    The Microbial Community of a Terrestrial Anoxic Inter-Tidal Zone: A Model for Laboratory-Based Studies of Potentially Habitable Ancient Lacustrine Systems on Mars

    Evidence indicates that Gale crater on Mars harboured a fluvio-lacustrine environment that was subjected to physio-chemical variations such as changes in redox conditions and evaporation with salinity changes, over time. Microbial communities from terrestrial environmental analogues sites are important for studying such potential habitability environments on early Mars, especially in laboratory-based simulation experiments. Traditionally, such studies have predominantly focused on microorganisms from extreme terrestrial environments. These are applicable to a range of Martian environments; however, they lack relevance to the lacustrine systems. In this study, we characterise an anoxic inter-tidal zone as a terrestrial analogue for the Gale crater lake system according to its chemical and physical properties, and its microbiological community. The sub-surface inter-tidal environment of the River Dee estuary, United Kingdom (53°21'015.40" N, 3°10'024.95" W) was selected and compared with available data from Early Hesperian-time Gale crater, and temperature, redox, and pH were similar. Compared to subsurface ‘groundwater’-type fluids invoked for the Gale subsurface, salinity was higher at the River Dee site, which are more comparable to increases in salinity that likely occurred as the Gale crater lake evolved. Similarities in clay abundance indicated similar access to, specifically, the bio-essential elements Mg, Fe and K. The River Dee microbial community consisted of taxa that were known to have members that could utilise chemolithoautotrophic and chemoorganoheterotrophic metabolism and such a mixed metabolic capability would potentially have been feasible on Mars. Microorganisms isolated from the site were able to grow under environment conditions that, based on mineralogical data, were similar to that of the Gale crater’s aqueous environment at Yellowknife Bay. Thus, the results from this study suggest that the microbial community from an anoxic inter-tidal zone is a plausible terrestrial analogue for studying habitability of fluvio-lacustrine systems on early Mars, using laboratory-based simulation experiments

    Designing avionics for lasers & optoelectronics

    Unlike imagery-based Earth observation (EO) which has become very widely and cheaply available, gravity sensing EO has not yet emerged from its fundamental science roots. The challenge therefore is to develop gravity sensing instruments that can replicate the success of widespread imagery based EO. There are three main gravity sensing mechanisms under investigation: laser ranging (e.g., GRACE-FO [1]); atom interferometers, which measure gravitation perturbations to the wavefunctions of individual atoms; and ‘relativistic geodesy’ which uses atomic clocks to measure the gravitational curvature of spacetime. All three of these measurement systems use stabilised lasers as their main enabling technology. However traditional laboratory laser systems struggle to meet the robustness, reliability, or low size, weight, and power (SWaP) requirements for use in space. A demonstrator was build that adapted telecommunications industry COTS components, and software radio FPGA/DSP techniques, to develop a new all-fibre space-qualified stabilised laser systems for geodesy that have equivalent performance to laboratory systems. This instrument was used to develop a 780 nm laser system that is stabilised to the Rubidium D2 line - the stabilised laser most commonly required by the quantum and atomic sensing field achieving sufficiently high laser performance for the laser system to be immediately useful for quantum applications (stability: 1-10 kHz, accuracy: 1 MHz); and in an ultra-compact package that has the potential to be used in space (1 litre, 0.5 kg, 10 W) [2]. This paper reports on the current student work that advances the instrument further towards a flight payload – and key avionics design considerations for future researchers. This takes lessons learnt from the ESA ESEO software radio payload in utilising ECSS design practices [3] to fabricate a robust and modular avionics back-end board that can operate with numerous front-end laser or opto-electronics configurations for different quantum applications. The new board consists of a single PCB containing circuitry for TT&C reporting of power supply and voltage conditioning, the current and temperature electronics needed to control a diode laser on orbit, interfaces for photo detectors and opto-electronics, and a high-speed analogue- to-digital conversion network centred around a FPGA. As an example, digital signal processing performed frequency-modulated spectroscopy on a warm Rubidium vapour using an all-fibre optical arrangement

    Forensic implications of the variation in morphology of marginal serrations on the teeth of the great white shark

