    A microsecond-response polymer-stabilized blue phase liquid crystal

    A polymer-stabilized blue-phase liquid crystal (BPLC) with microsecond response time is demonstrated using a vertical field switching cell. The measured decay time is 39 mu s at room temperature (21 degrees C) and then decreases to 9.6 mu s at 44.3 degrees C. Such a response time is 1-2 orders of magnitude faster than that of a typical BPLC device. The responsible physical mechanisms are the collective effects of short pitch length, strong polymer network, and low viscosity through temperature effect. The on-state voltage of our BPLC device is 44.2 V, hysteresis is below 0.7%, and contrast ratio is over 1300:1

    Changes in Physiological Parameters, Lipid Metabolism, and Expression of MicroRNAs in Genetically Improved Farmed Tilapia (Oreochromis niloticus) With Fatty Liver Induced by a High-Fat Diet

    Tilapia is susceptible to hepatic steatosis when grown in intensive farming systems. The aim of this study was to explore the mechanism of fatty liver induced by a high-fat diet (HFD) in genetically improved farmed tilapia (GIFT, Oreochromis niloticus). Juvenile GIFT were fed with HFD or a normal-fat diet (NFD) for 60 days. Substantial fat deposition in the liver of HFD-fed GIFT on days 20, 40, and 60 was observed using hematoxylin – eosin staining and oil red O staining. The increased fat deposition was consistent with increased triglyceride (TG) and total cholesterol (TC) levels in the liver of HFD-fed GIFT. There were significant differences (P < 0.05) in serum biochemical indexes (TG, TC, low density lipoprotein-cholesterol, and insulin contents, and alanine aminotransferase activity) between GIFT fed a HFD and GIFT fed a NFD on days 20, 40, and 60. Furthermore, 60 days of a HFD significantly changed (P < 0.05) the hepatic fatty acid composition, and led to increased polyunsaturated fatty acid levels and decreased saturated fatty acid and monounsaturated fatty acid levels. Hepatic antioxidant enzyme activities increased by day 20 and then declined, which led to an increase in malondialdehyde contents in the liver of HFD-fed GIFT. Molecular analyses revealed that the microRNAs miR-122, miR-29a, and miR-145-5p were upregulated, whereas miR-34a was downregulated in HFD-fed GIFT. SCD, ELOVL6, and SRD5A2 encode three important enzymes in lipid metabolism, and were identified as potential targets of miRNAs. The transcript levels of hepatic SCD and ELOVL6 were decreased and that of hepatic SRD5A2 was increased in GIFT fed a HFD. Overall, the results of this study revealed a potential link between miRNAs and fatty liver induced by HFD, and suggest that a HFD could lead to excess fat deposition in the GIFT liver, which may disrupt hepatic lipid metabolism and reduce the antioxidant defense capacity

    Effects of Growth Hormone Treatment on Height, Weight, and Obesity in Taiwanese Patients with Prader-Willi Syndrome

    BackgroundInformation regarding the efficacy of growth hormone (GH) therapy in Asian Prader-Willi syndrome (PWS) patients is lacking. We report our experience with GH treatment in children with PWS in Taiwan.MethodsForty-six PWS patients (27 males, 19 females; age range, 1 year 4 months to 13 years 7 months) who received and/or who are currently receiving GH treatment (0.1 IU/kg/day subcutaneously) for a period from 1 year to 3 years were retro-spectively analyzed. We evaluated height, weight, body mass index (BMI) and Rohrer index, before and after GH treatment.ResultsAfter patients had received GH for 1, 2 and 3 years, a significant improvement in mean height standard deviation score (SDS) was noted from −1.24 to −0.31 (p <0.01), 0.00 (p <0.001) and −0.26 (p <0.001), respectively. Mean BMI SDS decreased significantly from 1.93 to 1.13 (p <0.05) after 1 year of treatment; however, no significant changes were observed afterward. Mean Rohrer index decreased significantly, from 224.2 to 186.6 (p <0.001), 178.9 (p <0.001) and 169.3 (p <0.001). No significant gender or genotype pattern differences were noted among the 4 parameters examined.ConclusionThis 3-year, retrospective study indicates that PWS patients benefit from GH therapy in height increase and improved body composition

    Dielectric dispersion on the Kerr constant of blue phase liquid crystals

    Dielectric dispersions on the Kerr constant of two polymer-stabilized blue phase liquid crystals (BPLCs) are investigated. An extended Cole-Cole model is proposed to fit the experimental results and good agreement is obtained. As the electric field frequency increases, Kerr constant decreases and the associated dielectric heating effect gradually increases. These results will undoubtedly affect the high frequency operation of BPLC devices

