    The HLA-E Gene encodes two differentially regulated Transcripts and a Cell Surface Protein

    An HLA-E-specific oligonucleotide probe was used to study the expressioonf HLA-E. This probed etects two HLA-E transcripts, 1.8 and 2.7 kb in size, which are present in varying ratios in allt issues and cell lines investigated. We demonstrate that alternative poly(A) site usage accounts for the differential regulation of the two HLA-E mRNA species. Sequence analysis of three cDNA clones, representing the two transcripts of HLA-E, and of anH LA-E gene encoded by cosmid cd3.14, revealed identity of gene and cDNA in the 3’ untranslated region. S1 nuclease protection assays confirmed that the two HLA-E transcripts are not alternative splicing products. Introduction of cd3.14, together with human ,&m into the murine myeloma cell line P3X63-Ag8.653, resulted in a cell surface expresosf ioan HLA-class I heavy chain detectablbey indirect immunofluorescence whereas transfection into the humBaznr n expressing mouse L cell line, 527 was negative with regard to cell surface expressionC. ell surface labeling of transfectants and immunoprecipitation with a monomorphic HLA class I-specific antibodyo r an antibody against human &m confirmed the presence of an HLA-E H chain on the cell surface.     A sidereal rotation counting approach is demonstrated by resolving an ambiguity in the synodic rotation period of Koronis family member (3032) Evans, whose rotation lightcurves' features did not easily distinguish between doubly- and quadruply-periodic. It confirms that Evans's spin rate does not exceed the rubble-pile spin barrier and thus presents no inconsistency with being a ~14-km reaccumulated object. The full spin vector solution for Evans is comparable to those for the known prograde low-obliquity comparably-fast rotators in the Koronis family, consistent with having been spun up by YORP thermal radiation torques.Comment: 8 pages, 6 figures, accepted for publication in Icaru

    Charon's radius and density from the combined data sets of the 2005 July 11 occultation

    The 2005 July 11 C313.2 stellar occultation by Charon was observed by three separate research groups, including our own, at observatories throughout South America. Here, the published timings from the three data sets have been combined to more accurately determine the mean radius of Charon: 606.0 +/- 1.5 km. Our analysis indicates that a slight oblateness in the body (0.006 +/- 0.003) best matches the data, with a confidence level of 86%. The oblateness has a pole position angle of 71.4 deg +/- 10.4 deg and is consistent with Charon's pole position angle of 67 deg. Charon's mean radius corresponds to a bulk density of 1.63 +/- 0.07 g/cm3, which is significantly less than Pluto's (1.92 +/- 0.12 g/cm3). This density differential favors an impact formation scenario for the system in which at least one of the impactors was differentiated. Finally, unexplained differences between chord timings measured at Cerro Pachon and the rest of the data set could be indicative of a depression as deep as 7 km on Charon's limb.Comment: 25 pages including 4 tables and 2 figures.     Operational streamlining in a high-throughput genome sequencing center

It is therefore critical for the Genome Sequencing platform of the Broad Institute of MIT and Harvard to continually strive to reduce cost, improve throughput, and increase the quality of its data output. In the past, new technology in the form of both chemistry improvements and robotics has allowed the Institute to achieve these goals in a step-wise manner. However, as the rate of technology progression in sequencing has slowed, the Institute has been forced to look to continuous, incremental improvement in order to achieve its goals. The Core Sequencing/Detection group handles the high-throughput sequencing duties at the Broad Institute. Through the use of robotics and cutting edge biology, they are able to process and sequence upwards of 50 billion bases of DNA per year. The work that this thesis was based on took place primarily in this automated production area. This thesis utilizes a number of lean concepts, including the 7 Wastes and pull production control.(cont.) Kanban systems, workflow changes, and a 5S implementation were used to bring these concepts to life at the Broad Institute. In order to correctly size the kanban system, process buildup diagrams and discrete event simulation were used.     Long-term outcome of noninvasive positive pressure ventilation for obesity hypoventilation syndrome

