    Visceral adiposity and cancer: Role in pathogenesis and prognosis

    The prevalence of being overweight and obese has been expanded dramatically in recent years worldwide. Obesity usually occurs when the energetic introit overtakes energy expenditure from metabolic and physical activity, leading to fat accumulation mainly in the visceral depots. Excessive fat accumulation represents a risk factor for many chronic diseases, including cancer. Adiposity, chronic low-grade inflammation, and hyperinsulinemia are essential factors of obesity that also play a crucial role in tumor onset. In recent years, several strategies have been pointed toward boundary fat accumulation, thus limiting the burden of cancer attributable to obesity. While remodeling fat via adipocytes browning seems a tempting prospect, lifestyle interventions still represent the main pathway to prevent cancer and enhance the efficacy of treatments. Specifically, the Mediterranean Diet stands out as one of the best dietary approaches to curtail visceral adiposity and, therefore, cancer risk. In this Review, the close relationship between obesity and cancer has been investigated, highlighting the biological mechanisms at the basis of this link. Finally, strategies to remodel fat, including browning and lifestyle interventions, have been taken into consideration as a major perspective to limit excess body weight and tumor onset

    Influence of the rainfall in the content of nutrients in litter in agroforestry systems managed with burning and without burning in Amazon.

    This study evaluated the nutrient content of the litter, testing different treatments with burning and no burning, of the vegetation, to identify which one provides better efficiency in operation and production of nutrients in different seasonal conditions. The study area is located on the property of the family farmer, initially selected by a diagnosis socioeconomic, community Benjamin Constant, in northeastern Para. Litter was collected during two periods: dry season (November) and rainy (March) in 2009. For the collection of litter samples, we used collectors measuring (0.25 × 0.25 m2), which were placed directly on the soil surface. The collected material was stored in paper bags and taken to the laboratory for Chemical Analysis of the Brazilian Agricultural Research Corporation (EMBRAPA), which was determined by analyses of macronutrients (N, P, K, Ca, Mg, Na) and micronutrients (Fe, Cu, Mn, Zn). The highest concentrations of macronutrients were found in N for agroforestry systems with and without burning in two seasons (wet and dry). All macronutrients showed influence of seasonality, which was verified by the wide variation in nutritional behavior. The decreasing concentration of nutrients was presented N > Ca > Mg > Na > K > P in agroforestry system with burning, with maximum values of all nutrients in the rainy season, and N, P, K, Ca, Na in higher concentrations in agroforestry system without burning, and showed only the Mg peak in agroforestry system with burning. The behavior of the concentration of nutrients was opposite to that observed one, for all elements analyzed showed a reduction in the concentrations of nutrients in the dry season.     Calibração do medidor de clorofila Minolta SPAD-502 para avaliação do conteúdo de nitrogênio do milho.

    O objetivo deste trabalho foi calibrar o medidor de clorofila Minolta SPAD-502 para avaliação da nutrição nitrogenada das plantas de milho, baseando-se na prévia comparação das leituras de clorofila com os teores obtidos pelos extratores N,N-dimetilformamida (DMF) e acetona 80%. No campo, foram avaliados os teores de clorofila em folhas de milho cultivado sob plantio direto após a cultura de aveia-preta e após a de tremoço-branco, por duas safras consecutivas. A extração da clorofila com a solução DMF, por 72 horas, foi a mais indicada para a calibração do medidor de clorofila SPAD-502 na cultura do milho. O ajuste do modelo linear expressou melhor a relação entre o conteúdo de clorofila e as leituras do SPAD na cultura do milho, e seu uso produziu resultados coerentes com o estado nutricional da cultura

    PGC-1α induced browning promotes involution and inhibits lactation in mammary glands

