    Absence of seasonal patterns in MBT-CBT indices in mid-latitude soils

    The degree of methylation and cyclization of bacteria-derived branched glycerol dialkyl glycerol tetraether (GDGT) membrane lipids in soils depends on temperature and soil pH. Expressed in the methylation index of branched tetraethers (MBT) and cyclization ratio of branched tetraethers (CBT), these relationships are used to reconstruct past annual mean air temperature (MAT) based on the distribution of branched GDGTs in ancient sediments; the MBT-CBT proxy. Although it was shown that the best correlation of this proxy is with annual MAT, it remains unknown whether a seasonal bias in temperature reconstructions could occur, such as towards a seasonal period of optimal growth’ of the, as yet, unidentified soil bacteria which produce branched GDGTs. To investigate this possibility, soils were sampled from eight different plots in the USA (Minnesota and Ohio), The Netherlands (Texel) and the UK (Devon) in time series over 1 year and analyzed for their branched GDGT content. Further analyses of the branched GDGTs present as core lipids (CLs; the presumed fossil pool) and intact polar lipids (IPLs; the presumed extant pool) were undertaken for two of the investigated soil plots. The amount of IPL-derived branched GDGTs is low relative to the branched GDGT CLs, i.e. only 6–9% of the total branched GDGT pool.In all soils, no clear change was apparent in the distribution of branched GDGT lipids (either core or IPL-derived) with seasonal temperature change; the MBT–CBT temperature proxy gave similar temperature estimates year-round, which generally matched the mean annual soil temperature. In addition to a lack of coherent changes in relative distributions, concentrations of the branched GDGTs did not show clear changes over the seasons. For IPL-derived GDGTs these results suggest that their turnover time in soils is in the order of 1 year or more. Thus, our study does not provide evidence for seasonal effects on the distribution of branched GDGTs in soils, at least at mid-latitudes, and therefore, no direct evidence for a bias of MBT–CBT reconstructed temperatures towards a certain season of optimal growth of the source bacteria. If, however, there is a slight seasonal preference of branched GDGT production, which can easily be obscured by natural variability due to the heterogeneity of soils, then a seasonal bias may potentially still develop over time due to the long turnover time of branched GDGTs

    Photodynamic therapy as adjuvant therapy in surgically treated pleural malignancies.

    Five patients with a pleural malignancy (four malignant mesotheliomas and one localized low grade carcinoid) were treated with maximal surgical resection of the tumour followed by intraoperative adjuvant photodynamic therapy (PDT). The additional photodynamic treatment was performed with light of 652 nm from a high power diode laser, and meta-tetrahydroxy phenylchlorin as the photosensitizer. The light delivery to the thoracic cavity was monitored by in situ isotropic light detectors. The position of the light delivery fibre was adjusted to achieve optimal light distribution, taking account of reflected and scattered light in this hollow cavity. There was no 30-day post-operative mortality and only one patient suffered from a major complication (diaphragmatic rupture and haematopericardium). The operation time was increased by a maximum of 1 h to illuminate the total hemithoracic surface with 10 J cm(-2) (incident and scattered light). The effect of the adjuvant PDT was monitored by examination of biopsies taken 24 h after surgery under thoracoscopic guidance. Significant damage, including necrosis, was observed in the marker lesions with remaining malignancy compared with normal tissue samples, which showed only an infiltration with PMN cells and oedema of the striated muscles cells. Of the five patients treated, four are alive with no signs of recurrent tumour with a follow-up of 9-11 months. One patient was diagnosed as having a tumour dissemination in the skin around the thoracoscopy scar and died of abdominal tumour spread. Light delivery to large surfaces for adjuvant PDT is feasible in a relatively short period of time (< 1 h). In situ dosimetry ensures optimal light distribution and allows total doses (incident plus scattered light) to be monitored at different positions within the cavity. This combination of light delivery and dosimetry is well suited for adjuvant treatment with PDT in malignant pleural tumours

    Paleoceanography and ice sheet variability offshore Wilkes Land, Antarctica – Part 3: Insights from Oligocene–Miocene TEX86-based sea surface temperature reconstructions

