    Recent blooms of the dinoflagellate Ceratium in Albert Falls Dam (KZN): History, causes, spatial features and impacts on a reservoir ecosystem and its zooplankton

    A lake-wide bloom of the dinoflagellate Ceratium hirundinella, discovered in Albert Falls Dam in October 2006, exposed a significant ecological change indicative of reduced water quality in this historically mesotrophic reservoir. The spatial distribution of the bloom was examined synoptically in October 2006 and January 2007; these surveys revealed generally higher dinoflagellate densities in inshore reaches of the lake, and especially in the discharge plume of the inflowing Mgeni River. Ceratium totally dominated the phytoplankton assemblage, accounting almost completely for coincident chlorophyll levels, which generally increased with depth to generate a ‘deep’ chlorophyll maximum. Vertical oxygen profiles during the bloom differed substantially from corresponding profiles during non-bloom conditions historically typical in this reservoir. Direct count data and ordination analysis using non-metric multidimensional scaling exposed marked changes in zooplankton community structure compared to seasonally congruent non-bloom conditions in other years. Changes included the effective replacement of Moina by Bosmina, substantial reductions in Daphnia and Ceriodaphnia, and smaller but definite increases in abundance especially of calanoid copepods, as well as cyclopoid copepods and of Chaoborus, although not all of these differences were apparent in both survey months. These compositional changes are attributable to intrinsic differences in feeding biology among taxa and their associated susceptibility to the altered food environment, which was commensurate with Ceratium’s emergence. In addition, chydorid cladocerans appeared as a new (but spatially restricted) eutrophic bio-indicator component of the zooplankton, and the species diversity of cyclopoid copepods was enriched.The historical incidence of Ceratium in the lake since 1995 coincided with low NO3-N:TP values (used here as an N:P ratio proxy), particularly of inflow waters, and with broadly coincident values in the open lake. Ceratium was present but sparse in 1995, at average N:P ratios around 5.5. It disappeared in 1996 when the ratio increased radically to >10, and reappeared in 2004 after an erratic decline of the ratio to < 5 in the lake. The decline in N:P ratio of inflow waters since 1996 was clearly associated with a consistent rise in TP levels in inflows, most plausibly attributable to inputs of (Howick) wastewater treatment (WWT) plant origin. The appearance of Ceratium blooms is accordingly related to progressive elevations in mean annual P concentrations in inflows from ~40 ìg/l in 1995 to 120 ìg/l in 2007 (broadly mirrored in annual TP loadings), suggesting that improved operational efficiency (and capacity) of the WWT plant offers a plausible prospect for mitigation and reversa

    Knowledge and attitudes of parents and professionals to neonatal BCG vaccination in light of recent UK policy changes: A questionnaire study

    <p>Abstract</p> <p>Background</p> <p>Universal BCG vaccination in the UK ended in 2005. The new vaccination policy instead offers a number of different forms of selective vaccination to newborns based on risk of acquiring TB. We set out to assess the attitudes and knowledge of both parents and professionals to the new policy for neonatal BCG vaccination.</p> <p>Methods</p> <p>A short questionnaire was designed, made up of demographic and attitude questions, as well as very basic knowledge questions. The researchers handed out the questionnaire to all parents and professionals in the antenatal and postnatal areas, as well as the paediatric and neonatal units during a 6-week period. The site was the Royal Oldham hospital, a district general hospital with 3250 deliveries per year and multi-ethnic in its population mix.</p> <p>Results</p> <p>A total of 253 completed questionnaires were collected. The ethnic origin of responders was 50.6% White British, 18.2% Bangladeshi, 8.7% Indian, 4% White/Asian, the remaining 18.5% of other origins. 71.5% of responders said they had heard of BCG vaccine. When asked if they knew the new policy for its use, 33.2% answered yes. 24.5% gave the most accurate response when asked who now receives BCG.</p> <p>Conclusion</p> <p>We have found that amongst parents and professionals alike there is a lack of knowledge of the new policy. This has lead to confusion and as knowledge amongst the professionals who identify neonates for vaccination is low, uptake may be sub-optimal. We suggest that units investigate the issue and ensure that the new policy is understood and implemented correctly.</p

    Detection of metallic cobalt and chromium liver deposition following failed hip replacement using T2* and R2 magnetic resonance