    The PPARγ coactivator 1α (PGC-1α) is a transcriptional regulator of mitochondrial biogenesis and oxidative metabolism. Recent studies have highlighted a fundamental role of PGC-1α in promoting breast cancer progression and metastasis, but the physiological role of this coactivator in the development of mammary glands is still unknown. First, we show that PGC-1α is highly expressed during puberty and involution, but nearly disappeared in pregnancy and lactation. Then, taking advantage of a newly generated transgenic mouse model with a stable and specific overexpression of PGC-1α in mammary glands, we demonstrate that the re-expression of this coactivator during the lactation stage leads to a precocious regression of the mammary glands. Thus, we propose that PGC-1α action is non-essential during pregnancy and lactation, whereas it is indispensable during involution. The rapid preadipocyte–adipocyte transition, together with an increased rate of apoptosis promotes a premature mammary glands involution that cause lactation defects and pup growth retardation. Overall, we provide new insights in the comprehension of female reproductive cycles and lactation deficiency, thus opening new roads for mothers that cannot breastfeed

    Adhesion of platelets to colon cancer cells is necessary to promote tumor development in xenograft, genetic and inflammation models

    Platelets represent the linkage between tissue damage and inflammatory response with a putative role in tumorigenesis. Given the importance of the microenvironment in colon cancer development, we elucidated the eventual role of platelets‐cancer cells crosstalk in in vivo colon cancer models. To evaluate the involvement of platelets in intestinal tumorigenesis, we first analyzed if the ablation of β‐integrin P‐selectin that drives platelets‐cell adhesion, would contribute to platelets‐colon cancer cell interaction and drive cancer progression. In a xenograft tumor model, we observed that when tumors are inoculated with platelets, the ablation of P‐selectin significantly reduced tumor growth compared to control platelets. Furthermore, in genetic models, as well as in chronic colitis‐associated colorectal carcinogenesis, P‐selectin ablated mice displayed a significant reduction in tumor number and size compared to control mice. Taken together, our data highlights the importance of platelets in the tumor microenvironment for intestinal tumorigenesis. These results support the hypothesis that a strategy aimed to inhibit platelets adhesion to tumor cells are able to block tumor growth and could represent a novel therapeutic approach to colon cancer treatment

    Psychopathology in Young People With Intellectual Disability

    Context Comorbid severe mental health problems complicating intellectual disability are a common and costly public health problem. Although these problems are known to begin in early childhood, little is known of how they evolve over time or whether they continue into adulthood. Objective To study the course of psychopathology in a representative population of children and adolescents with intellectual disability. Design, Setting, and Participants The participants of the Australian Child to Adult Development Study, an epidemiological cohort of 578 children and adolescents recruited in 1991 from health, education, and family agencies that provided services to children with intellectual disability aged 5 to 19.5 years in 6 rural and urban census regions in Australia, were followed up for 14 years with 4 time waves of data collection. Data were obtained from 507 participants, with 84% of wave 1 (1991-1992) participants being followed up at wave 4 (2002-2003). Main Outcome Measures The Developmental Behaviour Checklist (DBC), a validated measure of psychopathology in young people with intellectual disability, completed by parents or other caregivers. Changes over time in the Total Behaviour Problem Score and 5 subscale scores of the DBC scores were modeled using growth curve analysis. Results High initial levels of behavioral and emotional disturbance decreased only slowly over time, remaining high into young adulthood, declining by 1.05 per year on the DBC Total Behaviour Problem Score. Overall severity of psychopathology was similar across mild to severe ranges of intellectual disability (with mean Total Behavior Problem Scores of approximately 44). Psychopathology decreased more in boys than girls over time (boys starting with scores 2.61 points higher at baseline and ending with scores 2.57 points lower at wave 4), and more so in participants with mild intellectual disability compared with those with severe or profound intellectual disability who diverged from having scores 0.53 points lower at study commencement increasing to a difference of 6.98 points below severely affected children by wave 4. This trend was observed in each of the subscales, except the social-relating disturbance subscale, which increased over time. Prevalence of participants meeting criteria for major psychopathology or definite psychiatric disorder decreased from 41% at wave 1 to 31% at wave 4. Few of the participants (10%) with psychopathology received mental health interventions during the study period. Conclusion These results provide evidence that the problem of psychopathology comorbid with intellectual disability is both substantial and persistent and suggest the need for effective mental health interventions
