    Genomic organization and phylogenetic utility of deer mouse (Peromyscus maniculatus) lymphotoxin-alpha and lymphotoxin-beta

    <p>Abstract</p> <p>Background</p> <p>Deer mice (<it>Peromyscus maniculatus</it>) are among the most common mammals in North America and are important reservoirs of several human pathogens, including Sin Nombre hantavirus (SNV). SNV can establish a life-long apathogenic infection in deer mice, which can shed virus in excrement for transmission to humans. Patients that die from hantavirus cardiopulmonary syndrome (HCPS) have been found to express several proinflammatory cytokines, including lymphotoxin (LT), in the lungs. It is thought that these cytokines contribute to the pathogenesis of HCPS. LT is not expressed by virus-specific CD4<sup>+ </sup>T cells from infected deer mice, suggesting a limited role for this pathway in reservoir responses to hantaviruses.</p> <p>Results</p> <p>We have cloned the genes encoding deer mouse LTα and LTβ and have found them to be highly similar to orthologous rodent sequences but with some differences in promoters elements. The phylogenetic analyses performed on the LTα, LTβ, and combined data sets yielded a strongly-supported sister-group relationship between the two murines (the house mouse and the rat). The deer mouse, a sigmodontine, appeared as the sister group to the murine clade in all of the analyses. High bootstrap values characterized the grouping of murids.</p> <p>Conclusion</p> <p>No conspicuous differences compared to other species are present in the predicted amino acid sequences of LTα or LTβ; however, some promoter differences were noted in LTβ. Although more extensive taxonomic sampling is required to confirm the results of our analyses, the preliminary findings indicate that both genes (analyzed both separately and in combination) hold potential for resolving relationships among rodents and other mammals at the subfamily level.</p

    An Upper Palaeolithic Proto-writing System and Phenological Calendar

    In at least 400 European caves such as Lascaux, Chauvet and Altamira, Upper Palaeolithic Homo sapiens groups drew, painted and engraved non-figurative signs from at least ~42,000 BP and figurative images (notably animals) from at least 37,000 BP. Since their discovery ~150 years ago, the purpose or meaning of European Upper Palaeolithic non-figurative signs has eluded researchers. Despite this, specialists assume that they were notational in some way. Using a database of images spanning the European Upper Palaeolithic, we suggest how three of the most frequently occurring signs—the line , the dot , and the —functioned as units of communication. We demonstrate that when found in close association with images of animals the line and dot constitute numbers denoting months, and form constituent parts of a local phenological/meteorological calendar beginning in spring and recording time from this point in lunar months. We also demonstrate that the sign, one of the most frequently occurring signs in Palaeolithic non-figurative art, has the meaning . The position of the within a sequence of marks denotes month of parturition, an ordinal representation of number in contrast to the cardinal representation used in tallies. Our data indicate that the purpose of this system of associating animals with calendar information was to record and convey seasonal behavioural information about specific prey taxa in the geographical regions of concern. We suggest a specific way in which the pairing of numbers with animal subjects constituted a complete unit of meaning—a notational system combined with its subject—that provides us with a specific insight into what one set of notational marks means. It gives us our first specific reading of European Upper Palaeolithic communication, the first known writing in the history of Homo sapiens

    The Impact of a Community–Wide Woodstove Changeout Intervention on Air Quality Within Two Schools

    Due to temperature inversions and widespread residential woodstove use, Libby, Montana historically experienced elevated levels of ambient woodsmoke PM2.5 throughout the winter months. In an effort to reduce wintertime PM2.5, a large community–wide woodstove changeout was conducted between 2005 and 2007, removing nearly 1 200 old polluting stoves from service. To determine the impact of this intervention on indoor air quality, PM2.5 sampling was conducted in the gymnasiums of an elementary and middle school before, during, and after the woodstove changeout over a four–year period. Throughout the program, results showed that indoor PM2.5 concentrations at the elementary school were moderately high regardless of year or season (mean±sd, 31.9±14.1 μg/m3), ranging from 11.0 μg/m3 to 79.3 μg/m3. At the middle school, the mean was 12.2±11.2 μg/m3, with no differences by season. Although there was an overall improvement in ambient air quality (and reduction of woodsmoke–PM2.5) when comparing pre– and post– changeout PM2.5 concentrations, results suggest that the community–wide woodstove changeout did not have a significant impact on indoor air quality within the gymnasiums over this same time period. These findings are supported by the results of selected chemical markers of woodsmoke measured from indoor PM (including levoglucosan) at both schools, which also demonstrated no significant reductions throughout the four–year sampling program