    BACKGROUND: Failed hip prostheses can cause elevated circulating cobalt and chromium levels, with rare reports of fatal systemic organ deposition, including cobalt cardiomyopathy. Although blood cobalt and chromium levels are easily measured, organ deposition is difficult to detect without invasive biopsy. The T2* magnetic resonance (MR) method is used to quantify tissue iron deposition, and plays an important role in the management of iron-loading conditions. Cobalt and chromium, like iron, also affect magnetism and are proposed MR contrast agents. CASE PRESENTATION: We describe a case of a 44-year-old male with a failed hip implant and very elevated blood cobalt and chromium levels. Despite normal cardiac MR findings, liver T2* and R2 values were abnormal, triggering tissue biopsy. Liver tissue analysis, including X-ray fluorescence, demonstrated heavy elemental cobalt and chromium deposition in macrophages, and no detectable iron. CONCLUSIONS: Our case demonstrates T2* and R2 quantification of liver metal deposition in a patient with a failed hip implant. Further work is needed to investigate the role of T2* and R2 MR in the detection of metal deposition from metal on metal hip prostheses

    Cholesterol and the risk of grade-specific prostate cancer incidence: evidence from two large prospective cohort studies with up to 37 years' follow up

    &lt;b&gt;Background&lt;/b&gt; High cholesterol may be a modifiable risk factor for prostate cancer but results have been inconsistent and subject to potential "reverse causality" where undetected disease modifies cholesterol prior to diagnosis.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Methods&lt;/b&gt; We conducted a prospective cohort study of 12,926 men who were enrolled in the Midspan studies between 1970 and 1976 and followed up to 31st December 2007. We used Cox-Proportional Hazards Models to evaluate the association between baseline plasma cholesterol and Gleason grade-specific prostate cancer incidence. We excluded cancers detected within at least 5 years of cholesterol assay.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Results&lt;/b&gt; 650 men developed prostate cancer in up to 37 years' follow-up. Baseline plasma cholesterol was positively associated with hazard of high grade (Gleason score[greater than or equal to]8) prostate cancer incidence (n=119). The association was greatest among men in the 4th highest quintile for cholesterol, 6.1 to &#60;6.69 mmol/l, Hazard Ratio 2.28, 95% CI 1.27 to 4.10, compared with the baseline of &#60;5.05 mmol/l. This association remained significant after adjustment for body mass index, smoking and socioeconomic status.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Conclusions&lt;/b&gt; Men with higher cholesterol are at greater risk of developing high-grade prostate cancer but not overall risk of prostate cancer. Interventions to minimise metabolic risk factors may have a role in reducing incidence of aggressive prostate cancer

    Leadership and decision-making practices in public versus private universities in Pakistan

    The goal of this study is to examine differences in leadership and decision-making practices in public and private universities in Pakistan, with a focus on transformational leadership (TL) and participative decision-making (PDM). We conducted semi-structured interviews with 46 deans and heads of department from two public and two private universities in Pakistan. Our findings indicate that leadership and decision-making practices are different in public and private universities. While differences were observed in all six types of TL-behaviour, the following three approaches emerged to be crucial in both public and private universities: (1) articulating a vision, (2) fostering the acceptance of group goals, and (3) high-performance expectations. In terms of PDM, deans and heads of department in public and private universities adopt a collaborative approach. However, on a practical level this approach is limited to teacher- and student-related matters. Overall, our findings suggest that the leadership and decision-making practices in Pakistani public and private universities are transformational and participative in nature

    Lipids and nutrition of the southern rock lobster, Jasus edwardsii, from hatch to puerulus

    We examined the lipid class and fatty-acid composition of the southern rock lobster, Jasus edwardsii, phyllosomas larvae and puerulus stage to improve understanding of their nutrition in relation to aquaculture. Lipid is critical in the nutrition of larval crustaceans, including lobsters. Specimens were from Tasmanian waters, Australia, and North Island, New Zealand, waters. Analyses were by TLC-FID and capillary GC and GC-MS. Phyllosomas larvae and nektonic pueruli were low in storage lipid (triacylglycerol), and phospholipid was the major lipid class. Sterol, mainly cholesterol, was the next most abundant class. The ratio of the essential omega-3 fatty acid eicosapentaenoic acid (EPA) to the omega-6 fatty acid arachidonic acid (AA) was lower in newly hatched phyllosomas (1.2-1.3) than in other phyllosomas (stages III-XI; 2.8-6.7) and pueruli (3.8). Ratios of the omega-3 fatty acid DHA (docosahexaenoic acid) to EPA were also lower in newly hatched phyllosomas (0.5) than in later-stage phyllosomas (1.5-2.1) and pueruli (1.2). We have followed up these compositional data by successfully enriching the live diet (Artemia) of early phyllosomas with AA, EPA and DHA. This dietary manipulation has achieved ratios of these key polyunsaturated fatty acids similar to those of wild phyllosomas. These findings will be of significance to the future of rock-lobster aquaculture

