    Resting-state fMRI using passband balanced steady-state free precession

    OBJECTIVE: Resting-state functional MRI (rsfMRI) has been increasingly used for understanding brain functional architecture. To date, most rsfMRI studies have exploited blood oxygenation level-dependent (BOLD) contrast using gradient-echo (GE) echo planar imaging (EPI), which can suffer from image distortion and signal dropout due to magnetic susceptibility and inherent long echo time. In this study, the feasibility of passband balanced steady-state free precession (bSSFP) imaging for distortion-free and high-resolution rsfMRI was investigated. METHODS: rsfMRI was performed in humans at 3 T and in rats at 7 T using bSSFP with short repetition time (TR = 4/2.5 ms respectively) in comparison with conventional GE-EPI. Resting-state networks (RSNs) were detected using independent component analysis. RESULTS AND SIGNIFICANCE: RSNs derived from bSSFP images were shown to be spatially and spectrally comparable to those derived from GE-EPI images with considerable intra- and inter-subject reproducibility. High-resolution bSSFP images corresponded well to the anatomical images, with RSNs exquisitely co-localized to the gray matter. Furthermore, RSNs at areas of severe susceptibility such as human anterior prefrontal cortex and rat piriform cortex were proved accessible. These findings demonstrated for the first time that passband bSSFP approach can be a promising alternative to GE-EPI for rsfMRI.     Enhanced performance in polymer photovoltaic cells with chloroform treated indium tin oxide anode modification

    Enhanced performance of a poly(3-hexylthiophene):(6,6)-phenyl C61 butyric acid methyl ester bulk heterojunction polymer photovoltaic cell is reported by modifying the indium tin oxide (ITO) anode with chloroform solution. Instead of the traditional UV-ozone treatment, the optimized chloroform modification on ITO anode can result in an enhancement in the power conversion efficiency of an identical device, originating from an increase in the photocurrent with negligible change in the open-circuit voltage. The performance enhancement is attributed to the work function modification of the ITO substrate through the surface incorporation of the chlorine, and thus improved charge collection efficiency. © 2011 American Institute of Physics

    Surface plasmon-enhanced electroluminescence in organic light-emitting diodes incorporating Au nanoparticles

    Surface plasmon-enhanced electroluminescence (EL) in an organic light-emitting diode is demonstrated by incorporating the synthesized Au nanoparticles (NPs) in the hole injection layer of poly(3,4-ethylene dioxythiophene):polystyrene sulfonic acid. An increase of ∼25% in the EL intensity and efficiency are achieved for devices with Au NPs, whereas the spectral and electrical properties remain almost identical to the control device. Time-resolved photoluminescence spectroscopy reveals that the EL enhancement is ascribed to the increase in spontaneous emission rate due to the plasmonic near-field effect induced by Au NPs. © 2012 American Institute of Physics

    A Self-Consistent Model for Positronium Formation from Helium Atoms

    The differential and total cross sections for electron capture by positrons from helium atoms are calculated using a first-order distorted wave theory satisfying the Coulomb boundary conditions. In this formalism a parametric potential is used to describe the electron screening in a consistent and realistic manner. The present procedure is self consistent because (i) it satisfies the correct boundary conditions and post-prior symmetry, and (ii) the potential and the electron binding energies appearing in the transition amplitude are consistent with the wave functions describing the collision system. The results are compared with the other theories and with the available experimental measurements. At the considered range of collision energies, the results agree reasonably well with recent experiments and theories.     Coherent anti-Stokes Raman scattering imaging of lipids in cancer metastasis

    <p>Abstract</p> <p>Background</p> <p>Lipid-rich tumours have been associated with increased cancer metastasis and aggressive clinical behaviours. Nonetheless, pathologists cannot classify lipid-rich tumours as a clinically distinctive form of carcinoma due to a lack of mechanistic understanding on the roles of lipids in cancer development.</p> <p>Methods</p> <p>Coherent anti-Stokes Raman scattering (CARS) microscopy is employed to study cancer cell behaviours in excess lipid environments <it>in vivo </it>and <it>in vitro</it>. The impacts of a high fat diet on cancer development are evaluated in a Balb/c mice cancer model. Intravital flow cytometry and histology are employed to enumerate cancer cell escape to the bloodstream and metastasis to lung tissues, respectively. Cancer cell motility and tissue invasion capability are also evaluated in excess lipid environments.</p> <p>Results</p> <p>CARS imaging reveals intracellular lipid accumulation is induced by excess free fatty acids (FFAs). Excess FFAs incorporation onto cancer cell membrane induces membrane phase separation, reduces cell-cell contact, increases surface adhesion, and promotes tissue invasion. Increased plasma FFAs level and visceral adiposity are associated with early rise in circulating tumour cells and increased lung metastasis. Furthermore, CARS imaging reveals FFAs-induced lipid accumulation in primary, circulating, and metastasized cancer cells.</p> <p>Conclusion</p> <p>Lipid-rich tumours are linked to cancer metastasis through FFAs-induced physical perturbations on cancer cell membrane. Most importantly, the revelation of lipid-rich circulating tumour cells suggests possible development of CARS intravital flow cytometry for label-free detection of early-stage cancer metastasis.</p

