    Flow injection determination of total catechins and procyanidins in white and red wines

    An easily automatable flow-injection (FI) method for the determination of total catechins is reported. The method is based on the reaction of vanillin in acid medium to yield a coloured product with maximum absorption at 500 nm. After optimisation by the univariate and multivariate approaches as required, the linear range was established (between 10 and 90 mgL-1 and 10 and 250 mgL-1 for white and red wines, respectively). Then, the assessment of the proposed versus the reference method was studied in terms of repeatability (2.57 mgL-1), reproducibility (3.56 mgL-1) (no significant differences were found), detection and quantification limits (not far from those of the reference method and always sufficient for the determination of catechins in any type of wine), traceability (excellent correlation under all conditions) and sample throughput (23 samples h-1 for the proposed method versus 3 samples h-1 for the reference method)

    Effects of highly active antiretroviral therapy on semen parameters of a cohort of 770 HIV-1 infected men

    Background HIV-1 infected patients show impaired semen parameters. Currently, it is not clear whether HIV-1 infection itself or antiretroviral therapy have an effect on semen parameters. We aim evaluate semen quality in a large cohort of fertile HIV-1 infected men under stable highly active antiretroviral therapy (HAART) and to assess the effect of HAART type and duration on semen parameters. Materials and methods Between January 2010 and June 2014, we enrolled in a retrospective case-control study 770 HIV-1 patients under stable HAART asking a reproductive counselling with their HIV negative partner. Co-infections with HBV or HCV, genital tract infections and known causes of infertility represented exclusion criteria. Semen samples were analysed and compared with the WHO reference values. A multivariate analysis including HAART type and duration, age, viral load and CD4 count, was performed on 600 patients out of 770. Results The median values of all semen parameters were significantly lower among HIV-1 infected patients compared to the WHO reference group, with a significant proportion of patients having values below the 5th percentile of the WHO reference value. In a multivariate analysis, only age and viral load negatively impacted progressive motility (\u3b2 -0.3 (95% CI: -0.5; -0.0) %, p<0.05) and semen morphology (\u3b2 -0.00 (95% CI: -0.00; -0.00) %, p0.01), while no associations were detected as regards HAART type and duration. Conclusions HIV-1 infected patients showed a significant impairment of semen parameters compared to the reference values. HAART type and duration showed no associations with semen quality. Further research is needed to investigate implications for clinical care of HIV infected men desiring a child

    The first report of Tityus (Scorpiones: Buthidae) in Anzoátegui State, Venezuela: a new species

    Tityus gonzalespongai n. sp. is a species endemic to the high mountains of Anzoátegui State, Venezuela. It is found between 1,600 and 2,200 m in ''La Laguna'' mountain. Its habitat includes the area of ''Bosque Húmedo Montano Bajo'' with the type of vegetation of ''Bosque Ombrófilo Montano Siempreverde'' (''Bosques Nublados Costeros'' that includes the ''Subpáramos Arbustivos''). It is distinguished from other Tityus species (T. caripitensis, T. monaguensis, and T. nororientalis) by the following characteristics: 1- the disposition of the trichobothria in the pedipalps; 2- ventral keels of the caudal segments of the metasoma (segment II, double and parallel in the proximal two thirds, then convergent and finally divergent in the base; segments III and IV, double and parallel in the basal third, then convergent in a single keel that divides in the base); 3- the number of lines of denticles of the movable finger of the right pedipalp (male = 14; female = 14); 4- pectineous teeth (right/left: male = 15/15: female = 16/15); 5- color: movable and fixed fingers dark brown; prosoma and metasoma, ochre; caudal segment IV, slightly darker than the previous ones; V and the telson, dark brown. T. gonzalespongai belongs to the ''androcottoides'' group and presents a marked sexual dimorphism. It is the first species of the Tityus genus described and reported in Anzoátegui State, expanding this taxa distribution in Venezuela

    The battle of the sexes starts in the oviduct: modulation of oviductal transcriptome by X and Y-bearing spermatozoa

