    Ageing of Natural Rubber under Stress

    We report a dynamical-mechanical study of stress relaxation at small deformation in a natural (polyisoprene) rubber well above its glass transition temperature Tg. We find that an almost complete relaxation of stress takes place over very long periods of time, even though the elastic network integrity is fully retained. The relaxation rate and the long-time equilibrium modulus are sensitive functions of temperature which do not follow time-temperature superposition. Many characteristic features of non-ergodic ageing response are apparent at both short and very long times. We interpret the observed behaviour in terms of the properties of rubber crosslinks, capable of isomerisation under stress, and relate the results to recent models of soft glassy rheology.Comment: Latex 2e (EPJ style), 5 EPS figure

    Morphology and stability of droplets sliding on soft viscoelastic substrates

    Soft solids such as gels and elastomers can be compliant enough to deform when a droplet lies on their surface, in particular at the line of contact between the solid, the liquid and the atmosphere. While axisymmetric contact line motion has received a lot of attention, much less is known about droplets moving on soft substrates, a configuration often encountered in applications in which symmetry may be lost. We investigate here the dynamic properties of droplets sliding on thick viscoelastic layers. We show that the partition of energy dissipation between the liquid and the solid sets the shape and velocity of droplets. When dissipation parts equally between the liquid and the solid, droplet dynamics are similar to that of droplets on rigid substrates. In the opposite case, we observe shapes that indicate the presence of contact angle hysteresis. We compare our observations to a non-linear model of the wetting of soft solids that we proposed recently. We find the model to be in excellent agreement with our data, in particular regarding the prediction of the hysteresis that we show to be only apparent. This work opens fondamental questions on the connection between the properties of the substrate and the dynamics, shapes and fragmentation of moving droplets that are relevant to all applications where soft gel coatings may be used.Comment: 19 pages, 7 figures, supplemental materials include

    SERPINB3 delays glomerulonephritis and attenuates the lupus-like disease in lupus murine models by inducing a more tolerogenic immune phenotype

    Objective: To explore the effects of SERPINB3 administration in murine lupus models with a focus on lupus-like nephritis. Methods: 40 NZB/W F1 mice were subdivided into 4 groups and intraperitoneally injected with recombinant SERPINB3 (7.5 \u3bcg/0.1 mL or 15 \u3bcg/0.1 mL) or PBS (0.1 mL) before (group 1 and 2) or after (group 3 and 4) the development of proteinuria ( 65100 mg/dl). Two additional mice groups were provided by including 20 MRL/lpr mice which were prophylactically injected with SERPINB3 (10 mice, group 5) or PBS (10 mice, group 6). Time of occurrence and levels of anti-dsDNA and anti-C1q antibodies, proteinuria and serum creatinine, overall- and proteinuria-free survival were assessed in mice followed up to natural death. Histological analysis was performed in kidneys of both lupus models. The Th17:Treg cell ratio was assessed by flow-cytometry in splenocytes of treated and untreated MRL/lpr mice. Statistical analysis was performed using non parametric tests and Kaplan-Meier curves, when indicated. Results: Autoantibody levels and proteinuria were significantly decreased and time of occurrence significantly delayed in SERPINB3-treated mice vs. controls. In agreement with these findings, proteinuria-free and overall survival were significantly improved in SERPINB3-treated groups vs. controls. Histological analysis demonstrated a lower prevalence of severe tubular lesions in kidneys of group 5 vs. group 6. SERPINB3-treated mice showed an overall trend toward a reduced prevalence of severe lesions in both strains. Th17:Treg ratio was significantly decreased in splenocytes of MRL/lpr mice treated with SERPINB3, compared to untreated control mice. Conclusions: SERPINB3 significantly improves disease course and delays the onset of severe glomerulonephritis in lupus-prone mice, possibly inducing a more tolerogenic immune phenotype

    The role of interferon regulatory factor five in systemic lupus erythematosus

    Systemic Lupus Erythematosus (SLE) is an autoimmune disorder associated with the development of immune complexes that deposit in multiple organ systems. The deposition of immune complexes is associated with tissue damage, most commonly seen in the form of kidney damage as lupus nephritis. IRF5 is an SLE susceptibility gene that is causal to type one interferon inflammatory cytokine production. We hypothesized that the peak luminescence of Donkey anti-mouse IgG Alexa-594 and for Goat anti-mouse C3-FITC in FcγRIIB-/- Yaa mice can show the disease state of SLE in murine models at which disease is most active. This thesis demonstrates that peak luminosity in FcγRIIB-/- Yaa mice is at 4.5 months of age. IHC staining was done to show the deposition of IgG and C3 protein in the glomeruli of FcγRIIB-/- Yaa at various ages. It also shows that the disease manifests between the ages of 2-4.5 months of age due to the differences in fluorescence intensity

