    Structural and functional analysis of ypt2, an essential ras-related gene in the fission yeast Schizosaccharomyces pombe encoding a Sec4 protein homologue.

    Using the cloned Saccharomyces cerevisiae YPT1 gene as hybridization probe, a gene, designated ypt2, was isolated from the fission yeast Schizosaccharomyces pombe and found to encode a 200 amino acid long protein most closely related to the ypt branch of the ras superfamily. Disruption of the ypt2 gene is lethal. The bacterially produced ypt2 gene product is shown to bind GTP. A region of the ypt2 protein corresponding to but different from the 'effector region' of ras proteins is also different from that of ypt1 proteins of different species but identical to the 'effector loop' of the S.cerevisiae SEC4 gene product, a protein known to be required for vesicular protein transport. The S.pombe ypt2 gene under control of the S.cerevisiae GAL10 promoter is able to suppress the temperature-sensitive phenotype of a S. cerevisiae sec4 mutant, indicating a functional similarity of these GTP-binding proteins from the two very distantly related yeasts

    Tripartite phase separation of two signal effectors with vesicles priming B cell responsiveness.

    Antibody-mediated immune responses rely on antigen recognition by the B cell antigen receptor (BCR) and the proper engagement of its intracellular signal effector proteins. Src homology (SH) 2 domain-containing leukocyte protein of 65 kDa (SLP65) is the key scaffold protein mediating BCR signaling. In resting B cells, SLP65 colocalizes with Cbl-interacting protein of 85 kDa (CIN85) in cytoplasmic granules whose formation is not fully understood. Here we show that effective B cell activation requires tripartite phase separation of SLP65, CIN85, and lipid vesicles into droplets via vesicle binding of SLP65 and promiscuous interactions between nine SH3 domains of the trimeric CIN85 and the proline-rich motifs (PRMs) of SLP65. Vesicles are clustered and the dynamical structure of SLP65 persists in the droplet phase in vitro. Our results demonstrate that phase separation driven by concerted transient interactions between scaffold proteins and vesicles is a cellular mechanism to concentrate and organize signal transducers

    A reduced-reference perceptual image and video quality metric based on edge preservation

    In image and video compression and transmission, it is important to rely on an objective image/video quality metric which accurately represents the subjective quality of processed images and video sequences. In some scenarios, it is also important to evaluate the quality of the received video sequence with minimal reference to the transmitted one. For instance, for quality improvement of video transmission through closed-loop optimisation, the video quality measure can be evaluated at the receiver and provided as feedback information to the system controller. The original image/video sequence-prior to compression and transmission-is not usually available at the receiver side, and it is important to rely at the receiver side on an objective video quality metric that does not need reference or needs minimal reference to the original video sequence. The observation that the human eye is very sensitive to edge and contour information of an image underpins the proposal of our reduced reference (RR) quality metric, which compares edge information between the distorted and the original image. Results highlight that the metric correlates well with subjective observations, also in comparison with commonly used full-reference metrics and with a state-of-the-art RR metric. © 2012 Martini et al

    Phylogeny of the Sepia pharaonis species complex (Cephalopoda: Sepiida) based on analyses of mitochondrial and nuclear DNA sequence data

    The pharaoh cuttlefish, Sepia pharaonis Ehrenberg, 1831, is a commercially fished species found from Japan to East Africa. Previous morphological and genetic work (the latter based on the 16S rRNA mitochondrial gene) suggested that S. pharaonis is a species complex, but relationships within the complex remained unresolved. To clarify these relationships, we have sequenced an additional mitochondrial gene region (cytochrome oxidase subunit I) and a nuclear gene region (rhodopsin) from over 50 specimens from throughout the range of S. pharaonis. We have also added sequence data from two specimens of Sepia ramani Neethiselvan, 2001, collected in southeastern India. Sepia ramani is a species that is morphologically very similar to S. pharaonis, and there is some question regarding its status as a distinct species. Phylogenetic analyses of a dataset comprising all three-gene regions revealed a monophyletic S. pharaonis complex consisting of a western Indian Ocean clade, a northeastern Australia clade, a Persian Gulf/Arabian Sea (‘Iranian’) clade, a western Pacific clade and a central Indian Ocean clade. Relationships among these clades remain somewhat poorly supported except for a clade comprising the Iranian clade, the western Pacific clade and the central Indian Ocean clade. One S. pharaonis specimen was collected in the Arabian Sea, but was found to be a member of the western Indian Ocean clade, suggesting that gene flow between these regions has either occurred recently or is ongoing. Both specimens of S. ramani are members of the S. pharaonis complex, but their mtDNA haplotypes are not closely related – one is a member of the central Indian Ocean clade, while the other is rather distantly related to the northeastern Australia clade. We suggest that ‘S. pharaonis’ may consist of several species, but morphological work is needed to clarify species-level taxonomy within this complex
