The fourth world : aboriginal women's activism and feminism

    Get PDF
The purpose of this thesis is to develop a framework for the discussion of Aboriginal women's oppression, one which will reflect Indigenous women's perspectives. It is suggested here that feminism often assumes that all women, cross-culturally, share the same oppression and that this assumption may be false. The writer believes that the Indigenous "Circle of Life" philosophy more appropriately embodies Aboriginal women's conceptions of human nature, their political philosophy, and their strategy for social change and liberation

Identifying low-Level sequence variants via next generation sequencing to aid stable CHO cell line screening

Developing stable Chinese hamster ovary (CHO) cell lines for biotherapeutics is an irreversible process and therefore, key quality attributes, such as sequence variants, must be closely monitored during cell line development (CLD) to avoid delay in the developmental timeline, and more importantly, to assure product safety and efficacy. Sequence variants, defined as unintended amino acid substitution in recombinant protein primary structure, result from alteration at either the DNA or the protein level. Here we report the application of transcriptome sequencing (RNAseq) in an IgG1 monoclonal antibody (mAb) CLD campaign to detect, identify and eliminate cell lines containing low-level point mutations in recombinant coding sequence. Among the top eleven mAb producers chosen from transfectant, clone or subclone stages, three of the cell lines contained either missense or nonsense point mutations at a low-level of less than 2%. Subsequent LC/MS/MS characterization detected ~3% sequence variants with an amino acid change from Ser to Leu at residue 117 in the heavy chain of transfectants 11 and 27. This substitution is consistent with the RNAseq finding of a C/T mutation located at 407 base pair (TCA→TTA) in the heavy chain coding sequence. Here, for the first time, we demonstrate that RNAseq is a rapid and highly sensitive method to identify low-level genetic mutation de novo corresponding to the amino acid substitution that elicits sequence variant(s). Its implementation in CLD constitutes an early and effective step in identifying desired CHO expression cell lines. As a continued effort to expedite the turnaround time and lower the developmental cost, we are further exploring other NGS approaches including cDNA (GOI) deep sequencing via Miseq and the preliminary data indicate a comparable sensitivity in detecting low-level point mutations

Apportioned Commodity Fetishism and the Transformative Power of Game Studies

    Get PDF
This chapter explores the ways in which the field of Game Studies helps shape popular understandings of player, play, and game, and specifically how the field alters the conceptual, linguistic, and discursive apparatuses that gamers use to contextualize, describe, and make sense of their experiences. The chapter deploys the concept of apportioned commodity fetishism to analyze the phenomena of discourse as practice, persona, and vagaries of game design, recursion, lexical formation, institutionalization, systems of self-effectiveness, theory as anti-theory, and commodification

YangMartin Luther King, Jr. is celebrated by most Americans today as a national hero, a person who gave his life leading his people to freedom. When Congress in 1983 established the third Monday of each January as a federal holiday in memory of King, he joined George Washington as the only individuals to be so honored. Since the holiday was first celebrated in 1986, tributes and festivities have annually taken place to remember his life and accomplishments

THE POWER OF COOL: CELEBRITY INFLUENCE IN THE IVORY TOWER

Since Donald Trump’s election in 2016, popular and scholarly commentators have been looking for speculative and/or dystopic literary works that might provide analogues for the Trump-era. Perhaps the most famous of these was the renewed popularity of Margaret Atwood’s The Handmaid’s Tale. In this regard, though, video games remain an underexplored fictional form. With its exaggerated and parodic satire of an America ruled by the corruption and greed of extreme right-wing populism, Grand Theft Auto (GTA): San Andreas (2004) offers a speculative fiction that players can enact as well as imagine, and simulate as well as prepare. Thus, reading the game through the lens of speculative fiction shows that GTA: San Andreas offers the kinds of intertexts, allusions, and parallels that Brabazon, Redhead, and Chivaura (2018) argue is essential for making sense of a dystopic present. Keywords: video games, game studies, popular culture, speculative fictio

Maine 4-H Afterschool Academy—A Professional Development Opportunity for Out-of-School-Time Providers

    Get PDF
The Maine 4-H Afterschool Academy trained 369 after-school and out of school time providers in 2011. This easy-to-adapt professional development opportunity used blended learning, a combination of in-person and Web-based opportunities. Providers successfully learned concepts and practical knowledge regarding 4-H, specifically 4-H Science. In post-training evaluations, 86% of participants reported feeling more confident in incorporating science, engineering, and technology in their afterschool program. This blended learning approach eliminated some barriers Extension faculty face when designing programing for afterschool providers
