    Competition, restructuring and firm performance: evidence of an inverted-U relationship from a cross-country survey of firms in transition economies

    This paper examines the importance of competition in the growth anddevelopment of firms. We draw on a survey of 3,300 firms in 25transition countries to shed light on the factors that influencerestructuring by firms and their subsequent performance. These datahave three main advantages over those used in previous work. First,they measure directly the degree of competition perceived by each firmin its principal market rather than attempting to infer this from marketdata as measured by statistical agencies. Second, the fact that transitioncountries have market structures inherited from the past avoids some ofthe endogeneity problems associated with measures of competition inmarket economies. Third, the breadth of cross-country variationprovides a method of dealing with the fact that firm-level measures ofthe external environment will not be independent of the firm?s ownperformance. We find evidence of a robust inverted-U effect ofcompetition on performance that is both statistically and economicallysignificant.     A Guide to Academic Freedom

    This brief introduction to the principles of academic freedom is intended for attorneys and other administrators who represent or work at colleges and universities. It has two purposes. The first is to introduce them to academic freedom as a set of professional principles regardless of whether or not they are legally enforceable. Attorneys and administrators need to understand the culture of the institutions they represent or serve. Nowhere is this more true than with colleges and universities, which have well established traditions and norms that influence the expectations and conduct of all those responsible for their governance, including faculty, administrators and trustees. The second purpose is to introduce the law relating to academic freedom as it has evolved over the last half century. As will become apparent, it is not always clear where academic freedom as a set of professional principles ends and the law begins. Academic freedom has received some recognition by the Supreme Court and considerably more by the lower federal courts in connection with the application of the First Amendment to cases involving both universities as institutions and the individual rights of faculty. However, the meaning of academic freedom in the context of constitutional law is confused. Apart from its constitutional dimension, academic freedom as a legal principle results from its incorporation into contracts or collective bargaining agreements between universities and faculty or into policies, guidelines or handbooks adopted or issued by universities that may or may not create contractual rights. It is not possible in an introduction to the subject of academic freedom to cover these complex issues of contract law and interpretation. Rather, the goal of the present work is merely to present what principles are or are not part of the definition of academic freedom and how they may be fairly applied in some of the most common contexts in which they arise. This guide was the outgrowth of several meetings over the course of two years sponsored by the Ford Foundation, as part of its “Difficult Dialogues Initiative,” and with the active support of the National Association of College and University Attorneys. I have benefitted greatly from the discussions at those meetings and from the comments of many of its participants on drafts of this guide

    Stochastic Gene Expression in a Lentiviral Positive Feedback Loop: HIV-1 Tat Fluctuations Drive Phenotypic Diversity

    Stochastic gene expression has been implicated in a variety of cellular processes, including cell differentiation and disease. In this issue of Cell, Weinberger et al. (2005) take an integrated computational-experimental approach to study the Tat transactivation feedback loop in HIV-1 and show that fluctuations in a key regulator, Tat, can result in a phenotypic bifurcation. This phenomenon is observed in an isogenic population where individual cells display two distinct expression states corresponding to latent and productive infection by HIV-1. These findings demonstrate the importance of stochastic gene expression in molecular "decision-making."Comment: Supplemental data available as q-bio.MN/060800

    Inference of Temporally Varying Bayesian Networks

    When analysing gene expression time series data an often overlooked but crucial aspect of the model is that the regulatory network structure may change over time. Whilst some approaches have addressed this problem previously in the literature, many are not well suited to the sequential nature of the data. Here we present a method that allows us to infer regulatory network structures that may vary between time points, utilising a set of hidden states that describe the network structure at a given time point. To model the distribution of the hidden states we have applied the Hierarchical Dirichlet Process Hideen Markov Model, a nonparametric extension of the traditional Hidden Markov Model, that does not require us to fix the number of hidden states in advance. We apply our method to exisiting microarray expression data as well as demonstrating is efficacy on simulated test data

    Single-cell western blotting.

    To measure cell-to-cell variation in protein-mediated functions, we developed an approach to conduct ∌10(3) concurrent single-cell western blots (scWesterns) in ∌4 h. A microscope slide supporting a 30-ÎŒm-thick photoactive polyacrylamide gel enables western blotting: settling of single cells into microwells, lysis in situ, gel electrophoresis, photoinitiated blotting to immobilize proteins and antibody probing. We applied this scWestern method to monitor single-cell differentiation of rat neural stem cells and responses to mitogen stimulation. The scWestern quantified target proteins even with off-target antibody binding, multiplexed to 11 protein targets per single cell with detection thresholds of <30,000 molecules, and supported analyses of low starting cell numbers (∌200) when integrated with FACS. The scWestern overcomes limitations of antibody fidelity and sensitivity in other single-cell protein analysis methods and constitutes a versatile tool for the study of complex cell populations at single-cell resolution

    Benefit-cost methodology study with example application of the use of wind generators

    An example application for cost-benefit methodology is presented for the use of wind generators. The approach adopted for the example application consisted of the following activities: (1) surveying of the available wind data and wind power system information, (2) developing models which quantitatively described wind distributions, wind power systems, and cost-benefit differences between conventional systems and wind power systems, and (3) applying the cost-benefit methodology to compare a conventional electrical energy generation system with systems which included wind power generators. Wind speed distribution data were obtained from sites throughout the contiguous United States and were used to compute plant factor contours shown on an annual and seasonal basis. Plant factor values (ratio of average output power to rated power) are found to be as high as 0.6 (on an annual average basis) in portions of the central U. S. and in sections of the New England coastal area. Two types of wind power systems were selected for the application of the cost-benefit methodology. A cost-benefit model was designed and implemented on a computer to establish a practical tool for studying the relative costs and benefits of wind power systems under a variety of conditions and to efficiently and effectively perform associated sensitivity analyses

    Quantum Phase Transition in Heisenberg-Kitaev Model

    We explore the nature of the quantum phase transition between a magnetically ordered state with collinear spin pattern and a gapless Z2Z_2 spin liquid in the Heisenberg-Kitaev model. We construct a slave particle mean field theory for the Heisenberg-Kitaev model in terms of complex fermionic spinons. It is shown that this theory, formulated in the appropriate basis, is capable of describing the Kitaev spin liquid as well as the transition between the gapless Z2Z_2 spin liquid and the so-called stripy antiferromagnet. In particular, within a mean field theory, we have a discontinuous transition from the Z2Z_2 spin liquid to the stripy antiferromagnet. We argue, however, that subtle spinon confinement effects, associated with the instability of gapped U(1) spin liquid in two spatial dimensions, are playing an important role at the transition. The possibility of an exotic continuous transition is briefly addressed.Comment: 12 pages, 6 figure