    Using Emotional Freedom Techniques (EFT) to Treat PTSD in Veterans: A Review of the Evidence, Survey of Practitioners, and Proposed Clinical Guidelines

    Abstract BACKGROUND: High prevalence rates of posttraumatic stress disorder (PTSD) in active military and veterans present a treatment challenge. Many PTSD studies have demonstrated the efficacy and safety of Emotional Freedom Techniques (EFT). OBJECTIVES: To develop clinical best practice guidelines for the use of EFT to treat PTSD, on the basis of the published literature, practitioner experience, and typical case histories. METHODS: We surveyed 448 EFT practitioners to gather information on their experiences with PTSD treatment. This included their demographic profiles, prior training, professional settings, use of assessments, and PTSD treatment practices. We used their responses, with the research evidence base, to formulate clinical guidelines applying the "stepped care" treatment model used by the United Kingdom's National Institute for Health and Clinical Excellence. RESULTS: Most practitioners (63%) reported that even complex PTSD can be remediated in 10 or fewer EFT sessions. Some 65% of practitioners found that more than 60% of PTSD clients are fully rehabilitated, and 89% stated that less than 10% of clients make little or no progress. Practitioners combined EFT with a wide variety of other approaches, especially cognitive therapy. Practitioner responses, evidence from the literature, and the results of a meta-analysis were aggregated into a proposed clinical guideline. CONCLUSION: We recommend a stepped care model, with 5 EFT therapy sessions for subclinical PTSD and 10 sessions for clinical PTSD, in addition to group therapy, online self-help resources, and social support. Clients who fail to respond should be referred for appropriate further care

    Community Woodstove Changeout and Impact on Ambient Concentrations of Polycyclic Aromatic Hydrocarbons and Phenolics

    A large woodstove changeout program was carried out in Libby, Montana, with the goal of reducing ambient levels of PM2.5. This provided researchers the opportunity to measure ambient concentrations of phenolic and polycyclic aromatic hydrocarbons (PAHs) before, during, and after the changeout of nearly 1200 stoves to evaluate the effectiveness of the intervention. Starting in the heating season of 2004/2005 and ending in the heating season of 2007/2008, 19 compounds were measured every three days using a high-volume polyurethane foam (PUF) sampler followed by gas chromatography and mass spectrometry analysis. Some of the organic species with the highest measured concentrations were also signature chemical markers for wood combustion. When comparing the measurements conducted during the heating season of 2004/2005 (prechangeout) to those of the heating season of 2007/2008 (postchangeout), there was a 64% average reduction in the measured concentrations of phenolics and PAHs, while the PM2.5 mass dropped by only 20% over the same time period. The results of this four year sampling program suggest that the Libby woodstove changeout program was successful in reducing overall concentrations of the measured phenolic and PAH compounds

    Residential Indoor PM\u3csub\u3e2.5\u3c/sub\u3e in Wood Stove Homes: Follow-up of the Libby Changeout Program

    In 2005 through 2008 a small rural mountain valley community engaged in a wood stove changeout program to address concerns of poor ambient air quality. During this program we assessed changes to indoor air quality before and after the introduction of a new, lower emission wood stove. We previously reported a greater than 70% reduction in indoor PM2.5 concentrations in homes following the installation of a new EPA-certified stove within the home. We report here on follow-up of the experiences in these and other homes over three winters of sample collection. In 21 homes, we compared pre-changeout PM2.5 concentrations (mean (sd) = 45.0 (33.0) μg/m3) to multiple post-changeout measures of PM2.5 concentrations using a DustTrak. The mean reduction (and 95% confidence interval) from pre-changeout to post-changeout was −18.5 μg/m3 (−31.9, −5.2), adjusting for ambient PM2.5, ambient temperature, and other factors. Findings across homes and across years were highly variable, and a subset of homes did not experience a reduction in PM2.5 following changeout. Reductions were also observed for organic carbon, elemental carbon, and levoglucosan, but increases were observed for dehydroabietic acid and abietic acid. Despite overall improvements in indoor air quality, the varied response across homes may be due to factors other than the introduction of a new wood stove

    The prevalence of low satisfaction and increased stress among faculty in academic medicine makes understanding facuity vitality in this field more important than ever before. To explore the contributors to and outcomes of faculty vitality, we conducted a multi-institutional study of faculty in academic medicine (N = 1,980, 42 percent response rate). Faculty were surveyed about climate and leadership, career and life management, satisfaction, engagement, productivity, and involvement in faculty development. Analysis reveals that controlling for other factors, academic medicine faculty who participate regularly in facuity development activ ities are significantly more satisfied, engaged, and productive
