    Femoral non-union above a fused knee: Successful treatment with exchange nailing and intramedullary delivery of bone morphogenetic protein

    Bone healing is a complex and well-orchestrated physiological process, in which bone repairs and regenerates regaining its original biomechanical and biochemical properties. It is estimated that 5 to 10% of all fractures are complicated by delayed union or non-union. Progression to non-union is thought to be multifactorial, even though the exact biological sequence remains obscure. Treatment should aim to addressing deficiencies in both the mechanical and biological components, along with eliminating co-factors that could negatively affect the locally induced fracture healing response. We report a case of a 78-year-old patient who presented with a distal femoral non-union above a previously fused knee, which was successfully managed with exchange nailing and intramedullary delivery of recombinant human bone morphogenetic protein-7 (rhBMP-7)

    Siamese Tracking of Cell Behaviour Patterns

    Tracking and segmentation of biological cells in video sequences is a challenging problem, especially due to the similarity of the cells and high levels of inherent noise. Most machine learning-based approaches lack robustness and suffer from sensitivity to less prominent events such as mitosis, apoptosis and cell collisions. Due to the large variance in medical image characteristics, most approaches are dataset-specific and do not generalise well on other datasets. In this paper, we propose a simple end-to-end cascade neural architecture that can effectively model the movement behaviour of biological cells and predict collision and mitosis events. Our approach uses U-Net for an initial segmentation which is then improved through processing by a siamese tracker capable of matching each cell along the temporal axis. By facilitating the re-segmentation of collided and mitotic cells, our method demonstrates its capability to handle volatile trajectories and unpredictable cell locations while being invariant to cell morphology. We demonstrate that our tracking approach achieves state-of-the-art results on PhC-C2DL-PSC and Fluo-N2DH-SIM+ datasets and ranks second on the DIC-C2DH-HeLa dataset of the cell tracking challenge benchmarks

    Learning a feature space for similarity in world music

    In this study we investigate computational methods for assessing music similarity in world music styles. We use state-of-the-art audio features to describe musical content in world music recordings. Our music collection is a subset of the Smithsonian Folkways Recordings with audio examples from 31 countries from around the world. Using supervised and unsupervised dimensionality reduction techniques we learn feature representations for music similarity. We evaluate how well music styles separate in this learned space with a classification experiment. We obtained moderate performance classifying the recordings by country. Analysis of misclassifications revealed cases of geographical or cultural proximity. We further evaluate the learned space by detecting outliers, i.e. identifying recordings that stand out in the collection. We use a data mining technique based on Mahalanobis distances to detect outliers and perform a listening experiment in the ‘odd one out’ style to evaluate our findings. We are able to detect, amongst others, recordings of non-musical content as outliers as well as music with distinct timbral and harmonic content. The listening experiment reveals moderate agreement between subjects’ ratings and our outlier estimation

    The in vitro effects of platelet products on the biophysiological functions of human bone marrow mesenchymal stromal cells: a systematic review

    Platelet products (PP) and bone-marrow aspirate are popular sources of osteoinductive signalling molecules and osteogenic bone marrow mesenchymal stromal cells (BM-MSCs) used in the treatment of impaired bone healing. However, the combined use of PP and BM-MSCs in clinical studies has reported mixed results. Understanding the cellular and molecular interactions between PP and BM-MSCs plays the important role of guiding future research and clinical application. This systematic review investigates the effects of PP on the biophysiological functions of BM-MSCs in in vitro human studies, including (i) proliferation, (ii) migration, (iii) differentiation, (iv) growth factor/cytokine/protein expression, (v) immunomodulation, (vi) chemotactic effect on haematopoietic stem cells, (vii) response to apoptotic stress, and (viii) gene expression. In vitro studies in human have demonstrated the multi-faceted ‘priming effect’ of PP on the biophysiological functions of BM-MSCs. PP has been shown to improve proliferation, migration, osteogenic differentiation, reaction to apoptotic stress as well as immunomodulatory, pro-angiogenic and pro-inflammatory capacities of BM-MSCs. Several factors are highlighted that restrict the transferability of these findings into clinical practice. Therefore, more collaborative in vitro research in humans modelled to reflect clinical practice is required to better understand the effects of PP exposure on the biophysiological function(s) of BM-MSCs in human

    Tenebrio molitor larvae meal inclusion affects hepatic proteome and apoptosis and/or autophagy of three farmed fish species

    Herein, the effect of dietary inclusion of insect (Tenebrio molitor) meal on hepatic pathways of apoptosis and autophagy in three farmed fish species, gilthead seabream (Sparus aurata), European seabass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss), fed diets at 25%, 50% and 60% insect meal inclusion levels respectively, was investigated. Hepatic proteome was examined by liver protein profiles from the three fish species, obtained by two-dimensional gel electrophoresis. Although cellular stress was evident in the three teleost species following insect meal, inclusion by T. molitor, D. labrax and O. mykiss suppressed apoptosis through induction of hepatic autophagy, while in S. aurata both cellular procedures were activated. Protein abundance showed that a total of 30, 81 and 74 spots were altered significantly in seabream, European seabass and rainbow trout, respectively. Insect meal inclusion resulted in individual protein abundance changes, with less number of proteins altered in gilthead seabream compared to European seabass and rainbow trout. This is the first study demonstrating that insect meal in fish diets is causing changes in liver protein abundances. However, a species-specific response both in the above mentioned bioindicators, indicates the need to strategically manage fish meal replacement in fish diets per species

    Cascading failure in electric power systems is a complicated problem for which a variety of models, software tools, and analytical tools have been proposed but are difficult to verify. Benchmarking and validation are necessary to understand how closely a particular modeling method corresponds to reality, what engineering conclusions may be drawn from a particular tool, and what improvements need to be made to the tool in order to reach valid conclusions. The community needs to develop the test cases tailored to cascading that are central to practical benchmarking and validation. In this paper, the IEEE PES working group on cascading failure reviews and synthesizes how benchmarking and validation can be done for cascading failure analysis, summarizes and reviews the cascading test cases that are available to the international community, and makes recommendations for improving the state of the art