    Dynamics of absorption properties of CDOM and its composition in Likas estuary, North Borneo, Malaysia

Available from Elsevier via the DOI in this record. Chromophoric Dissolved Organic Matter (CDOM) is a vital water constituent in aquatic ecosystems that contributes to water colour, affects light penetration, and impacts primary production. This study aims to determine the spatial and monsoonal variability of CDOM absorption properties in the Likas estuary, characterise the source of CDOM, and investigate the correlations between CDOM absorption properties and salinity. Likas estuary is a small estuary located in Kota Kinabalu city on the west coast of Sabah, facing the South China Sea. A mangrove ecosystem surrounds it with manufactured structures such as residential areas and public facilities. Surface water samples were collected at 19 stations: upstream of rivers to the river mouth and coastal area during spring tides every month, from June 2018 to July 2019, for 14-months. The distribution of aCDOM(440) in the study area is predictable as a signature in a coastal area with a decreasing gradient from the upstream towards coastal water (0.29 ± 0.19 m−1 to 1.05 ± 0.39 m−1). There are increasing spatial patterns of spectral slopes S275-295 and SR. However, S350-400 and S300-600 declined spatial gradients from the upstream to coastal water. Thus, S300-600 indicates a linear relationship between aCDOM(440), which unconventional results in coastal water. We suspect this is due to a small coverage of the study site with a distance of 0.5 m intervals of each station. This could be why the S300-600 had constant values throughout the study area (with no statistical difference between stations). In addition, S300-600 was merely varied in the stations located at the river mouth and coastal water. Based on the spectral slope ratio (SR), most of the stations located in the Darau, Inanam, and Bangka-Bangka rivers had SR values less than 1. Hence, CDOM in these stations is a terrestrial-dominated source.     Flavor Violating Higgs Decays

    We study a class of nonstandard interactions of the newly discovered 125 GeV Higgs-like resonance that are especially interesting probes of new physics: flavor violating Higgs couplings to leptons and quarks. These interaction can arise in many frameworks of new physics at the electroweak scale such as two Higgs doublet models, extra dimensions, or models of compositeness. We rederive constraints on flavor violating Higgs couplings using data on rare decays, electric and magnetic dipole moments, and meson oscillations. We confirm that flavor violating Higgs boson decays to leptons can be sizeable with, e.g., h -> tau mu and h -> tau e branching ratios of order 10% perfectly allowed by low energy constraints. We estimate the current LHC limits on h -> tau mu and h -> tau e decays by recasting existing searches for the SM Higgs in the tau-tau channel and find that these bounds are already stronger than those from rare tau decays. We also show that these limits can be improved significantly with dedicated searches and we outline a possible search strategy.     Seminiferous tubule transfection in vitro to define post-meiotic gene regulation

    The electronic version of this article is the complete one and can be found online at: http://www.rbej.com/content/7/1/67Background: Post-meiotically expressed genes in the testis are essential for the proper progression of spermatogenesis, and yet, aside from the construction of individual transgenic mice using specific promoters to drive reporter plasmids, there are only very limited possibilities for relevant and quantitative analysis of gene promoters. This is due to the special nature of post-meiotic haploid cells, which to date are not represented in any appropriate cell-lines. This article reports the development of novel methodology using isolated and cultured rat seminiferous tubules in a multiwell format, into which promoter-reporter constructs can be introduced by a combination of microinjection and electroporation. Methods: Culture conditions were developed which allowed the continued incubation of isolated rat seminiferous tubules for up to 48 h without obvious cell death and loss of post-meiotic cells. Transfection of intact seminiferous tubules by microinjection and electroporation was optimized to achieve high expression efficiencies of control plasmids, using either fluorescent protein or luciferase as reporters, thereby allowing both morphological as well as quantitative assessment. Results: Successful transfection was achieved into all cell types except for mature spermatozoa. However, there appeared to be only limited cell-type specificity for the promoters used, even though these had appeared to be specific when used in transgenic animals. Conclusion: We have devised a methodology which allows relatively high throughput analysis of post-meiotic gene promoters into primary cells of intact seminiferous tubules. An apparent lack of cell-type specificity suggests that the gene fragments used do not contain sufficient targeting information, or that the transient episomal expression of the constructs does not encourage appropriate expression specificity.
