    Podoplanin expression in the development and progression of laryngeal squamous cell carcinomas

    <p>Abstract</p> <p>Background</p> <p>Podoplanin expression is attracting interest as a marker for cancer diagnosis and prognosis. We therefore investigated the expression pattern and clinical significance of podoplanin during the development and progression of laryngeal carcinomas.</p> <p>Results</p> <p>Podoplanin expression was determined by immunohistochemistry in paraffin-embedded tissue specimens from 84 patients with laryngeal premalignancies and 53 patients with laryngeal squamous cell carcinomas. We found podoplanin expression extending from the basal to the suprabasal layer of the epithelium in 37 (44%) of 84 dysplastic lesions, whereas normal epithelium showed negligible expression. Patients carrying podoplanin-positive lesions had a higher laryngeal cancer incidence than those with negative expression reaching borderline statistical significance (51% <it>versus </it>30%, <it>P </it>= 0.071). Podoplanin expression in laryngeal carcinomas exhibited two distinct patterns. 20 (38%) cases showed diffuse expression in most tumour cells and 33 (62%) focal expression at the proliferating periphery of tumour nests. High podoplanin expression was inversely correlated with T classification (<it>P </it>= 0.033), disease stage (<it>P </it>= 0.006), and pathological grade (<it>P </it>= 0.04). There was a trend, although not significant, towards reduced disease-specific survival for patients with low podoplanin levels (<it>P </it>= 0.31) and diffuse expression pattern (<it>P </it>= 0.08).</p> <p>Conclusions</p> <p>Podoplanin expression increases in the early stages of laryngeal tumourigenesis and it seems to be associated with a higher laryngeal cancer risk. Podoplanin expression in laryngeal squamous cell carcinomas, however, diminishes during tumour progression. Taken together, these data support a role for podoplanin expression in the initiation but not in the progression of laryngeal cancers.</p

    Deciphering the Potential Coding of Human Cytomegalovirus: New Predicted Transmembrane Proteome

    CMV is a major cause of morbidity and mortality in immunocompromised individuals that will benefit from the availability of a vaccine. Despite the efforts made during the last decade, no CMV vaccine is available. An ideal CMV vaccine should elicit a broad immune response against multiple viral antigens including proteins involved in virus-cell interaction and entry. However, the therapeutic use of neutralizing antibodies targeting glycoproteins involved in viral entry achieved only partial protection against infection. In this scenario, a better understanding of the CMV prote-ome potentially involved in viral entry may provide novel candidates to include in new potential vaccine design. In this study, we aimed to explore the CMV genome to identify proteins with puta-tive transmembrane domains to identify new potential viral envelope proteins. We have performed in silico analysis using the genome sequences of nine different CMV strains to predict the trans-membrane domains of the encoded proteins. We have identified 77 proteins with transmembrane domains, 39 of which were present in all the strains and were highly conserved. Among the core proteins, 17 of them such as UL10, UL139 or US33A have no ascribed function and may be good candidates for further mechanistic studies

    O III Congresso Ibero-Americano de Empreendedorismo, Energia, Ambiente e Tecnologia (CIEEMAT), dá continuidade ao trabalho desenvolvido no Brasil pelo Centro Federal de Educação Tecnológica Celso Suckow da Fonseca – Rio de Janeiro, atravessando o oceano Atlântico para a sua primeira edição no contexto da Península Ibérica. Concebido para englobar um conjunto abrangente de áreas temáticas, este evento serve de ponto de encontro entre profissionais, investigadores, professores e estudantes dos países Ibero-Americanos, num fórum que reforça o perfil internacional do Instituto Politécnico de Bragança, assim como das demais Instituições Organizadoras e Parceiras, designadamente, no âmbito de Programas de Dupla Diplomação. Nos três dias do evento, esperamos que encontrem oportunidades para a partilha de ideias e resultados de investigação, reforçando ou estabelecendo novos contactos e parcerias no espaço Latino-Americano.     Analysis of vibration impact on stability of dewetting thin liquid film

    Dynamics of a thin dewetting liquid film on a vertically oscillating substrate is considered. We assume moderate vibration frequency and large (compared to the mean film thickness) vibration amplitude. Using the lubrication approximation and the averaging method, we formulate the coupled sets of equations governing the pulsatile and the averaged fluid flows in the film, and then derive the nonlinear amplitude equation for the averaged film thickness. We show that there exists a window in the frequency-amplitude domain where the parametric and shear-flow instabilities of the pulsatile flow do not emerge. As a consequence, in this window the averaged description is reasonable and the amplitude equation holds. The linear and nonlinear analyses of the amplitude equation and the numerical computations show that such vibration stabilizes the film against dewetting and rupture.Comment: 19 pages, 11 figure