    The volume of the Antarctic continental ice sheet(s) varied substantially during the Oligocene and Miocene ( 34–5 Ma) from smaller to substantially larger than today, both on million-year and on orbital timescales. However, reproduction through physical modeling of a dynamic response of the ice sheets to climate forcing remains problematic, suggesting the existence of complex feedback mechanisms between the cryosphere, ocean, and atmosphere systems. There is therefore an urgent need to improve the models for better predictions of these systems, including resulting potential future sea level change. To assess the interactions between the cryosphere, ocean, and atmosphere, knowledge of ancient sea surface conditions close to the Antarctic margin is essential. Here, we present a new TEX86- based sea surface water paleotemperature record measured on Oligocene sediments from Integrated Ocean Drilling Program (IODP) Site U1356, offshore Wilkes Land, East Antarctica. The new data are presented along with previously published Miocene temperatures from the same site. Together the data cover the interval between 34 and 11 Ma and encompasses two hiatuses. This record allows us to accurately reconstruct the magnitude of sea surface temperature (SST) variability and trends on both million-year and glacial–interglacial timescales.Julian D.     Assessing branched tetraether lipids as tracers of soil organic carbon transport through the Carminowe Creek catchment (southwest England)

    Soils represent the largest reservoir of organic carbon (OC) on land. Upon mobilization, this OC is either returned to the atmosphere as carbon dioxide (CO2) or transported and ultimately locked into (marine) sediments, where it will act as a long-term sink of atmospheric CO2. These fluxes of soil OC are, however, difficult to evaluate, mostly due to the lack of a soil-specific tracer. In this study, a suite of branched glycerol dialkyl glycerol tetraethers (brGDGTs), which are membrane lipids of soil bacteria, is tested as specific tracers for soil OC from source (soils under arable land, ley, grassland, and woodland) to sink (Loe Pool sediments) in a small catchment located in southwest England (i.e. Carminowe Creek draining into Loe Pool). The analysis of brGDGTs in catchment soils reveals that their distribution is not significantly different across different land use types (p > 0:05) and thus does not allow land-use-specific soil contributions to Loe Pool sediments to be traced. Furthermore, the significantly higher contribution of 6-methyl brGDGT isomers in creek sediments (isomerization ratio (IR) D 0:48 ± 0:10, mean ± standard deviation (SD); p < 0:05) compared to that in catchment soils (IR D 0:28±0:11) indicates that the initial soil signal is substantially altered by brGDGT produced in situ. Similarly, the riverine brGDGT signal appears to be overwritten by lacustrine brGDGTs in the lake sedimentary record, indicated by remarkably lower methylation of branched tetraethers (MBT05ME D 0:46 ± 0:02 in creek bed sediments and 0:38 ± 0:01 in lake core sediments; p < 0:05) and a higher degree of cyclization (DC D 0:23±0:02 in creek bed sediments and 0:32±0:08 in lake core sediments). Thus, in this small catchment, brGDGTs do not allow us to trace soil OC transport. Nevertheless, the downcore changes in the degree of cyclization and the abundance of isoprenoid GDGTs produced by methanogens in the Loe Pool sediment do reflect local environmental conditions over the past 100 years and have recorded the eutrophication history of the lake. © 2020 Author(s)

    Lipid-biomarker-based sea surface temperature record offshore Tasmania over the last 23 million years

