic content (TPC) ranged from 148 to 237 mg gallic acid equivalent (GAE)/g of dried peels (DW), the radical-scavenging ability (RSA) ranged from 307 to 472 mg ascorbic acid equivalent (AAE)/g DW, the free ellagic acid content (EA) ranged from 49 to 94 g/mL, and the ellagitannins (ETs) ranged from 242 to 340 g/mL. For both cultivars, PUAE extracts had higher ET content and a lower EC50, while the decoctions had slightly higher TPC, RSA, and free EA amounts. Principal component analysis (PCA) highlighted the direct correlation between the ET content and the tyrosinase enzyme inhibition (lower values of EC50).     Functional over-redundancy and vulnerability of lichen communities decouple across spatial scales and environmental severity

    According to the insurance hypothesis, high taxonomic diversity should ensure ecosystem stability because of functional redundancy, whereas reduced functional diversity that results from species loss should affect ecosystem sensitivity, resilience, and vulnerability. However, even in species-rich ecosystems, functional over-redundancy (FOR; i.e., the tendency of most species to cluster into a few over-represented functional entities) in some cases may result in under-representation of many functions, and the ecosystem might become highly vulnerable. Using a stratified random sampling design with nested spatial levels (nine land use strata, 70 plots, 435 trees/rock outcrops, and 9845 quadrats), we recorded the occurrence of over 350 species of epiphytic and rock-dwelling lichens in semi-arid ecosystems in western Sardinia, where solar radiation defines a wide environmental gradient. By accounting for species functional traits, such as growth form, photosynthetic strategies, and reproductive strategies, we obtained 43 functional entities (>60% of all possible combinations) and tested the scale-dependency of FOR and functional vulnerability (FV, i.e., the risk of losing functional entities) by generalized linear mixed models. We found that FOR increased and FV decreased with increasing spatial scale, which supports the hypothesis of a cross-scale functional reinforcement. Decoupling of FOR and FV was far more evident for rock-dwelling compared with epiphytic communities, which reflects differing environmental conditions associated with substrate type. Our results indicate that increased warming and climatic extremes could exacerbate species clustering into the most resistant functional entities and thus enhance FOR at the community level. Therefore, high taxonomic diversity may not ensure systematic buffering of climate change impacts

    Eucalyptus cinerea and E. nicholii by-Products as Source of Bioactive Compounds for Agricultural Applications

    The cultivation of different species of Eucalyptus has recently expanded in Liguria (Italy) due to the growing demand of the North European floricultural market. Eucalyptus tree branches are cut and selected for their quality, resulting in large amounts of waste biomass to be disposed of. The aim of our study was to evaluate the phytotoxic and antimicrobial activities of essential oils (EOs) from pruning wastes of E. cinerea (EC) and E. nicholii (EN), for potential applications in agriculture. Phytochemical analyses showed eucalyptol (1,8-cineole) as the major component in both EOs, but the EO yield of EN was higher than that of EC, in agreement with a significantly higher oil gland density on EN leaves. EOs from both species showed phytotoxicity on both weeds tested, but no significant inhibition on horticultural crop seed germination, except for Raphanus sativus. The EO from EC showed the strongest antibacterial activity, while the EO from EN showed the strongest antifungal activity. Concluding, EOs from Eucalyptus pruning may be used as possible alternatives to synthetic herbicides and pesticides, acting as antimicrobial and antifungal agents, thus representing a safe strategy for crop management programs

    Construction of a YAC contig covering human chromosome 6p22

    A contig covering human chromosome 6p22 that consists of 134 YAC clones aligned based on the presence/absence of 52 DNA markers is presented. This contig overlaps with the 6p23 contig at its telomeric end and with the 6p21.3 contig at its centromeric end. The order of loci within the contig resolves the relative positions of several genetically mapped markers. Among the additional markers used here, there are eight novel PCR assays. The 12 known genes and anonymous ESTs located within the contig establish a first step toward a transcriptional map of this region.     Depigmenting potential of lichen extracts evaluated by in vitro and in vivo tests

    Melanin is the main pigment of human skin, playing the primary role of protection from ultraviolet radiation. Alteration of the melanin production may lead to hyperpigmentation diseases, with both aesthetic and health consequences. Thus, suppressors of melanogenesis are considered useful tools for medical and cosmetic treatments. A great interest is focused on natural sources, aimed at finding safe and quantitatively available depigmenting substances. Lichens are thought to be possible sources of this kind of compounds, as the occurrence of many phenolic molecules suggests possible effects on phenolase enzymes involved in melanin synthesis, like tyrosinase. In this work, we used four lichen species, Cetraria islandica Ach., Flavoparmelia caperata Hale, Letharia vulpina (L.) Hue, and Parmotrema perlatum (Hudson) M. Choisy, to obtain extracts in solvents of increasing polarity, viz. chloroform, chloroform-methanol, methanol, and water. Cell-free, tyrosinase inhibition experiments showed highest inhibition for L. vulpina methanol extract, followed by C. islandica chloroform-methanol one. Comparable results for depigmenting activities were observed by means of in vitro and in vivo systems, such as MeWo melanoma cells and zebrafish larvae. Our study provides first evidence of depigmenting effects of lichen extracts, from tyrosinase inhibition to cell and in vivo models, suggesting that L. vulpina and C. islandica extracts deserve to be further studied for developing skin-whitening products

    Expected performance of the ASTRI-SST-2M telescope prototype

    ASTRI (Astrofisica con Specchi a Tecnologia Replicante Italiana) is an Italian flagship project pursued by INAF (Istituto Nazionale di Astrofisica) strictly linked to the development of the Cherenkov Telescope Array, CTA. Primary goal of the ASTRI program is the design and production of an end-to-end prototype of a Small Size Telescope for the CTA sub-array devoted to the highest gamma-ray energy region. The prototype, named ASTRI SST-2M, will be tested on field in Italy during 2014. This telescope will be the first Cherenkov telescope adopting the double reflection layout in a Schwarzschild-Couder configuration with a tessellated primary mirror and a monolithic secondary mirror. The collected light will be focused on a compact and light-weight camera based on silicon photo-multipliers covering a 9.6 deg full field of view. Detailed Monte Carlo simulations have been performed to estimate the performance of the planned telescope.
