    Review on Fenugreek (Trigonella foenum-graecum L.) and its important secondary metabolite diosgenin

    Fenugreek (Trigonella foenum-graecum L.) is a medicinal plant used worldwide since ancient times. Its use as smelling agent and spice was documented since 15th century. The genus Trigonella includes around 260 species diffused worldwide and belonging to Fabaceae family. In the last decades, a number of studies highlighted the biological activities and therapeutic properties of this species mainly attributed to bioactive secondary metabolites such as alkaloids, flavonoids, steroids and saponins. In particular, diosgenin, a steroidal saponin, has been investigated for its medicinal uses and fenugreek has been reported as source of raw material for the production of steroidal hormones. This review article focuses on the cultivation, genetics, ecophysiology and traditional uses of fenugreek, as well as on its medicinal properties, phytochemical and nutrient contents. Extraction procedures and pharmacological activities of diosgenin are also reviewed, as well as methods for its chemical analyses. This review focuses on the medicinal importance of Fenugreek and its important secondary metabolite diosgenin. The review article complies the results of pre-clinical studies conducted to establish the various medicinal applications of diosgenin. This will help researcher to discover fundamental role of diosgenin as a potential product for drug manufacturers and use of fenugreek as a source of diosgenin

    Validation of a method for diosgenin extraction from fenugreek (Trigonella foenum-graecum L.)

    Background. Diosgenin is a very important plant secondary metabolite and raw material for the drug industry. Plant sources rich in diosgenin include yam (Dioscorea spp.) and fenugreek (Trigonella foenum-graecum L.). A method for diosgenin extraction from yam extracts has previously been validated, but its extraction from fenugreek plants still requires validation. In addition, all available methods require time-consuming additional purification steps. The present study was aimed at developing a low cost, less time-consuming single-step method for diosgenin extraction from fenugreek. Material and methods. This study represents a method developed for diosgenin extraction from fenugreek plants without any additional/supportive purification methods such as chromatography or thin-layer chromatography. Diosgenin yield estimation and purity analysis by HPLC method, along with accuracy and precision analysis, is presented. Results. Five different fenugreek varieties were subjected to a newly developed diosgenin extraction method, and an HPLC chromatogram showed a single peak corresponding to diosgenin. Yield was determined by the standard curve method. Limit of detection (LOD) and limit of quantification (LOQ) for the assay were found to be 0.0312 and 0.102 \u3bcg, respectively; t calculated for slope and other statistical parameters were found to be significant (P value < 0.001) for this method. Conclusion. We have developed a fast, accurate and low cost method for diosgenin extraction from fenugreek. Although the authors have studied this method only in fenugreek plants, it could be applied to the extraction of a few other plant secondary metabolites, which will help researchers to save time and effort

    Elicitation of diosgenin production in Trigonella foenum-graecum (fenugreek) seedlings by methyl jasmonate

    The effects of methyl jasmonate (MeJA), an elicitor of plant defense mechanisms, on the biosynthesis of diosgenin, a steroidal saponin, were investigated in six fenugreek (Trigonella foenum-graecum) varieties (Gujarat Methi-2, Kasuri-1, Kasuri-2, Pusa Early Branching, Rajasthan Methi and Maharashtra Methi-5). Treatment with 0.01% MeJA increased diosgenin levels, in 12 days old seedlings, from 0.5%-0.9% to 1.1%-1.8%. In addition, MeJA upregulated the expression of two pivotal genes of the mevalonate pathway, the metabolic route leading to diosgenin: 3-hydroxy-3-methylglutaryl-CoA reductase (HMG) and sterol-3-\u3b2-glucosyl transferase (STRL). In particular, MeJA increased the expression of HMG and STRL genes by 3.2- and 22.2-fold, respectively, in the Gujarat Methi-2 variety, and by 25.4- and 28.4-fold, respectively, in the Kasuri-2 variety. Therefore, MeJA may be considered a promising elicitor for diosgenin production by fenugreek plants

