    On the Change in Archivability of Websites Over Time

    As web technologies evolve, web archivists work to keep up so that our digital history is preserved. Recent advances in web technologies have introduced client-side executed scripts that load data without a referential identifier or that require user interaction (e.g., content loading when the page has scrolled). These advances have made automating methods for capturing web pages more difficult. Because of the evolving schemes of publishing web pages along with the progressive capability of web preservation tools, the archivability of pages on the web has varied over time. In this paper we show that the archivability of a web page can be deduced from the type of page being archived, which aligns with that page's accessibility in respect to dynamic content. We show concrete examples of when these technologies were introduced by referencing mementos of pages that have persisted through a long evolution of available technologies. Identifying these reasons for the inability of these web pages to be archived in the past in respect to accessibility serves as a guide for ensuring that content that has longevity is published using good practice methods that make it available for preservation.Comment: 12 pages, 8 figures, Theory and Practice of Digital Libraries (TPDL) 2013, Valletta, Malt

    An evaluation of the suitability of ERTS data for the purposes of petroleum exploration

    This experiment was designed to determine the types and amounts of information valuable to petroleum exploration extractable from ERTS data and the cost of obtaining the information using traditional or conventional means. It was desired that an evaluation of this new petroleum exploration tool be made in a geologically well known area in order to assess its usefulness in an unknown area. The Anadarko Basin lies in western Oklahoma and the panhandle of Texas. It was chosen as a test site because there is a great deal of published information available on the surface and subsurface geology of the area, and there are many known structures that act as traps for hydrocarbons. This basin is similar to several other large epicontinental sedimentary basins. It was found that ERTS imagery is an excellent tool for reconnaissance exploration of large sedimentary basins or new exploration provinces. For the first time, small and medium size oil companies can rapidly and effectively analyze exploration provinces as a whole

    Continuous Commissioning of a LEED-EB Gold Certified Office Building

    A 320,000 square foot Class A high-rise office building in Nashville, TN achieved LEED-EB Gold Certification. LEED-EB version 2.0 was utilized, which requires Retro-Commissioning (RCx) as a prerequisite for certification. SSRCx performed the RCx services during the LEED-EB performance period, and was contracted after building certification to utilize the Continuous CommissioningĀ® (CCĀ®) process to seek additional energy savings. The building was constructed in 1999 as a Core and Shell project and build-outs were done through 2001. By January 2008, a single company managed and occupied the facility. This paper describes the RCx process for LEED-EB v2.0 and the CC process after LEED Certification. Findings and implementation strategies for energy savings are presented for both processes

    Busting the Myth That Green Costs More Green

    Buildings are one of the largest consumers of resources and energy in this country, and according to the AIA (American Institute of Architects) are responsible for almost half of all carbon emissions in the United States. Since Americans spend nearly 90 percent of their lives indoors, buildings are clearly important to our way of life. The most common misconception about green building is that these approaches cost more to implement than traditional strategies and techniques of design and construction. Any decision made in the early stages of programming and design will have economic impact on the overall building cost. How many floors will our building have? Will we use marble in the lobby? Can we use fancy fixtures in the bathrooms? But according to a Davis Langdon study, there was ā€œā€¦no significant difference in the construction costs for LEEDĀ®-seeking versus non- LEEDĀ® buildingsā€¦ā€ In addition to this widely referenced report, other independent studies by the State of California and the GSA indicate that cost premiums are minimal. More importantly, first cost is only a small part of the total cost of building ownership. Cost-of-ownership studies agree that first cost only accounts for around 10 percent of all costs a building owner will spend over the life of the building. The other 90 percent comes in the form of operation and maintenance ā€“ two areas in which designing for LEEDĀ® certification can save enormously. Any additional costs for building green are recouped in one to two years on average, with exponential cost savings thereafter that leave traditional construction far behind

    Novel Transcriptional Regulatory Signals in the Adeno-Associated Virus Terminal Repeat A/D Junction Element