    Spatial Ecology of Sub-Adult Green Turtles in Coastal Waters of the Turks and Caicos Islands: Implications for Conservation Management

Requests to access the datasets should be directed to [email protected] turtles are of conservation concern throughout their range, with past population declines largely due to exploitation through both legal and illegal take, and incidental capture in fisheries. Whilst much research effort has been focused on nesting beaches and elaborating migratory corridors, these species spend the vast majority of their lifecycle in foraging grounds, which are, in some species, quite discrete. To understand and manage these populations, empirical data are needed on distribution, space-use, and habitats to best inform design of protective measures. Here we describe space-use, occupancy, and wide-ranging movements derived from conventional flipper tagging and satellite tracking of sub-adult green turtles (Chelonia mydas) within the coastal waters of the Turks and Caicos Islands (TCI; 2011–2017). 623 turtles were fitted with flipper tags, with 69 subsequently recaptured, five of which in international waters. Sixteen individual turtles of between 63 and 81 cm curved carapace length were satellite tracked for a mean 226 days (range: 38–496). Data revealed extended periods of occupancy in the shallow coastal waters within a RAMSAR protected area. Satellite tracking and flipper tagging showed wide-ranging movements, with flipper tag recaptures occurring in waters off Nicaragua (n = 4), and Venezuela (n = 1). Also, four of 16 satellite tracked turtles exhibiting directed movements away (displaced >450 km) from TCI waters traveling through nine geo-political zones within the Caribbean-Atlantic basin, as well as on the High Seas. One turtle traveled to the Central American coast before settling on inshore habitat in Colombia’s waters for 162 days before transmission ceased, indicating ontogenetic dispersal to a distant foraging habitat. These data highlight connectivity throughout the region, displaying key linkages between countries that have previously only been linked by genetic evidence.     BACKGROUND: Cellulose acetate phthalate (CAP), a pharmaceutical excipient used for enteric film coating of capsules and tablets, was shown to inhibit infection by the human immunodeficiency virus type 1 (HIV-1) and several herpesviruses. CAP formulations inactivated HIV-1, herpesvirus types 1 (HSV-1) and 2 (HSV-2) and the major nonviral sexually transmitted disease (STD) pathogens and were effective in animal models for vaginal infection by HSV-2 and simian immunodeficiency virus. METHODS: Enzyme-linked immunoassays and flow cytometry were used to demonstrate CAP binding to HIV-1 and to define the binding site on the virus envelope. RESULTS: 1) CAP binds to HIV-1 virus particles and to the envelope glycoprotein gp120; 2) this leads to blockade of the gp120 V3 loop and other gp120 sites resulting in diminished reactivity with HIV-1 coreceptors CXCR4 and CCR5; 3) CAP binding to HIV-1 virions impairs their infectivity; 4) these findings apply to both HIV-1 IIIB, an X4 virus, and HIV-1 BaL, an R5 virus. CONCLUSIONS: These results provide support for consideration of CAP as a topical microbicide of choice for prevention of STDs, including HIV-1 infection

    Anti-HIV-1 activity of cellulose acetate phthalate: Synergy with soluble CD4 and induction of "dead-end" gp41 six-helix bundles

    BACKGROUND: Cellulose acetate phthalate (CAP), a promising candidate microbicide for prevention of sexual transmission of the human immunodeficiency virus type 1 (HIV-1) and other sexually transmitted disease (STD) pathogens, was shown to inactivate HIV-1 and to block the coreceptor binding site on the virus envelope glycoprotein gp120. It did not interfere with virus binding to CD4. Since CD4 is the primary cellular receptor for HIV-1, it was of interest to study CAP binding to HIV-1 complexes with soluble CD4 (sCD4) and its consequences, including changes in the conformation of the envelope glycoprotein gp41 within virus particles. METHODS: Enzyme-linked immunosorbent assays (ELISA) were used to study CAP binding to HIV-1-sCD4 complexes and to detect gp41 six-helix bundles accessible on virus particles using antibodies specific for the α-helical core domain of gp41. RESULTS: 1) Pretreatment of HIV-1 with sCD4 augments subsequent binding of CAP; 2) there is synergism between CAP and sCD4 for inhibition of HIV-1 infection; 3) treatment of HIV-1 with CAP induced the formation of gp41 six-helix bundles. CONCLUSIONS: CAP and sCD4 bind to distinct sites on HIV-1 IIIB and BaL virions and their simultaneous binding has profound effects on virus structure and infectivity. The formation of gp41 six-helical bundles, induced by CAP, is known to render the virus incompetent for fusion with target cells thus preventing infection