    BACKGROUND:Sex allocation of offspring in mammals is usually considered as a matter of chance, being dependent on whether an X- or a Y-chromosome-bearing spermatozoon reaches the oocyte first. Here we investigated the alternative possibility, namely that the oviducts can recognise X- and Y- spermatozoa, and may thus be able to bias the offspring sex ratio. RESULTS:By introducing X- or Y-sperm populations into the two separate oviducts of single female pigs using bilateral laparoscopic insemination we found that the spermatozoa did indeed elicit sex-specific transcriptomic responses. Microarray analysis revealed that 501 were consistently altered (P-value <0.05) in the oviduct in the presence of Y-chromosome-bearing spermatozoa compared to the presence of X-chromosome-bearing spermatozoa. From these 501 transcripts, 271 transcripts (54.1%) were down-regulated and 230 transcripts (45.9%) were up-regulated when the Y- chromosome-bearing spermatozoa was present in the oviduct. Our data showed that local immune responses specific to each sperm type were elicited within the oviduct. In addition, either type of spermatozoa elicits sex-specific signal transduction signalling by oviductal cells. CONCLUSIONS:Our data suggest that the oviduct functions as a biological sensor that screens the spermatozoon, and then responds by modifying the oviductal environment.     Design Time Optimization for Hardware Watermarking Protection of HDL Designs

    HDL-level design offers important advantages for the application of watermarking to IP cores, but its complexity also requires tools automating these watermarking algorithms. A new tool for signature distribution through combinational logic is proposed in this work. IPP@HDL, a previously proposed high-level watermarking technique, has been employed for evaluating the tool. IPP@HDL relies on spreading the bits of a digital signature at the HDL design level using combinational logic included within the original system. The development of this new tool for the signature distribution has not only extended and eased the applicability of this IPP technique, but it has also improved the signature hosting process itself. Three algorithms were studied in order to develop this automated tool. The selection of a cost function determines the best hosting solutions in terms of area and performance penalties on the IP core to protect. An 1D-DWT core and MD5 and SHA1 digital signatures were used in order to illustrate the benefits of the new tool and its optimization related to the extraction logic resources. Among the proposed algorithms, the alternative based on simulated annealing reduces the additional resources while maintaining an acceptable computation time and also saving designer effort and time

    Effects of Antiretroviral Therapy on Sperm DNA Integrity of HIV-1-Infected Men

    HIV-1-affected couples\u2019 desire to have children and free sexual intercourses with the use of pre-exposure prophylaxis for the negative partner has emerged as an alternative option to assisted reproduction in aviremic patients under highly active antiretroviral therapy (HAART). It is already known that sperm quality may be impaired in HIV-infected men. The underlying physiopathological mechanism is still debated. The aim of this study was to evaluate the effects of HAART on sperm DNA fragmentation, comparing HIV-1-infected patients taking HAART versus na\uefve HIV-1-infected patients. This is a prospective case-control study. Sperm nuclear DNA fragmentation rate was evaluated by the sperm chromatin dispersion test in 77 HIV-infected men: 53 HIV-1 patients receiving HAART (Group 1) versus 24 na\uefve HIV-1 patients not receiving HAART (Group 2). Complete semen analysis was performed according to WHO 2010 recommendations. Patients with HBV infection or HCV infection coinfections and genital tract infections wre excluded. All the patients did not present any clinical signs of their disease. Seminal parameters were examined in the two groups, showing no significant differences. Increased sperm DNA fragmentation > 30% was demonstrated in 67.9% of patients in Group 1 and 37.5% of patients in Group 2, respectively (p =.02). A positive but nonsignificant trend toward increased fragmentation was reported with advancing patients\u2019 age. In conclusion, sperm nuclear fragmentation rate is increased in HIV-1-infected patients taking HAART compared to HIV-1 patients not receiving HAART

    La Cantalera: an exceptional window onto the vertebrate biodiversity of the Hauterivian-Barremian transition in the Iberian Peninsula.

