    Evaluation of the impact energy of the samples produced by the additive manufacturing technology

    The article covered the evaluation of the impact energy, notch toughness and morphology of the fracture surfaces of the specimens manufactured by the Direct Metal Laser Sintering Technology. Specimens without heat treatment with no notch were not broken through in course of testing, therefore there was no fracture surface present. The heat treatment resulted in the increase in hardness values. The values of impact energy after the heat treatment was approximately 60 % lower. Ductile intergranular fracture with more or less segmented dimple morphology appeared in every specimen. At places where the internal plastic bond was resisting the test, cracks remaining after particles broke away from the surface can be seen as craters

    Zr alloy protection against high-temperature oxidation: Coating by a double-layered structure with active and passive functional properties

    In this work, a new concept of metal surface protection against degradation caused by high-temperature oxidation in water environment is presented. We were the first to create a double-layered coating consisting of an active and passive part to protect Zr alloy surface against high-temperature oxidation in a hot water environment. We investigated the hot steam corrosion of ZIRLO fuel cladding coated with a double layer consisting of 500 nm nanocrystalline diamond (NCD) as the bottom layer and 2 m chromium-aluminum-silicon nitride (CrAlSiN) as the upper layer. Coated and noncoated ZIRLO samples were exposed for 4 days at 400 Ā°C in an autoclave (working water-cooled nuclear reactor temperature) and for 60 minutes at 1000 Ā°C (nuclear reactor accident temperature) in a hot steam furnace. We have shown that the NCD coating protects the Zr alloy surface against oxidation in an active way: carbon from NCD layer enters the Zr alloy surface and, by changing the physical and chemical properties of the Zr cladding tube surface, limits the Zr oxidation process. In contrast, the passive CrAlSiN coating prevents the Zr cladding tube surface from coming into physical contact with the hot steam. The advantages of the double layer were demonstrated, particularly in terms of hot (accident-temperature) oxidation kinetics: in the initial stage, CrAlSiN layer with low number of defects acts as an impermeable barrier. But after a longer time (more than 20 minutes) the protection by more cracked CrAlSiN decreases. At the same time, the carbon from NCD strongly penetrates the Zr cladding surface and worsen conditions for Zr oxidation. For the double-layer coating, the underlying NCD layer mitigates thermal expansion, reducing cracks and defects in upper layer CrAlSiN

    Use of Ultra High Performance Liquid Chromatography-Tandem Mass Spectrometry to Demonstrate Decreased Serum Statin Levels after Extracorporeal LDL-Cholesterol Elimination

    Background. Using our statin analysis method, it was possible to uncover a significant drop in statin levels (atorvastatin, simvastatin, and metabolites) after extracorporeal LDL-cholesterol elimination (EE) in severe familial hypercholesterolemia (FH). The purpose of this work was to identify the mechanism underlying this drop and its clinical significance as well as to propose measures to optimize a pharmacotherapeutical regimen that can prevent the loss of statins. Methods. Ultra High Performance Liquid Chromatography (UHPLC) connected to the triple quadrupole MS/MS system was used. Patients. A group of long-term treated patients (3ā€“12 years of treatment) with severe FH (12 patients) and treated regularly by LDL-apheresis (immunoadsorption) or haemorheopheresis (cascade filtration) were included in this study. Results. After EE, the level of statins and their metabolites decreased (atorvastatin before/after LDL-apheresis: 8.83/3.46ā€‰nmol/l; before/after haemorheopheresis: 37.02/18.94ā€‰nmol/l). A specific loss was found (concentration of atorvastatin for LDL-apheresis/haemorheopheresis: 0.28/3.04ā€‰nmol/l in washing fluids; 11.07ā€‰nmol/l in filters). To prevent substantial loss of statin concentrations, a pharmacotherapeutic regimen with a longer time interval between the dose of statins and EE is recommended (15 hours). Conclusions. A specific loss of statins was found in adsorbent columns and filters. The decrease can be prevented by the suggested dosage scheme

