    An interpolation matched interface and boundary method for elliptic interface problems

    AbstractAn interpolation matched interface and boundary (IMIB) method with second-order accuracy is developed for elliptic interface problems on Cartesian grids, based on original MIB method proposed by Zhou et al. [Y. Zhou, G. Wei, On the fictious-domain and interpolation formulations of the matched interface and boundary method, J. Comput. Phys. 219 (2006) 228–246]. Explicit and symmetric finite difference formulas at irregular grid points are derived by virtue of the level set function. The difference scheme using IMIB method is shown to satisfy the discrete maximum principle for a certain class of problems. Rigorous error analyses are given for the IMIB method applied to one-dimensional (1D) problems with piecewise constant coefficients and two-dimensional (2D) problems with singular sources. Comparison functions are constructed to obtain a sharp error bound for 1D approximate solutions. Furthermore, we compare the ghost fluid method (GFM), immersed interface method (IIM), MIB and IMIB methods for 1D problems. Finally, numerical examples are provided to show the efficiency and robustness of the proposed method

    Aberrant lysosomal carbohydrate storage accompanies endocytic defects and neurodegeneration in Drosophila benchwarmer

    Lysosomal storage is the most common cause of neurodegenerative brain disease in preadulthood. However, the underlying cellular mechanisms that lead to neuronal dysfunction are unknown. Here, we report that loss of Drosophila benchwarmer (bnch), a predicted lysosomal sugar carrier, leads to carbohydrate storage in yolk spheres during oogenesis and results in widespread accumulation of enlarged lysosomal and late endosomal inclusions. At the bnch larval neuromuscular junction, we observe similar inclusions and find defects in synaptic vesicle recycling at the level of endocytosis. In addition, loss of bnch slows endosome-to-lysosome trafficking in larval garland cells. In adult bnch flies, we observe age-dependent synaptic dysfunction and neuronal degeneration. Finally, we find that loss of bnch strongly enhances tau neurotoxicity in a dose-dependent manner. We hypothesize that, in bnch, defective lysosomal carbohydrate efflux leads to endocytic defects with functional consequences in synaptic strength, neuronal viability, and tau neurotoxicity

    Multi-scale fusion visual attention network for facial micro-expression recognition

    IntroductionMicro-expressions are facial muscle movements that hide genuine emotions. In response to the challenge of micro-expression low-intensity, recent studies have attempted to locate localized areas of facial muscle movement. However, this ignores the feature redundancy caused by the inaccurate locating of the regions of interest.MethodsThis paper proposes a novel multi-scale fusion visual attention network (MFVAN), which learns multi-scale local attention weights to mask regions of redundancy features. Specifically, this model extracts the multi-scale features of the apex frame in the micro-expression video clips by convolutional neural networks. The attention mechanism focuses on the weights of local region features in the multi-scale feature maps. Then, we mask operate redundancy regions in multi-scale features and fuse local features with high attention weights for micro-expression recognition. The self-supervision and transfer learning reduce the influence of individual identity attributes and increase the robustness of multi-scale feature maps. Finally, the multi-scale classification loss, mask loss, and removing individual identity attributes loss joint to optimize the model.ResultsThe proposed MFVAN method is evaluated on SMIC, CASME II, SAMM, and 3DB-Combined datasets that achieve state-of-the-art performance. The experimental results show that focusing on local at the multi-scale contributes to micro-expression recognition.DiscussionThis paper proposed MFVAN model is the first to combine image generation with visual attention mechanisms to solve the combination challenge problem of individual identity attribute interference and low-intensity facial muscle movements. Meanwhile, the MFVAN model reveal the impact of individual attributes on the localization of local ROIs. The experimental results show that a multi-scale fusion visual attention network contributes to micro-expression recognition

    Eps15 and Dap160 control synaptic vesicle membrane retrieval and synapse development

    Epidermal growth factor receptor pathway substrate clone 15 (Eps15) is a protein implicated in endocytosis, endosomal protein sorting, and cytoskeletal organization. Its role is, however, still unclear, because of reasons including limitations of dominant-negative experiments and apparent redundancy with other endocytic proteins. We generated Drosophila eps15-null mutants and show that Eps15 is required for proper synaptic bouton development and normal levels of synaptic vesicle (SV) endocytosis. Consistent with a role in SV endocytosis, Eps15 moves from the center of synaptic boutons to the periphery in response to synaptic activity. The endocytic protein, Dap160/intersectin, is a major binding partner of Eps15, and eps15 mutants phenotypically resemble dap160 mutants. Analyses of eps15 dap160 double mutants suggest that Eps15 functions in concert with Dap160 during SV endocytosis. Based on these data, we hypothesize that Eps15 and Dap160 promote the efficiency of endocytosis from the plasma membrane by maintaining high concentrations of multiple endocytic proteins, including dynamin, at synapses

    Mini percutaneous nephrolithotomy is a noninferior modality to standard percutaneous nephrolithotomy for the management of 20-40 mm renal calculi: A Multicenter randomized controlled trial

    Background: High quality of evidence comparing mini percutaneous nephrolithotomy (mPNL) with standard percutaneous nephrolithotomy (sPNL) for the treatment of larger-sized renal stones is lacking. Objective: To compare the efficacy and safety of mPNL and sPNL for the treatment of 20–40 mm renal stones. Design, setting, and participants: A parallel, open-label, and noninferior randomized controlled trial was performed at 20 Chinese centers (2016–2019). The inclusion criteria were patients 18–70 yr old, with normal renal function, and 20–40 mm renal stones. Intervention: Percutaneous nephrolithotomy PNL was performed using either 18 F or 24 F percutaneous nephrostomy tracts. Outcome measurements and statistical analysis: The primary outcome was the one-session stone-free rate (SFR). The secondary outcomes included operating time, visual analog pain scale (VAS) score, blood loss, complications as per the Clavien-Dindo grading system, and length of hospitalization. Results and limitations: The 1980 intention-to-treat patients were randomized. The mPNL group achieved a noninferior one-session SFR to the sPNL group by the one-side noninferiority test (0.5% [difference], p < 0.001). The transfusion and embolization rates were comparable; however, the sPNL group had a higher hemoglobin drop (5.2 g/l, p < 0.001). The sPNL yielded shorter operating time (–2.2 min, p = 0.008) but a higher VAS score (0.8, p < 0.001). Patients in the sPNL group also had longer hospitalization (0.6 d, p < 0.001). There was no statistically significant difference in fever or urosepsis occurrences. The study's main limitation was that only 18F or 24F tract sizes were used. Conclusions: Mini mPNL achieves noninferior SFR outcomes to sPNL, but with reduced bleeding, less postoperative pain, and shorter hospitalization. Patient summary: We evaluated the surgical outcomes of percutaneous nephrolithotomy using two different sizes of nephrostomy tracts in a large population. We found that the smaller tract might be a sensible alternative for patients with 20–40 mm renal stones. This multicenter, parallel, open-label, and noninferior randomized controlled trial showed that mini percutaneous nephrolithotomy achieved noninferior stone-free rate with advantages of reduced blood loss, less postoperative pain, and shorter hospitalization.