    The Neogene (23.04–2.58 Ma) is characterised by progressive buildup of ice volume and climate cooling in the Antarctic and the Northern Hemisphere. Heat and moisture delivery to Antarctica is, to a large extent, regulated by the strength of meridional temperature gradients. However, the evolution of the Southern Ocean frontal systems remains scarcely studied in the Neogene. Here, we present the first long-term continuous sea surface temperature (SST) record of the subtropical front area in the Southern Ocean at Ocean Drilling Program (ODP) Site 1168 off western Tasmania. This site is, at present, located near the subtropical front (STF), as it was during the Neogene, despite a 10∘ northward tectonic drift of Tasmania. We analysed glycerol dialkyl glycerol tetraethers (GDGTs – on 433 samples) and alkenones (on 163 samples) and reconstructed the paleotemperature evolution using TEX86 and U37k′ as two independent quantitative proxies. Both proxies indicate that Site 1168 experienced a temperate ∼ 25 ∘C during the early Miocene (23–17 Ma), reaching ∼ 29 ∘C during the mid-Miocene climatic optimum. The stepwise ∼ 10 ∘C cooling (20–10 ∘C) in the mid-to-late Miocene (12.5–5.0 Ma) is larger than that observed in records from lower and higher latitudes. From the Pliocene to modern (5.3–0 Ma), STF SST first plateaus at ∼ 15 ∘C (3 Ma), then decreases to ∼ 6 ∘C (1.3 Ma), and eventually increases to the modern levels around ∼ 16 ∘C (0 Ma), with a higher variability of 5∘ compared to the Miocene. Our results imply that the latitudinal temperature gradient between the Pacific Equator and the STF during late Miocene cooling increased from 4 to 14 ∘C. Meanwhile, the SST gradient between the STF and the Antarctic margin decreased due to amplified STF cooling compared to the Antarctic margin. This implies a narrowing SST gradient in the Neogene, with contraction of warm SSTs and northward expansion of subpolar conditions.</p

    E-cadherin inactivation in lobular carcinoma in situ of the breast: an early event in tumorigenesis.

    In breast cancer, inactivating point mutations in the E-cadherin gene are frequently found in invasive lobular carcinoma (ILC) but never in invasive ductal carcinoma (IDC). Lobular carcinoma in situ (LCIS) adjacent to ILC has previously been shown to lack E-cadherin expression, but whether LCIS without adjacent invasive carcinoma also lacks E-cadherin expression and whether the gene mutations present in ILC are already present in LCIS is not known. We report here that E-cadherin expression is absent in six cases of LCIS and present in 150 cases of ductal carcinoma in situ (DCIS), both without an adjacent invasive component. Furthermore, using mutation analysis, we could demonstrate the presence of the same truncating mutations and loss of heterozygosity (LOH) of the wild-type E-cadherin in the LCIS component and in the adjacent ILC. Our results indicate that E-cadherin is a very early target gene in lobular breast carcinogenesis and plays a tumour-suppressive role, additional to the previously suggested invasion-suppressive role

    Organic matter characteristics in yedoma and thermokarst deposits on Baldwin Peninsula, west Alaska

    As Arctic warming continues and permafrost thaws, more soil and sedimentary organic matter (OM) will be decomposed in northern high latitudes. Still, uncertainties remain in the quality of the OM and the size of the organic carbon (OC) pools stored in different deposit types of permafrost landscapes. This study presents OM data from deep permafrost and lake deposits on the Baldwin Peninsula which is located in the southern portion of the continuous permafrost zone in west Alaska. Sediment samples from yedoma and drained thermokarst lake basin (DTLB) deposits as well as thermokarst lake sediments were analyzed for cryostratigraphical and biogeochemical parameters and their lipid biomarker composition to identify the belowground OC pool size and OM quality of ice-rich permafrost on the Baldwin Peninsula. We provide the first detailed characterization of yedoma deposits on Baldwin Peninsula. We show that three-quarters of soil OC in the frozen deposits of the study region (total of 68&thinsp;Mt) is stored in DTLB deposits (52&thinsp;Mt) and one-quarter in the frozen yedoma deposits (16&thinsp;Mt). The lake sediments contain a relatively small OC pool (4&thinsp;Mt), but have the highest volumetric OC content (93&thinsp;kg&thinsp;m−3) compared to the DTLB (35&thinsp;kg&thinsp;m−3) and yedoma deposits (8&thinsp;kg&thinsp;m−3), largely due to differences in the ground ice content. The biomarker analysis indicates that the OM in both yedoma and DTLB deposits is mainly of terrestrial origin. Nevertheless, the relatively high carbon preference index of plant leaf waxes in combination with a lack of a degradation trend with depth in the yedoma deposits indicates that OM stored in yedoma is less degraded than that stored in DTLB deposits. This suggests that OM in yedoma has a higher potential for decomposition upon thaw, despite the relatively small size of this pool. These findings show that the use of lipid biomarker analysis is valuable in the assessment of the potential future greenhouse gas emissions from thawing permafrost, especially because this area, close to the discontinuous permafrost boundary, is projected to thaw substantially within the 21st century.</p