    Origin and Examination of a Leafhopper Facultative Endosymbiont

    Eukaryotes engage in intimate interactions with microbes that range in age and type of association. Although many conspicuous examples of ancient insect associates are studied (e.g., Buchneraaphidicola), fewer examples of younger associations are known. Here, we further characterize a recently evolved bacterial endosymbiont of the leafhopper Euscelidius variegatus (Hemiptera, Cicadellidae), called BEV. We found that BEV, continuously maintained in E. variegatus hosts at UC Berkeley since 1984, is vertically transmitted with high fidelity. Unlike many vertically transmitted, ancient endosymbioses, the BEVā€“E. variegatus association is not obligate for either partner, and BEV can be cultivated axenically. Sufficient BEV colonies were grown and harvested to estimate its genome size and provide a partial survey of the genome sequence. The BEV chromosome is about 3.8Ā Mbp, and there is evidence for an extrachromosomal element roughly 53Ā kb in size (e.g., prophage or plasmid). We sequenced 438Ā kb of unique short-insert clones, representing about 12% of the BEV genome. Nearly half of the gene fragments were similar to mobile DNA, including 15 distinct types of insertion sequences (IS). Analyses revealed that BEV not only shares virulence genes with plant pathogens, but also is closely related to the plant pathogenic genera Dickeya, Pectobacterium, and Brenneria. However, the slightly reduced genome size, abundance of mobile DNA, fastidious growth in culture, and efficient vertical transmission suggest that symbiosis with E. variegatus has had a significant impact on genome evolution in BEV

    Weight Gain Is Associated with Medial Contact Site of Subthalamic Stimulation in Parkinson's Disease

    The aim of our study was to assess changes in body-weight in relation to active electrode contact position in the subthalamic nucleus. Regular body weight measurements were done in 20 patients with advanced Parkinson's disease within a period of 18 months after implantation. T1-weighted (1.5T) magnetic resonance images were used to determine electrode position in the subthalamic nucleus and the Unified Parkinson's disease rating scale (UPDRS-III) was used for motor assessment. The distance of the contacts from the wall of the third ventricle in the mediolateral direction inversely correlated with weight gain (rā€Š=ā€Šāˆ’0.55, p<0.01) and with neurostimulation-related motor condition expressed as the contralateral hemi-body UPDRS-III (rā€Š=ā€Šāˆ’0.42, p<0.01). Patients with at least one contact within 9.3 mm of the wall experienced significantly greater weight gain (9.4Ā±(SD)4.4 kg, Nā€Š=ā€Š11) than those with both contacts located laterally (3.9Ā±2.7 kg, Nā€Š=ā€Š9) (p<0.001). The position of the active contact is critical not only for motor outcome but is also associated with weight gain, suggesting a regional effect of subthalamic stimulation on adjacent structures involved in the central regulation of energy balance, food intake or reward

    Walking of antitumor bifunctional trinuclear PtII complex on double-helical DNA

    The trinuclear BBR3464 ([{trans-PtCl(NH3)2}2Āµ-(trans-Pt(NH3)2(H2N(CH2)6NH2)2)]4+) belongs to the polynuclear class of platinum-based anticancer agents. DNA adducts of this complex differ significantly in structure and type from those of clinically used mononuclear platinum complexes, especially, long-range (Pt, Pt) intrastrand and interstrand cross-links are formed in both 5ā€²ā€“5ā€² and 3ā€²ā€“3ā€² orientations. We show employing short oligonucleotide duplexes containing single, site-specific cross-links of BBR3464 and gel electrophoresis that in contrast to major DNA adducts of clinically used platinum complexes, under physiological conditions the coordination bonds between platinum and N7 of G residues involved in the cross-links of BBR3464 can be cleaved. This cleavage may lead to the linkage isomerization reactions between this metallodrug and double-helical DNA. Differential scanning calorimetry of duplexes containing single, site-specific cross-links of BBR3464 reveals that one of the driving forces that leads to the lability of DNA cross-links of this metallodrug is a difference between the thermodynamic destabilization induced by the cross-link and by the adduct into which it could isomerize. The rearrangements may proceed in the way that cross-links originally formed in one strand of DNA can spontaneously translocate from one DNA strand to its complementary counterpart, which may evoke walking of the platinum complex on DNA molecule