    Thermomagnetic history effects in SmMn2_2Ge2_2

    The intermetallic compound SmMn2_2Ge2_2, displaying multiple magnetic phase transitions, is being investigated in detail for its magnetization behavior near the 145 K first order ferromagnetic to antiferromagnetic transition occuring on cooling, in particular for thermomagnetic history effects in the magnetization data. The most unusual finding is that the thermomagnetic irreversibility, [= MFCW^{FCW}(T)-MZFC^{ZFC}(T)] at 135 K is higher in intermediate magnetic field strengths. By studying the response of the sample (i.e., thermomagnetic irreversibility and thermal hysteresis) to different histories of application of magnetic field and temperature, we demonstrate how the supercooling and superheating of the metastable magnetic phases across the first order transition at 145 K contribute to overall thermomagnetic irreversibility.Comment: 15 pages, 5 figures, to appear in Physical Review

    Reducing the environmental impact of surgery on a global scale: systematic review and co-prioritization with healthcare workers in 132 countries

    Background Healthcare cannot achieve net-zero carbon without addressing operating theatres. The aim of this study was to prioritize feasible interventions to reduce the environmental impact of operating theatres. Methods This study adopted a four-phase Delphi consensus co-prioritization methodology. In phase 1, a systematic review of published interventions and global consultation of perioperative healthcare professionals were used to longlist interventions. In phase 2, iterative thematic analysis consolidated comparable interventions into a shortlist. In phase 3, the shortlist was co-prioritized based on patient and clinician views on acceptability, feasibility, and safety. In phase 4, ranked lists of interventions were presented by their relevance to high-income countries and low–middle-income countries. Results In phase 1, 43 interventions were identified, which had low uptake in practice according to 3042 professionals globally. In phase 2, a shortlist of 15 intervention domains was generated. In phase 3, interventions were deemed acceptable for more than 90 per cent of patients except for reducing general anaesthesia (84 per cent) and re-sterilization of ‘single-use’ consumables (86 per cent). In phase 4, the top three shortlisted interventions for high-income countries were: introducing recycling; reducing use of anaesthetic gases; and appropriate clinical waste processing. In phase 4, the top three shortlisted interventions for low–middle-income countries were: introducing reusable surgical devices; reducing use of consumables; and reducing the use of general anaesthesia. Conclusion This is a step toward environmentally sustainable operating environments with actionable interventions applicable to both high– and low–middle–income countries

    Not AvailableUsing a unique set of household-level data from the Indian state of Punjab, this paper assesses the performance and financing of dairy value chains at their upstream. We find co-existence of formal value chains driven by dairy cooperatives and private processors including multinationals and informal value chains driven by vendors or local traders and consumer-households. The resource-rich dairy farmers prefer partnerships with private dairy processors or vice versa. The small dairy farmers are more dependent on informal channels for the sale of their produce. Although, there is no significant difference in milk yield across herd sizes and value chains, the farmers associated with cooperative value chain earn more profit. The findings also indicate the practice of scale-based price discrimination in the formal segment, especially by the multinationals. Further, more than half of the dairy farmers finance their dairying activities borrowing from the formal as well as informal sources. The chain-based financing is restricted to the value chains driven by the local traders and private domestic processors. The financing by commercial banks is limited and is biased in favour of resource-rich dairy farmers.Not Availabl

    A Web Server and Mobile App for Computing Hemolytic Potency of Peptides

    Numerous therapeutic peptides do not enter the clinical trials just because of their high hemolytic activity. Recently, we developed a database, Hemolytik, for maintaining experimentally validated hemolytic and non-hemolytic peptides. The present study describes a web server and mobile app developed for predicting, and screening of peptides having hemolytic potency. Firstly, we generated a dataset HemoPI-1 that contains 552 hemolytic peptides extracted from Hemolytik database and 552 random non-hemolytic peptides (from Swiss-Prot). The sequence analysis of these peptides revealed that certain residues (e.g., L, K, F, W) and motifs (e.g., “FKK”, “LKL”, “KKLL”, “KWK”, “VLK”, “CYCR”, “CRR”, “RFC”, “RRR”, “LKKL”) are more abundant in hemolytic peptides. Therefore, we developed models for discriminating hemolytic and non-hemolytic peptides using various machine learning techniques and achieved more than 95% accuracy. We also developed models for discriminating peptides having high and low hemolytic potential on different datasets called HemoPI-2 and HemoPI-3.
