    Information Seeking Behaviour of Shrimp Farmers and their Perception towards Technology Dissemination through Mobile Phones

    A study was conducted among the shrimp farmers to ascertain their information seeking behaviour and perception concerning extension service through mobile phones. The findings indicate that the farmer respondents were of the perception that mobile phone-aided extension service is a viable, expedient, prompt, cost-effective and novel approach. Further they expressed that a dedicated mobile app on shrimp farming with dynamic content modules on ‘disease diagnosis, calculations for water quality corrections, feed management, pond risk management and post your query ’would be very useful for farmers and extension workers to update their capacity and sharing of field experiences. Therefore, developing a mobile app on shrimp farming, incorporating the above features would enhance the access to quality information and minimise the information gap among the shrimp farmers

    INSULIN SECRETAGOGUE EFFECT OF ROOTS OF RAVENALA MADAGASCARIENSIS SONN. - AN IN VITRO STUDY

    Objective: The objective of this study was to establish the cytotoxicity profile and to evaluate the insulin secretagogue effect of ethanolic root extract of Ravenala madagascariensis Sonn. Methods: The cell viability of rat insulinoma 5F (RIN5F) cell lines over the treatment of plant extract was assessed by 3-(4,5-dimethyl-2-thiazolyl)- 2,5-diphenyltetrazolium bromide assay. The insulin-releasing effect was evaluated by insulin secretion assay over RIN5F cell lines by enzyme-linked immunosorbent assay. Results: The ethanolic extract of the roots of R. madagascariensis Sonn. showed negligible cytotoxicity at 20–40 ÎŒg/ml, and hence, concentrations up to 40 ÎŒg/ml were used in insulin secretion assay. The ethanolic root extract at 20 and 40 ÎŒg/ml significantly (p<0.05 compared to control) stimulated the insulin release in a dose-dependent manner even in the presence of glucose at lower and higher concentrations (5 and 10 mM). Conclusion: Thus, our results validate its traditional claim in the treatment of diabetes by stimulating the secretion of insulin, thereby suggesting a possible mechanism of its antidiabetic effect

    When are the bigger fish in the small pond better citizens? A multilevel examination of relative overqualification in workgroups

Drawing upon social comparison theory and integrating with social exchange theory, we contend that employees' relative overqualification (ROQ, defined as individual overqualification relative to other group members' overqualification perceptions) is associated with their relative standing with their leader (measured as LMXSC, leader–member exchange social comparison), which in turn relates to employees' organizational citizenship behaviors (OCB). Furthermore, we assert that workgroup structural attributes and individual values (leader span of control and power distance orientation) influence the ROQ–LMXSC–OCB relationship. Multilevel modeling using data from 243 employees nested in 36 workgroups suggested patterns of moderated mediation where leader span of control and employee power distance orientation moderate the indirect link between ROQ and OCB through LMXSC. That is, the indirect relationship between ROQ and OCB is stronger in workgroups with a narrow leader span of control and for employees high in power distance orientation. Implications and directions for meso- and group-level research are discussed

    Socio-personal contour, information flow and productivity of brackishwater aquaculture systems - An Appraisal

    Brackishwater aquaculture, the farming of shrimps and fishes in coastal areas is practiced both in traditional and scientific methods. A study was conducted to assess the connectedness between the profile characteristics, their information sourcing and productivity levels. The study indicated that systems and states differ significantly (p<0.01) both in terms of profile status and productivity levels. Private extension services were dominant in scientific farming while traditional systems depend on state fisheries departments for inputs and technical advisory. Therefore, taking into account the relative strengths the state departments may partner with research institutions and input companies respectively for technology support and reaching the farmers located in remote locations. Mobile phone applications may be developed and launched for connecting the technology, inputs, services and market with the farming community

    Assessment of Antisocial Behaviour in the Juveniles

    Background: High level of crime, particularly by the people below 18 years, has been a matter of concern for many years. Assessing and preventing the antisocial behaviour in juveniles has become a policy priority.Methods: A cross sectional study was conducted on 52 males less than 18 years in an observation home in Bangalore, India. The data was collected in an interview by filling a semi structured questionnaire. Revised Wisconsin Delinquency Risk Assessment Scale (RWDRAS) was filled up simultaneously and then results were analysed.Results: Majority of the offenders were in the age group 16-18 years. In the majority the cause for antisocial behaviour was unknown followed by the family problems and poverty. The RWDRAS identified 47% of the juveniles with antisocial behaviour as low risk group.Conclusion: The Education, counselling and prevention of poverty are some of the preventive measures of antisocial behaviour. There is a need to develop a better criteria and scale for the diagnosis of antisocial behaviour as the presently used RWDRAS failed in 47% of the juveniles