    BACKGROUND: Few data are available on the long-term outcome of noninvasive positive pressure ventilation (NPPV) for obesity hypoventilation syndrome (OHS). This study was designed to determine long-term survival, treatment adherence, and prognostic factors in patients with OHS in whom NPPV was initiated in an acute setting vs under stable clinical conditions.METHODS: One hundred thirty consecutive patients with OHS (56 women) who started NPPV between January 1995 and December 2006 either under stable conditions (stable group, n = 92) or during ICU management of acute hypercapnic exacerbation (acute group, n = 38) were retrospectively analyzed. RESULTS: Arterial blood gases and the Epworth sleepiness scale were both significantly improved after 6 months of NPPV. With a mean follow-up of 4.1 +/- 2.9 years, 24 (18.5%) patients died and 24 (18.5%) discontinued NPPV. On Kaplan-Meier analysis, 1-, 2-, 3-, and 5-year survival probabilities were 97.5%, 93%, 88.3%, and 77.3%, respectively. Mortality was lower than that described in a previous series of patients with untreated OHS. Supplemental oxygen therapy was the only independent predictor of mortality. The probability of continuing NPPV was 80% at 3 years with a high rate of daily use ( > 7 h). Female sex was predictive of lower long-term adherence to NPPV. The acute and stable groups did not differ in terms of arterial blood gases and Epworth sleepiness scale at 6 months, long-term survival, and treatment adherence. CONCLUSIONS: The results of this study support long-term NPPV as an effective and well-tolerated treatment of OHS whether initiated in the acute or chronic setting

    Introduction Venlafaxine and propranolol have rarely been identified as causes of pulmonary pathology. We describe a case of drug-induced pneumonitis occurring in a patient treated with these two medications. Case report A 55 years old woman with liver cirrhosis treated with venlafaxine for 1 year and propranolol for 1 month was admitted to the intensive care unit because of acute respiratory failure. A Mycoplasma pneumoniae pneumonitis was diagnosed. After initial improvement under antibiotics, a new deterioration of respiratory status was observed 4 days after the reintroduction of venlafaxine and propranolol. Spontaneous recovery occurred after these treatments were withheld. Co administration of venlafaxine and propranolol, 2 drugs with affinity for the same cytochrome P450 isoenzyme (CYP2D6), may have contributed to drug accumulation and pulmonary toxicity. The liver cirrhosis of our patient may also have contributed to decreased cytochrome P450 enzymatic activity. Conclusions Venlafaxine and propranolol share the same metabolic pathway and their co-administration may be complicated by drug induced pneumonitis

The antenna is based on a double-side printed multilayer paper substrate and is fed with a coaxial cable for the testing.     The Mission Accessible Near-Earth Objects Survey: Four years of photometry

    Over 4.5 years, the Mission Accessible Near-Earth Object Survey (MANOS) assembled 228 Near-Earth Object (NEO) lightcurves. We report rotational lightcurves for 82 NEOs, constraints on amplitudes and periods for 21 NEOs, lightcurves with no detected variability within the image signal to noise and length of our observing block for 30 NEOs, and 10 tumblers. We uncovered 2 ultra-rapid rotators with periods below 20s; 2016MA with a potential rotational periodicity of 18.4s, and 2017QG18_{18} rotating in 11.9s, and estimate the fraction of fast/ultra-rapid rotators undetected in our project plus the percentage of NEOs with a moderate/long periodicity undetectable during our typical observing blocks. We summarize the findings of a simple model of synthetic NEOs to infer the object morphologies distribution using the measured distribution of lightcurve amplitudes. This model suggests a uniform distribution of axis ratio can reproduce the observed sample. This suggests that the quantity of spherical NEOs (e.g., Bennu) is almost equivalent to the quantity of highly elongated objects (e.g., Itokawa), a result that can be directly tested thanks to shape models from Doppler delay radar imaging analysis. Finally, we fully characterized 2 NEOs as appropriate targets for a potential robotic/human mission: 2013YS2_{2} and 2014FA7_{7} due to their moderate spin periods and low Δv\Delta v.Comment: Accepted for Publication, The Astrophysical Journal Supplement Serie
