    Development of a Fiber Optic Sensor for Online Monitoring of Thin Coatings

     The thickness measurement of gas, liquid and solid layers is not only important for the basic research on nanoscience but equally valuable in contemporary applied biomedical research. Here, we have developed an optical spectroscopy based technique for the online monitoring of thin films (coatings). A low cost light emitting diode (LED) source combined with a fiber optic bundle and grating based spectrograph have been used to generate white light interferogram. We have monitored online change of refractive index of an air film (~4 μm thickness) with temperature following the change in the intensity profile of the interferogram. A thin film of water between two cover slips (thin glass plates) has also been monitored. We have proposed a schematic for further lowering the cost of the developed instrument for the online monitoring of the coating thickness (semitransparent liquid/gas/solid films) during manufacturing/processing. A brief theoretical analysis on the detection limit of the developed technique has also been discussed in the paper

    An investigation on prevalence of Anemia in relation to BMI and nutrient intake among adult rural population of West Bengal, India

    Background: Anemia is now globally recognized major public health problem. The present study was undertaken to investigate the prevalence of anemia and interrelationship of anemia with BMI among adult rural population of West Bengal state, India. Method: Anthropometric measures, nutritional parameters, heart rate and hemoglobin were measured from 310 adult subjects by using standard technique. Results: The prevalence of anemia was high among both sexes and more than the global prevalence. The anemic individuals consumed nutritionally deficient diet than the non-anemic individuals. The prevalence of anemia was significantly high in underweight and normal groups. The Odds ratios became significantly higher in underweight group than normal and overweight/obese groups of both sexes. Conclusions: The prevalence of anemia in the adult rural population might be related to dietary deficiency and poor dietary patterns. The present study was hypothesized that the subjects with low BMI would have a higher risk of anemia compared to normal or overweight subjects

    A Design of Digital Microfluidic Biochip along with Structural and Behavioural Features in Triangular Electrode Based Array

    AbstractDigital microfluidic based biochip manoeuvres on the theory of microfluidic technology, having a broad variety of applications in chemistry, biology, environmental monitoring, military etc. Being concerned about the technological advancement in this domain, we have focused on equilateral triangular electrodes based DMFB systems. Accepting the associated design issues, here, we have addressed many facets of such electrodes regarding their structural and behavioural issues in comparison to the existing square electrodes. As the requisite voltage reduction is a key challenging design issues, to implement all the tasks using triangular electrodes that are possible in square electrode arrays as well, is a tedious job. Furthermore, to deal with this new design deploying triangular electrodes, we have analyzed all the necessary decisive factors including fluidic constraints to ensure safe droplet movements and other modular operations together with mixing and routing. Moreover, an algorithm has been developed to find a route for a given source and destination pair in this newly designed DMFB. Finally, we have included a comparative study between this new design and the existing one while encountering the above mentioned issues

    Placental Toll-Like Receptor 3 and Toll-Like Receptor 7/8 Activation Contributes to Preeclampsia in Humans and Mice

    Preeclampsia (PE) is a pregnancy-specific hypertensive syndrome characterized by excessive maternal immune system activation, inflammation, and endothelial dysfunction. Toll-like receptor (TLR) 3 activation by double-stranded RNA (dsRNA) and TLR7/8 activation by single-stranded RNA (ssRNA) expressed by viruses and/or released from necrotic cells initiates a pro-inflammatory immune response; however it is unknown whether viral/endogenous RNA is a key initiating signal that contributes to the development of PE. We hypothesized that TLR3/7/8 activation will be evident in placentas of women with PE, and sufficient to induce PE-like symptoms in mice. Placental immunoreactivity and mRNA levels of TLR3, TLR7, and TLR8 were increased significantly in women with PE compared to normotensive women. Treatment of human trophoblasts with the TLR3 agonist polyinosine-polycytidylic acid (poly I:C), the TLR7-specific agonist imiquimod (R-837), or the TLR7/8 agonist CLO97 significantly increased TLR3/7/8 levels. Treatment of mice with poly I:C, R-837, or CLO97 caused pregnancy-dependent hypertension, endothelial dysfunction, splenomegaly, and placental inflammation. These data demonstrate that RNA-mediated activation of TLR3 and TLR7/8 plays a key role in the development of PE

    Dysregulated miRNAome and Proteome of PPRV Infected Goat PBMCs Reveal a Coordinated Immune Response

    In this study, the miRNAome and proteome of virulent Peste des petits ruminants virus (PPRV) infected goat peripheral blood mononuclear cells (PBMCs) were analyzed. The identified differentially expressed miRNAs (DEmiRNAs) were found to govern genes that modulate immune response based on the proteome data. The top 10 significantly enriched immune response processes were found to be governed by 98 genes. The top 10 DEmiRNAs governing these 98 genes were identified based on the number of genes governed by them. Out of these 10 DEmiRNAs, 7 were upregulated, and 3 were downregulated. These include miR-664, miR-2311, miR-2897, miR-484, miR-2440, miR-3533, miR-574, miR-210, miR-21-5p, and miR-30. miR-664 and miR-484 with proviral and antiviral activities, respectively, were upregulated in PPRV infected PBMCs. miR-210 that inhibits apoptosis was downregulated. miR-21-5p that decreases the sensitivity of cells to the antiviral activity of IFNs and miR-30b that inhibits antigen processing and presentation by primary macrophages were downregulated, indicative of a strong host response to PPRV infection. miR-21-5p was found to be inhibited on IPA upstream regulatory analysis of RNA-sequencing data. This miRNA that was also highly downregulated and was found to govern 16 immune response genes in the proteome data was selected for functional validation vis-a-vis TGFBR2 (TGF-beta receptor type-2). TGFBR2 that regulates cell differentiation and is involved in several immune response pathways was found to be governed by most of the identified immune modulating DEmiRNAs. The decreased luciferase activity in Dual Luciferase Reporter Assay indicated specific binding of miR-21-5p and miR-484 to their target thus establishing specific binding of the miRNAs to their targets.This is the first report on the miRNAome and proteome of virulent PPRV infected goat PBMCs

    In vitro induction of nitric oxide by mouse peritoneal macrophages treated with human placental extract

    Nitric oxide (NO) is an important cellular mediator of tissue repair. It is produced in macrophages by the enzyme inducible nitric oxide synthase (iNOS) during wound healing. An aqueous extract of human placenta used as wound healer, has been investigated in terms of induction of NO by mouse peritoneal macrophages as well as human monocyte derived macrophages. NO production was estimated in macrophages culture supernatants. Incubation of 0.1 to 20 mg/ml of placental extract with 2 × 106 cells in vitro produced 10 to 100 μ M of nitrite (n = 4) in a dose dependent manner suggesting production of NO. With increase of NO production, NADPH present in the applied extract decreased proportionately. Application of L-NG monomethyl arginine (L-NMMA), an NO synthase (NOS) inhibitor, reduced the production of NO at the basal level. Dose dependent release of IFN-γ with respect to placental extract by the mouse macrophages was observed. It has been observed that human monocytes derived macrophages also produced significant amount of NO by induction of the extract. Similar induction of NO by placental extract in presence and absence of polymyxin B suggested that this property is not likely to be mediated by the endotoxin/LPS