    The teeth of the Great White Shark have been examined to ascertain whether there is any commonality in the arrangement or number of the marginal serrations (peaks) or, indeed, whether individual sharks have a unique pattern of shapes or size of the peaks. The teeth of the White Shark are characteristic in size and shape with serrations along almost the entire mesial and distal margins. This study has revealed no consistent pattern of size or arrangement of the marginal serrations that was sufficiently characteristic within an individual shark to serve as a reliable index of identification of a tooth as originating from that particular shark. Nonetheless, the serrations are sufficiently distinctive to enable the potential identification of an individual tooth as having been the cause of a particular bitemark.     Allometric relationships of the dentition of the great White Shark, Carcharodon carcharias, in forensic investigations of shark attacks

    As a result of a systematic morphometric study of shark dentitions, a system of notation for describing the location of shark teeth has been developed and is proposed as a standard to be adopted for use in similar studies in the future. The macroscopic morphology of White Shark teeth has been characterised in order to gain quantitative data which might assist in identification of these sharks from bite marks on victims or objects or from shark carcasses. Using these data, a nomogram has been developed which can be used to estimate the body length of a White Shark from measurements of tooth or bite mark morphology. An example of the forensic application of such allometric data is provided as it applied to a recent fatal attack on a diver by a White Shark.     Quantitative forensic evaluation of bite marks with the aid of a shape analysis computer program: Part 1; the development of 'SCIP' and the Similarity Index

    Bite marks left on human tissue and bitten material have become an important aspect of scientific evidence used for the conviction or acquittal of a suspect. Expert opinion has often been based on subjective comparisons rather than any objective metrical analysis and many experts will agree that there is a need to employ additional comparative tests to achieve unbiased objectivity in their investigation. In this study, an interactive shape analysis computer program ('SCIP' - Shape Comparison Interactive Program) has been employed in an attempt to derive experimentally a quantitative comparison, in the form of a Similarity Index (S.I.), between the 'offender's' teeth and the bite marks produced on a standard flat wax form. The S.I. values obtained using 'SCIP' were evaluated in a variety of experimental bite mark situations. It was found that in no case could the S.I. values produced by comparison of the bite mark with the dental casts from non-perpetrators be confused with the much lower S.I. from comparison of the bite mark with the dental cast of the perpetrator. The use of the Similarity Index derived using the 'SCIP' program is recommended as a simple, accurate and objective means of comparing bite marks in suitable forensic cases.     A Transcriptomal Analysis of Bovine Oviductal Epithelial Cells Collected During the Follicular Phase Versus the Luteal Phase of the Estrous Cycle

    BACKGROUND: Reproductive success depends on a functional oviduct for gamete storage, maturation, fertilization, and early embryonic development. The ovarian-derived steroids estrogen and progesterone are key regulators of oviductal function. The objective of this study was to investigate luteal and follicular phase-specific oviductal epithelial cell function by using microarray-based transcriptional profiling, to increase our understanding of mRNAs regulating epithelial cell processes, and to identify novel genes and biochemical pathways that may be found to affect fertility in the future. METHODS: Six normally cycling Angus heifers were assigned to either luteal phase (LP, n = 3) or follicular phase (FP, n = 3) treatment groups. Heifers in the LP group were killed between day 11 and 12 after estrus. Heifers in the FP group were treated with 25 mg PGF2α (Lutalyse, Pfizer, NY) at 8 pm on day 6 after estrus and killed 36 h later. Transcriptional profiling by microarray and confirmation of selected mRNAs by real-time RT-PCR analyses was performed using total RNA from epithelial cells isolated from sections of the ampulla and isthmus collected from LP and FP treatment groups. Differentially expressed genes were subjected to gene ontology classification and bioinformatic pathway analyses. RESULTS: Statistical one-way ANOVA using Benjamini-hochberg multiple testing correction for false discovery rate (FDR) and pairwise comparison of epithelial cells in the ampulla of FP versus LP groups revealed 972 and 597 transcripts up- and down-regulated, respectively (P \u3c 0.05). Within epithelial cells of the isthmus in FP versus LP groups, 946 and 817 transcripts were up- and down-regulated, respectively (P \u3c 0.05). Up-regulated genes from both ampulla and isthmus were found to be largely involved in cholesterol biosynthesis and cell cycle pathways, while down-regulated genes were found in numerous inflammatory response pathways. CONCLUSIONS: Microarray-based transcriptional profiling revealed phase of the cycle-dependent changes in the expression of mRNA within the epithelium of the oviducts\u27 ampulla and isthmus