    Dynamic mechanical response of polymer networks

    The dynamic-mechanical response of flexible polymer networks is studied in the framework of tube model, in the limit of small affine deformations, using the approach based on Rayleighian dissipation function. The dynamic complex modulus G* is calculated from the analysis of a network strand relaxation to the new equilibrium conformation around the distorted primitive path. Chain equilibration is achieved via a sliding motion of polymer segments along the tube, eliminating the inhomogeneity of the polymer density caused by the deformation. The characteristic relaxation time of this motion separates the low-frequency limit of the complex modulus from the high-frequency one, where the main role is played by chain entanglements, analogous to the rubber plateau in melts. The dependence of storage and loss moduli, G' and G'', on crosslink and entanglement densities gives an interpolation between polymer melts and crosslinked networks. We discuss the experimental implications of the rather short relaxation time and the slow square-root variation of the moduli and the loss factor tan at higher frequencies.Comment: Journal of Chemical Physics (Oct-2000); Lates, 4 EPS figures include

    Biodiversity and pollen feeding habits of syrphids in olive groves and surrounding landscape in Northeastern Portugal during spring

    Many syrphid larvae are predators of aphids and early stages of moths and psyllids, being potential control agents of some olive pests. However, adults need pollen and nectar for energy and reproduction. An increase of plant resources biodiversity in heterogeneous landscape could benefit these natural enemies. Thus, our goal of was to study the syrphid biodiversity and their pollen sources in olive agroecosystems from the northeast of Portugal during spring, that coincides with the availability of pests. For that, syrphids were captured in three not-tilled olive groves and two surrounding field areas (an herbaceous vegetation patch and a scrubland) next to each olive grove. Simultaneously, flowering plant inventories were carried out. Syrphids and the pollen contained in the guts of the most abundant species were identified. Thereafter pollen in guts was compared with pollen in the environment to evaluate a potential pollen selection. The most abundant species were Sphaerophoria scripta and Melanostoma mellinum. During the spring syrphids did not consume all the occurring plant species but did not actively selected pollen types. Results indicated that Asteraceae, Plantaginaceae, Caryophyllaceae, Echium type, Rumex type and Jasione type are important food sources for M. mellinum and S. scripta. These results will allow managing ground cover vegetation more efficiently in order to conserve syrphids in the olive agroecosystem.M.     EULAR recommendations for the management of systemic lupus erythematosus : 2023 update

    To update the EULAR recommendations for the management of systemic lupus erythematosus (SLE) based on emerging new evidence.An international Task Force formed the questions for the systematic literature reviews (January 2018-December 2022), followed by formulation and finalisation of the statements after a series of meetings. A predefined voting process was applied to each overarching principle and recommendation. Levels of evidence and strengths of recommendation were assigned, and participants finally provided their level of agreement with each item.The Task Force agreed on 5 overarching principles and 13 recommendations, concerning the use of hydroxychloroquine (HCQ), glucocorticoids (GC), immunosuppressive drugs (ISDs) (including methotrexate, mycophenolate, azathioprine, cyclophosphamide (CYC)), calcineurin inhibitors (CNIs, cyclosporine, tacrolimus, voclosporin) and biologics (belimumab, anifrolumab, rituximab). Advice is also provided on treatment strategies and targets of therapy, assessment of response, combination and sequential therapies, and tapering of therapy. HCQ is recommended for all patients with lupus at a target dose 5 mg/kg real body weight/day, considering the individual's risk for flares and retinal toxicity. GC are used as 'bridging therapy' during periods of disease activity; for maintenance treatment, they should be minimised to equal or less than 5 mg/day (prednisone equivalent) and, when possible, withdrawn. Prompt initiation of ISDs (methotrexate, azathioprine, mycophenolate) and/or biological agents (anifrolumab, belimumab) should be considered to control the disease and facilitate GC tapering/discontinuation. CYC and rituximab should be considered in organ-threatening and refractory disease, respectively. For active lupus nephritis, GC, mycophenolate or low-dose intravenous CYC are recommended as anchor drugs, and add-on therapy with belimumab or CNIs (voclosporin or tacrolimus) should be considered. Updated specific recommendations are also provided for cutaneous, neuropsychiatric and haematological disease, SLE-associated antiphospholipid syndrome, kidney protection, as well as preventative measures for infections, osteoporosis, cardiovascular disease.The updated recommendations provide consensus guidance on the management of SLE, combining evidence and expert opinion