    The Escherichia coli transcriptome mostly consists of independently regulated modules

    Underlying cellular responses is a transcriptional regulatory network (TRN) that modulates gene expression. A useful description of the TRN would decompose the transcriptome into targeted effects of individual transcriptional regulators. Here, we apply unsupervised machine learning to a diverse compendium of over 250 high-quality Escherichia coli RNA-seq datasets to identify 92 statistically independent signals that modulate the expression of specific gene sets. We show that 61 of these transcriptomic signals represent the effects of currently characterized transcriptional regulators. Condition-specific activation of signals is validated by exposure of E. coli to new environmental conditions. The resulting decomposition of the transcriptome provides: a mechanistic, systems-level, network-based explanation of responses to environmental and genetic perturbations; a guide to gene and regulator function discovery; and a basis for characterizing transcriptomic differences in multiple strains. Taken together, our results show that signal summation describes the composition of a model prokaryotic transcriptome

    T7 RNA Polymerase Functions In Vitro without Clustering

    Many nucleic acid polymerases function in clusters known as factories. We investigate whether the RNA polymerase (RNAP) of phage T7 also clusters when active. Using ‘pulldowns’ and fluorescence correlation spectroscopy we find that elongation complexes do not interact in vitro with a Kd<1 ”M. Chromosome conformation capture also reveals that genes located 100 kb apart on the E. coli chromosome do not associate more frequently when transcribed by T7 RNAP. We conclude that if clustering does occur in vivo, it must be driven by weak interactions, or mediated by a phage-encoded protein

    Kinetic Pathway of Pyrophosphorolysis by a Retrotransposon Reverse Transcriptase

    DNA and RNA polymerases use a common phosphoryl transfer mechanism for base addition that requires two or three acidic amino acid residues at their active sites. We previously showed, for the reverse transcriptase (RT) encoded by the yeast retrotransposon Ty1, that one of the three conserved active site aspartates (D211) can be substituted by asparagine and still retain in vitro polymerase activity, although in vivo transposition is lost. Transposition is partially restored by second site suppressor mutations in the RNAse H domain. The novel properties of this amino acid substitution led us to express the WT and D211N mutant enzymes, and study their pre-steady state kinetic parameters. We found that the kpol was reduced by a factor of 223 in the mutant, although the Kd for nucleotide binding was unaltered. Further, the mutant enzyme had a marked preference for Mn2+ over Mg2+. To better understand the functions of this residue within the Ty1 RT active site, we have now examined the in vitro properties of WT and D211N mutant Ty1 RTs in carrying out pyrophosphorolysis, the reverse reaction to polymerization, where pyrophosphate is the substrate and dNTPs are the product. We find that pyrophosphorolysis is efficient only when the base-paired primer template region is >14 bases, and that activity increases when the primer end is blunt-ended or recessed by only a few bases. Using pre-steady state kinetic analysis, we find that the rate of pyrophosphorolysis (kpyro) in the D211N mutant is nearly 320 fold lower than the WT enzyme, and that the mutant enzyme has an ∌170 fold lower apparent Kd for pyrophosphate. These findings indicate that subtle substrate differences can strongly affect the enzyme's ability to properly position the primer-end to carry out pyrophosphorolysis. Further the kinetic data suggests that the D211 residue has a role in pyrophosphate binding and release, which could affect polymerase translocation, and help explain the D211N mutant's transposition defect

    HIV Transmission Potential Among Local and Migrant Factory Workers in Kolkata, India

    Migrant workers in India play a key role in the spread of HIV. Kolkata is a common destination for workers, who may acquire infection and transmit it to their wives and/or other sexual partners. We investigated sexual relations and condom use by factory workers. Migrant and local factory workers were randomly selected from five wards of Kolkata. Information was collected about demographic and socio-economic characteristics, sexual relationships, condom usage, and perceptions and intent to use condoms. Condom use was very low in both groups of workers, particularly among migrants. Many married workers visited female sex workers but never used condoms. Few intended to use condoms, and if they did, it did not always translate into actual usage. There is great potential for transmission of HIV/sexually transmitted infections by these workers. Carefully designed intervention and education programs in the context of low literacy and cultural norms are urgently needed
