    Fibroblast Growth Factor-10 (FGF-10) Mobilizes Lung-resident Mesenchymal Stem Cells and Protects Against Acute Lung Injury.

    FGF-10 can prevent or reduce lung specific inflammation due to traumatic or infectious lung injury. However, the exact mechanisms are poorly characterized. Additionally, the effect of FGF-10 on lung-resident mesenchymal stem cells (LR-MSCs) has not been studied. To better characterize the effect of FGF-10 on LR-MSCs, FGF-10 was intratracheally delivered into the lungs of rats. Three days after instillation, bronchoalveolar lavage was performed and plastic-adherent cells were cultured, characterized and then delivered therapeutically to rats after LPS intratracheal instillation. Immunophenotyping analysis of FGF-10 mobilized and cultured cells revealed expression of the MSC markers CD29, CD73, CD90, and CD105, and the absence of the hematopoietic lineage markers CD34 and CD45. Multipotency of these cells was demonstrated by their capacity to differentiate into osteocytes, adipocytes, and chondrocytes. Delivery of LR-MSCs into the lungs after LPS injury reduced the inflammatory response as evidenced by decreased wet-to-dry ratio, reduced neutrophil and leukocyte recruitment and decreased inflammatory cytokines compared to control rats. Lastly, direct delivery of FGF-10 in the lungs of rats led to an increase of LR-MSCs in the treated lungs, suggesting that the protective effect of FGF-10 might be mediated, in part, by the mobilization of LR-MSCs in lungs

    Prediction of Drug–Target Interaction Using Dual-Network Integrated Logistic Matrix Factorization and Knowledge Graph Embedding

    Nowadays, drug–target interactions (DTIs) prediction is a fundamental part of drug repositioning. However, on the one hand, drug–target interactions prediction models usually consider drugs or targets information, which ignore prior knowledge between drugs and targets. On the other hand, models incorporating priori knowledge cannot make interactions prediction for under-studied drugs and targets. Hence, this article proposes a novel dual-network integrated logistic matrix factorization DTIs prediction scheme (Ro-DNILMF) via a knowledge graph embedding approach. This model adds prior knowledge as input data into the prediction model and inherits the advantages of the DNILMF model, which can predict under-studied drug–target interactions. Firstly, a knowledge graph embedding model based on relational rotation (RotatE) is trained to construct the interaction adjacency matrix and integrate prior knowledge. Secondly, a dual-network integrated logistic matrix factorization prediction model (DNILMF) is used to predict new drugs and targets. Finally, several experiments conducted on the public datasets are used to demonstrate that the proposed method outperforms the single base-line model and some mainstream methods on efficiency

    Sevoflurane Induces a Cyclophilin D-Dependent Decrease of Neural Progenitor Cells Migration

    Clinical studies have suggested that repeated exposure to anesthesia and surgery at a young age may increase the risk of cognitive impairment. Our previous research has shown that sevoflurane can affect neurogenesis and cognitive function in young animals by altering cyclophilin D (CypD) levels and mitochondrial function. Neural progenitor cells (NPCs) migration is associated with cognitive function in developing brains. However, it is unclear whether sevoflurane can regulate NPCs migration via changes in CypD. To address this question, we treated NPCs harvested from wild-type (WT) and CypD knockout (KO) mice and young WT and CypD KO mice with sevoflurane. We used immunofluorescence staining, wound healing assay, transwell assay, mass spectrometry, and Western blot to assess the effects of sevoflurane on CypD, reactive oxygen species (ROS), doublecortin levels, and NPCs migration. We showed that sevoflurane increased levels of CypD and ROS, decreased levels of doublecortin, and reduced migration of NPCs harvested from WT mice in vitro and in WT young mice. KO of CypD attenuated these effects, suggesting that a sevoflurane-induced decrease in NPCs migration is dependent on CypD. Our findings have established a system for future studies aimed at exploring the impacts of sevoflurane anesthesia on the impairment of NPCs migration

    Circular RNAs: a rising star in respiratory diseases

    Abstract Circular RNAs (CircRNAs), as a new class of non-coding RNA molecules that, unlike linear RNAs, have covalently closed loop structures from the ligation of exons, introns, or both. CircRNAs are widely expressed in various organisms in a specie-, tissue-, disease- and developmental stage-specific manner, and have been demonstrated to play a vital role in the pathogenesis and progression of human diseases. An increasing number of recent studies has revealed that circRNAs are intensively associated with different respiratory diseases, including lung cancer, acute respiratory distress syndrome, pulmonary hypertension, pulmonary tuberculosis, and silicosis. However, to the best of our knowledge, there has been no systematic review of studies on the role of circRNAs in respiratory diseases. In this review, we elaborate on the biogenesis, functions, and identification of circRNAs and focus particularly on the potential implications of circRNAs in respiratory diseases

    The Impact of Zero-Mode Inrush Current of T-Hin on Zero-Sequence Overcurrent Protection and an Improved Protection with the Second Harmonic Restraint

    In recent years, the zero-mode inrush current of high-impedance transformer with built-in high-voltage winding (T-Hin), which has large amplitude and decays slowly, causes the misoperation of zero-sequence overcurrent protection. Compared with magnetizing inrush current, the waveform of zero-mode inrush current is inconsistent and irregular, and few researches have proposed the mathematical analysis as well as the improved protection using waveform characteristics. In this paper, the mathematical expression of transformer zero-mode inrush current is derived. Further considering the parameter differences, the zero-mode inrush current of T-Hin is larger, which tends to cause the misoperation. The mathematical waveforms fit well with the recorded waveforms. Both recorded waveforms and mathematical waveforms in various conditions prove that the second harmonic ratio (the ratio between the second harmonic and first harmonic) of zero-mode inrush current is significant. Based on the above analysis, a criterion based on the second harmonic ratio restraint of zero-mode inrush current is proposed. If the second harmonic ratio exceeds the setting value, it is considered that the inrush current is generated and sends a signal to restrain the protection. The theoretical setting value of the proposed criterion and the practical engineering method for determining the setting value are obtained