    Adeno-associated virus (AAV) type 2 vectors transfer stable, long-term gene expression to diverse cell types in vivo. Many gene therapy applications require the control of long-term transgene expression, and AAV vectors, similar to other gene transfer systems, are being evaluated for delivery of regulated gene expression cassettes. Previously, we (R. P. Haberman, T. J. McCown, and R. J. Samulski, Gene Ther. 5:1604ā€“1611, 1998) demonstrated the use of the tetracycline-responsive system for long-term regulated expression in rat brains. In that study, we also observed residual expression in the ā€œoffā€ state both in vitro and in vivo, suggesting that the human cytomegalovirus (CMV) major immediate-early minimal promoter or other cis-acting elements (AAV terminal repeats [TR]) were contributing to this activity. In the present study, we identify that the AAV TR, minus the tetracycline-responsive minimal CMV promoter, will initiate mRNA expression from vector templates. Using deletion analysis and specific PCR-derived TR reporter gene templates, we mapped this activity to a 37-nucleotide stretch in the A/D elements of the TR. Although the mRNA derived from the TR is generated from a non-TATA box element, the use of mutant templates failed to identify function of canonical initiator sequences as previously described. Finally, we demonstrated the presence of green fluorescent protein expression both in vitro and in vivo in brain by using recombinant virus carrying only the TR element. Since the AAV terminal repeat is a necessary component of all recombinant AAV vectors, this TR transcriptional activity may interfere with all regulated expression cassettes and may be a problem in the development of novel TR split gene vectors currently being considered for genes too large to be packaged

    Adeno-associated virus-mediated expression and constitutive secretion of NPY or NPY13-36 suppresses seizure activity in vivo

    Neuropeptide Y (NPY) is a 36-amino-acid peptide that attenuates seizure activity following direct infusion or adeno-associated virus (AAV)-mediated expression in the central nervous system. However, NPY activates all NPY receptor subtypes, potentially causing unwanted side effects. NPY13-36 is a C-terminal peptide fragment of NPY that primarily activates the NPY Y2 receptor, thought to mediate the antiseizure activity. Therefore, we investigated if recombinant adeno-associated virus-mediated expression and constitutive secretion of NPY or NPY13-36 could alter limbic seizure sensitivity. Rats received bilateral piriform cortex infusions of AAV vectors that express and constitutively secrete full-length NPY (AAV-FIB-NPY) or NPY13-36 (AAV-FIB-NPY13-36). Control rats received no infusion, as we have previously shown that vectors expressing and secreting reporter genes like GFP (AAV-FIB-EGFP), as well as vectors expressing peptides that lack secretion sequences (AAV-GAL) have no effect on seizures. One week later, all animals received kainic acid (10 mg kgāˆ’1, intraperitoneally), and the latencies to wet dog shakes and limbic seizure behaviors were determined. Although both control and vector-treated rats developed wet dog shake behaviors with similar latencies, the latencies to class III and class IV limbic seizures were significantly prolonged in both NPY- and NPY13-36-treated groups. Thus, AAV-mediated expression and constitutive secretion of NPY and NPY13-36 is effective in attenuating limbic seizures, and provides a platform for delivering therapeutic peptide fragments with increased receptor selectivity

    Role of modelling in improving nutrient efficiency in cropping systems

    The applicability of models in addressing resource management issues in agriculture has been widely promoted by the research community, yet examples of real impacts of such modelling efforts on current farming practices are rare. Nevertheless, simulation models can compliment traditional field experimentation in researching alternative management options. The first objective of this paper is, therefore, to provide four case study examples of where models were used to help research issues relating to improved nutrient efficiency in low-input cropping systems. The first two cases addressed strategies of augmenting traditional farming practices with small applications of chemical fertilizer (N and P). The latter two cases explicitly addressed the question of what plant genetic traits can be beneficial in low-nutrient farming systems. In each of these case studies, the APSIM (Agricultural Production Systems Simulator) systems model was used to simulate the impacts of alternative crop management systems. The question of whether simulation models can assist the research community in contributing to purposeful change in farming practice is also addressed. Recent experiences in Australia are reported where simulation models have contributed to practice change by farmers. Finally, current initiatives aimed at testing whether models can also contribute to improving the nutrient efficiency of smallholder farmers in the SAT are discussed

    Treatment Strategies Targeting Excess Hippocampal Activity Benefit Aged Rats with Cognitive Impairment

