    Antioxidant, Anti-inflammatory and Cytotoxicity of Phaleria macrocarpa (Boerl.) Scheff Fruit

    <p>Abstract</p> <p>Background</p> <p><it>Phaleria macrocarpa </it>(Scheff.) Boerl (Thymelaceae) originates from Papua Island, Indonesia and grows in tropical areas. The different parts of the fruit of <it>P. macrocarpa </it>were evaluated for antioxidant, anti-inflammatory, and cytotoxic activities.</p> <p>Methods</p> <p><it>Phaleria macrocarpa </it>fruit were divided into pericarp, mesocarp and seed. All parts of the fruit were reflux extracted with methanol. The antioxidant activity of the extracts were characterized in various <it>in vitro </it>model systems such as FTC, TBA, DPPH radical, reducing power and NO radical. Anti-inflammatory assays were done by using NO production by macrophage RAW 264.7 cell lines induced by LPS/IFN-γ and cytotoxic activities were determined by using several cancer cell lines and one normal cell line</p> <p>Results</p> <p>The results showed that different parts (pericarp, mesocarp, and seed) of <it>Phaleria macrocarpa </it>fruit contain various amount of total phenolic (59.2 ± 0.04, 60.5 ± 0.17, 47.7 ± 1.04 mg gallic acid equivalent/g DW) and flavonoid compounds (161.3 ± 1.58, 131.7 ± 1.66, 35.9 ± 2.47 mg rutin equivalent/g DW). Pericarp and mesocarp showed high antioxidant activities by using DPPH (71.97%, 62.41%), ferric reducing antioxidant power (92.35%, 78.78%) and NO scavenging activity (65.68%, 53.45%). Ferric thiocyanate and thiobarbituric acid tests showed appreciable antioxidant activity in the percentage hydroperoxides inhibitory activity from pericarp and mesocarp in the last day of the assay. Similarly, the pericarp and mesocarp inhibited inducible nitric oxide synthesis with values of 63.4 ± 1.4% and 69.5 ± 1.4% in macrophage RAW 264.7 cell lines induced by LPS/IFN-γ indicating their notable anti-inflammatory potential. Cytotoxic activities against HT-29, MCF-7, HeLa and Chang cell lines were observed in all parts.</p> <p>Conclusions</p> <p>These results indicated the possible application of <it>P. macrocarpa </it>fruit as a source of bioactive compounds, potent as an antioxidant, anti inflammatory and cytotoxic agents.</p

    Antioxidant activity and in vitro inhibition of tumor cell growth by leaf extracts from the carob tree (Ceratonia siliqua)

    The methanol leaf extracts of female cultivars of the carob tree [Ceratonia siliqua L. (Fabaceae)] and of hermaphrodite and male trees were investigated for their contents of phenolic compounds, their in vitro antioxidant activity, measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging and linoleic acid system assays, and their in vitro tumor growth inhibition on HeLa cells. The different cultivars and trees showed high levels of phenols, and considerable variations in the amount of these compounds. The extracts showed significant radical scavenging activity (RSA), which was not significantly affected by the gender of the tree. From the female cultivars tested, Galhosa exhibited the highest RSA. Gender significantly affected the antioxidant activity of the extracts measured by the linoleic acid system assay, and males and hermaphrodites showed the highest activities. The extracts displayed a remarkable ability to inhibit tumor cell proliferation, and their bioactivity varied with different cultivars or trees tested. Extracts from male and hermaphrodite trees exhibited higher capacity to inhibit the proliferation of HeLa cells than the female cultivars

    Genetic variation in multiple biologic pathways, flavonoid intake, and breast cancer

    PURPOSE: We previously reported an inverse association between flavonoid intake and breast cancer incidence, which has been confirmed by others; but no studies have considered simultaneously potential interactions of flavonoids with multiple genetic polymorphisms involved in biologically-relevant pathways (oxidative stress, carcinogen metabolism, DNA repair, and one-carbon metabolism). METHODS: To estimate interaction effects between flavonoids and 13 polymorphisms in these four pathways on breast cancer risk, we used population-based data (N = 875 cases and 903 controls) and several statistical approaches, including conventional logistic regression and semi-Bayesian hierarchical modeling (incorporating prior information on the possible biological functions of genes), which also provides biologic pathway-specific effect estimates. RESULTS: Compared to the standard multivariate model, the results from the hierarchical model indicate that gene-by-flavonoid interaction estimates are attenuated, but more precise. In the hierarchical model, the average effect of the deleterious versus beneficial gene, controlling for average flavonoid intake in the DNA repair pathway, and adjusted for the three other biologically-relevant pathways (oxidative stress, carcinogen metabolism, and one-carbon metabolism), resulted in a 27% increase risk for breast cancer [Odds Ratio (OR) = 1.27; 95% Confidence Interval (CI) = 0.70, 2.29]. However, the CI was wide. CONCLUSIONS: Based on results from the semi-Bayesian model, breast cancer risk may be influenced jointly by flavonoid intake and genes involved in DNA repair, but our findings require confirmation

    The effectiveness of mathematics teaching: a cross-national investigation in primary schools in England and China

    This study focuses on the effectiveness of mathematics teaching to children aged 9-10 years, applies a mixture of six methods to classroom-level data collected in England and China, correlates observable teacher behaviours with pupil mathematics performance and collects multiple perceptions that indirectly connect with the differences of teaching and learning cross-nationally. It has been found in the study that 9-to 10-year-olds (n = 343) from China outscored their English peers (n = 236) at the same age by over 20 per cent in each of two mathematics tests derived from TIMSS 2003. Structured analysis of lesson videos has revealed that Chinese mathematics teachers scored much higher than their English colleagues on an internationally validated observation instrument which focused on the quality of six dimensions of teacher behaviours. Furthermore, the quantity of teacher behaviours was also measured and the subsequent correlational analysis on pooled data indicated a positive effect of whole-class interactive teaching (r = 0.97, p &lt; 0.01) and pupil time on task (r = 0.95, p &lt; 0.01) and a negative impact of whole-class lecture (r = -0.91, p &lt; 0.01), individual/group work (r=-0.81, p &lt; 0.05) and classroom management (r = -0.77, p &lt; 0.05) on pupils’ mathematics performance cross-nationally. Qualitative findings are connected with quantitative results to explain how teachers think, how this relates to the way they teach and how the differences of teaching result in the performance gap cross-nationally. The study replicated previous TER findings from the West across two geographically and culturally different countries, suggested possible directions for future enquiries, and recommended potential ways for practice and policy innovations