    Isotretinoin Oil-Based Capsule Formulation Optimization

    The purpose of this study was to develop and optimize an isotretinoin oil-based capsule with specific dissolution pattern. A three-factor-constrained mixture design was used to prepare the systemic model formulations. The independent factors were the components of oil-based capsule including beeswax (X1), hydrogenated coconut oil (X2), and soybean oil (X3). The drug release percentages at 10, 30, 60, and 90 min were selected as responses. The effect of formulation factors including that on responses was inspected by using response surface methodology (RSM). Multiple-response optimization was performed to search for the appropriate formulation with specific release pattern. It was found that the interaction effect of these formulation factors (X1X2, X1X3, and X2X3) showed more potential influence than that of the main factors (X1, X2, and X3). An optimal predicted formulation with Y10 min, Y30 min, Y60 min, and Y90 min release values of 12.3%, 36.7%, 73.6%, and 92.7% at X1, X2, and X3 of 5.75, 15.37, and 78.88, respectively, was developed. The new formulation was prepared and performed by the dissolution test. The similarity factor f2 was 54.8, indicating that the dissolution pattern of the new optimized formulation showed equivalence to the predicted profile

    A-Dorf liegt im Einzugsgebiet von zwei Großstädten und besteht seit dem Jahr 953. Ursprünglich und für den größten Teil seiner Geschichte war das Dorf eine bäuerliche Siedlung, hat aber seit den 1950er-Jahren einen deutlichen sozialstrukturellen Wandel durchgemacht. Die Einwohnerschaft hat sich seit den 1970er-Jahren durch Neubaugebiete nahezu verdoppelt. Insbesondere die Zugezogenen weisen eine eher pragmatische, zweckrationale Ortsbindung auf und sind überlokal orientiert. Ihr Lebens- und Aktionsraum geht weit über die Grenzen des Dorfes hinaus. Dies hat auch Konsequenzen für dörfliche Formen der Gemeinschaftsbildung, die inzwischen weit stärker Ergebnisse von individuellen Selektionen sind und nicht mehr durch Strukturen des Kontextes erzwungen werden. Sehr deutlich wurde dies z. B. bei den Beziehungen zu Nachbarn/ innen oder dörflichen Vereinen und Interessengruppen. »Die« Dorfgemeinschaft im Singular wird ersetzt durch interessenspezifische Teilgemeinschaften.»A-Village« is located in the catchment area of two major cities and exists since 1953. Originally, and for the most part of its history, it was a rural village, but since the 1950s it went through a major change concerning its social structure. The number of inhabitants has almost doubled since the 1970s, due to housing estates. Especially the recent settlers have a more pragmatic and function-guided binding to the village and are globally orientated. Their space of living and action exceeds the narrow boundaries of the village to a greater extent. These facts have consequences for the way of forming communities, and are less the implications of contextual structures, but reflect more results of selections by individuals. This is evident, for example, in the case of relations with neighbours or associations and interest groups. The »village-community« in the singular is being replaced by segmented communities, which are being formed to serve special interests

    Loss of FHL1 induces an age-dependent skeletal muscle myopathy associated with myofibrillar and intermyofibrillar disorganization in mice

    Recent human genetic studies have provided evidences that sporadic or inherited missense mutations in four-and-a-half LIM domain protein 1 (FHL1), resulting in alterations in FHL1 protein expression, are associated with rare congenital myopathies, including reducing body myopathy and Emery–Dreifuss muscular dystrophy. However, it remains to be clarified whether mutations in FHL1 cause skeletal muscle remodeling owing to gain- or loss of FHL1 function. In this study, we used FHL1-null mice lacking global FHL1 expression to evaluate loss-of-function effects on skeletal muscle homeostasis. Histological and functional analyses of soleus, tibialis anterior and sternohyoideus muscles demonstrated that FHL1-null mice develop an age-dependent myopathy associated with myofibrillar and intermyofibrillar (mitochondrial and sarcoplasmic reticulum) disorganization, impaired muscle oxidative capacity and increased autophagic activity. A longitudinal study established decreased survival rates in FHL1-null mice, associated with age-dependent impairment of muscle contractile function and a significantly lower exercise capacity. Analysis of primary myoblasts isolated from FHL1-null muscles demonstrated early muscle fiber differentiation and maturation defects, which could be rescued by re-expression of the FHL1A isoform, highlighting that FHL1A is necessary for proper muscle fiber differentiation and maturation in vitro. Overall, our data show that loss of FHL1 function leads to myopathy in vivo and suggest that loss of function of FHL1 may be one of the mechanisms underlying muscle dystrophy in patients with FHL1 mutations