    Excess neural activity in the CA3 region of the hippocampus has been linked to memory impairment in aged rats. We tested whether interventions aimed at reducing this excess activity would improve memory performance. Aged (24 to 28 months old) male Longā€“Evans rats were characterized in a spatial memory task known to depend on the functional integrity of the hippocampus, such that aged rats with identified memory impairment were used in a series of experiments. Overexpression of the inhibitory neuropeptide Y 13ā€“36 in the CA3 via adeno-associated viral transduction was found to improve hippocampal-dependent long-term memory in aged rats, which had been characterized with impairment. Subsequent experiments with two commonly used antiepileptic agents, sodium valproate and levetiracetam, similarly produced dose-dependent memory improvement in such aged rats. Improved spatial memory with low doses of these agents was observed in both appetitve and aversive spatial tasks. The benefits of these different modalities of treatment are consistent with the concept that excess activity in the CA3 region of the hippocampus is a dysfunctional condition that may have a key role underlying age-related impairment in hippocampal-dependent memory processes. Because increased hippocampal activation occurs in age-related memory impairment in humans as observed in functional neuroimaging, the current findings also suggest that low doses of certain antiepileptic drugs in cognitively impaired elderly humans may have therapeutic potential and point to novel targets for this indication

    Abstract From six years of participatory action research has emerged Farmers', Advisers' and Researchers' Monitoring, Simulation. Communication And Performance Evaluation (FARMSCAPE) as an approach for supporting farmers' management of dryland crop production. In contrast to the strategy of producing decision support software for farmers, FARMSCAPE features simulation-aided discussions about management among farmers, advisers, and (sometimes) researchers. The key is a capability to flexibly simulate the consequences of a wide range of crop and cropland management alternatives in a variable climate at a paddock scale using local soil and weather data. The high level of interest among farmers has led to a current focus on transfer of the technology to agricultural service providers. Keywords: Farming systems, on-farm, simulation, soil monitoring, action research The term "farming systems research" is most commonly used in Australia to mean "research on bio-physical sub-systems aimed at improving systems of farming". Research methodology tends to be a flexible and pragmatic use of formal experimental design and statistical analysis. Experiments are designed to represent aspects of farming sufficiently realistically for results to be meaningful to farmers and advisers but without unnecessarily or overly straining professional standards for methodology concerning making valid comparisons with adequate confidence. In the interest of the former, experiments are often located on commercial farms, and, increasingly, with farmers. A second established way of interpreting the term "farming systems research" is "systems research which is about farming". Here the emphasis is the application to farming of systems concepts and methodologies that have evolved over the past 50 years, mainly outside agriculture. This paradigm has been termed "systems agriculture" (1). Emphasis here is on approaches to learning/ research/ intervention when the system under study does not lend itself readily to scientific experimentation. Feasibility of the latter declines with increases in scale and/or, complexity and temporal variability. Two pools of methodological resources for addressing such systems are available—often termed "hard" and "soft" approaches. "Hard" systems approaches have, at their core, mathematical models of the systems of interest designed to represent the essential aspects of function in relation to environment. But the hard lesson in the main stream of the hard systems movement has been that the approach turns out to be appropriate only to those aspects of systems that are not complicated by people with purposes and freedom of choice (3). The fact that the specific nature of a farm system substantially reflects the design and management efforts of a farmer means that a "soft" systems approach, eg participative action research, should enhance the usefulness and impact of the research on real farming.     Farmers, advisers and researchers learning together better management of crops and croplands

    Summary. Farmers in the northeastern sub-tropics of Australia must cope with very high climatic variability in order to succeed in crop production. Their capacity for innovation was tapped by means of an on-farm research project that brought farmers, advisers and researchers together on the Darling Downs and in central Queensland. The researchers added value to the farmers' own experiments on fertility and water use efficiency by soil and weather monitoring at specific sites and then using a simulation model of cropping systems to extend findings to a wider context of climate and soil. The advisers extended knowledge aquired from this experience via local farmer networks and have undertaken training in the use of simulation to support farmers' management decisions. The experience described opens up possibilities for developing new, cost-effective ways for devising and testing improved farm management