    La Cantalera is an accumulation site for fossil vertebrates consisting mainly of teeth and isolated postcranial remains. It has the greatest vertebrate biodiversity of any site from the Hauterivian-Barremian transition in the Iberian Peninsula. Up to now, 31 vertebrate taxa have been recognized: an osteichthyan (Teleostei indet.), two amphibians (Albanerpetonidae indet. and Discoglossidae indet.), a chelonian (Pleurosternidae? indet.), a lizard (Paramacellodidae? indet.), four crocodylomorphs (cf. Theriosuchus sp., Bernissartiidae indet., Goniopholididae indet., cf. Lisboasaurus sp.), two pterosaurs (Istiodactylidae? indet., Ornithocheiridae? indet.), four ornithopod dinosaurs (Iguanodontoidea indet., Hadrosauroidea? indet., “Hypsilophodontidae” indet., Rhabdodontidae? indet.), a thyreophoran (Polacanthidae indet.), a sauropod (Euhelopodidae indet.), eleven theropods (Carcharodontosauridae? indet., Baryonychinae indet., aff. Paronychodon sp., Maniraptora indet. 1-3, Dromaeosaurinae indet. 1-3, Velociraptorinae indet., Avialae? indet.) and three or four multituberculate mammals (Cantalera abadi, Eobaatar sp., Plagiaulacidae or Eobaataridae gen. et sp. indet., “Plagiaulacida” indet.). Nine ootaxa have also been distinguished at the site of La Cantalera. Oofamilies assigned to dinosaurs (Elongaloolithidae, Prismatoolithidae, cf. Spheroolithidae), to crocodiles (Krokolithidae) and eggshells of two oofamilies incertidae sedis are represented. This association is consistent with the record of bone and tooth remains from the site. La Cantalera was formed in a marshy environment where there was an intermittent body of water. The great majority of the vertebrate fossil remains lack evidence of transport, so the preserved association is a good representation of the vertebrate ecosystem in or around the marshy area of La Cantalera. The vertebrate association of La Cantalera displays certain differences with respect to those from lacustrine environments of the Hauterivian-Barremian transition of the Iberian Range. These differences include, for example, the absence of Chondrichthyes, the merely token presence of the osteichthyans, the scarcity of chelonians, the presence of exclusively multituberculate mammals, the lower diversity of sauropods and the greater diversity of theropods

    Mowat-Wilson syndrome (MWS) is a multiple congenital anomaly syndrome characterized by a distinct facial phenotype (high forehead, frontal bossing, large eyebrows, medially flaring and sparse in the middle part, hypertelorism, deep set but large eyes, large and uplifted ear lobes, with a central depression, saddle nose with prominent rounded nasal tip, prominent columella, open mouth, with M-shaped upper lip, frequent smiling, and a prominent but narrow and triangular pointed chin), moderate-to-severe intellectual deficiency, epilepsy and variable congenital malformations including Hirschsprung disease (HSCR), genitourinary anomalies (in particular hypospadias in males), congenital heart defects, agenesis of the corpus callosum and eye anomalies. The prevalence of MWS is currently unknown, but 171 patients have been reported so far. It seems probable that MWS is under-diagnosed, particularly in patients without HSCR. MWS is caused by heterozygous mutations or deletions in the Zinc finger E-box-binding homeobox 2 gene, ZEB2, previously called ZFHX1B (SIP1). To date, over 100 deletions/mutations have been reported in patients with a typical phenotype; they are frequently whole gene deletions or truncating mutations, suggesting that haploinsufficiency is the main pathological mechanism. Studies of genotype-phenotype analysis show that facial gestalt and delayed psychomotor development are constant clinical features, while the frequent and severe congenital malformations are variable. In a small number of patients, unusual mutations can lead to an atypical phenotype. The facial phenotype is particularly important for the initial clinical diagnosis and provides the hallmark warranting ZEB2 mutational analysis, even in the absence of HSCR. The majority of MWS cases reported so far were sporadic, therefore the recurrence risk is low. Nevertheless, rare cases of sibling recurrence have been observed. Congenital malformations and seizures require precocious clinical investigation with intervention of several specialists (including neonatologists and pediatricians). Psychomotor development is delayed in all patients, therefore rehabilitation (physical therapy, psychomotor and speech therapy) should be started as soon as possible
