    Statistically Locked-in Transport Through Periodic Potential Landscapes

    Classical particles driven through periodically modulated potential energy landscapes are predicted to follow a Devil's staircase hierarchy of commensurate trajectories depending on the orientation of the driving force. Recent experiments on colloidal spheres flowing through arrays of optical traps do indeed reveal such a hierarchy,but not with the predicted structure. The microscopic trajectories, moreover,appear to be random, with commensurability emerging only in a statistical sense. We introduce an idealized model for periodically modulated transport in the presence of randomness that captures both the structure and statistics of such statistically locked-in states.Comment: REVTeX with EPS figures, 4 pages, 4 figure

    Circuit for Full Charging of Series Lithium-Ion Cells

    An advanced charger has been proposed for a battery that comprises several lithium-ion cells in series. The proposal is directed toward charging the cells in as nearly an optimum manner as possible despite unit-to-unit differences among the nominally identical cells. The particular aspect of the charging problem that motivated the proposal can be summarized as follows: During bulk charging (charging all the cells in series at the same current), the voltages of individual cells increase at different rates. Once one of the cells reaches full charge, bulk charging must be stopped, leaving other cells less than fully charged. To make it possible to bring all cells up to full charge once bulk charging has been completed, the proposed charger would include a number of top-off chargers one for each cell. The top-off chargers would all be powered from the same DC source, but their outputs would be DC-isolated from each other and AC-coupled to their respective cells by means of transformers, as described below. Each top-off charger would include a flyback transformer, an electronic switch, and an output diode. For suppression of undesired electromagnetic emissions, each top-off charger would also include (1) a resistor and capacitor configured to act as a snubber and (2) an inductor and capacitor configured as a filter. The magnetic characteristics of the flyback transformer and the duration of its output pulses determine the energy delivered to the lithium-ion cell. It would be necessary to equip the cell with a precise voltage monitor to determine when the cell reaches full charge. In response to a full-charge reading by this voltage monitor, the electronic switch would be held in the off state. Other cells would continue to be charged similarly by their top-off chargers until their voltage monitors read full charge

    CD45 immunoaffinity depletion of vesicles from Jurkat T cells demonstrates that exosomes contain CD45: no evidence for a distinct exosome/HIV-1 budding pathway

    The presence of relatively high levels of cellular protein contamination in density-purified virion preparations is a confounding factor in biochemical analyses of HIV and SIV produced from hematopoietic cells. A major source of this contamination is from vesicles, either microvesicles or exosomes, that have similar physical properties as virions. Thus, these particles can not be removed by size or density fractionation. Although virions and vesicles have similar cellular protein compositions, CD45 is excluded from HIV-1 yet is present in vesicles produced from hematopoietic cells. By exploiting this finding, we have developed a CD45 immunoaffinity depletion procedure that removes vesicles from HIV-1 preparations. While this approach has been successfully applied to virion preparations from several different cell types, some groups have concluded that "exosomes" from certain T cell lines, specifically Jurkat, do not contain CD45. If this interpretation is correct, then these vesicles could not be removed by CD45 immunoaffinity depletion. Here we show that dense vesicles produced by Jurkat and SupT1/CCR5 cells contain CD45 and are efficiently removed from preparations by CD45-immunoaffinity depletion. Also, contaminating cellular proteins were removed from virion preparations produced by these lines. Previously, the absence of CD45 from both "exosomes" and virions has been used to support the so called Trojan exosome hypothesis, namely that HIV-1 is simply an exosome containing viral material. The presence of CD45 on vesicles, including exosomes, and its absence on virions argues against a specialized budding pathway that is shared by both exosomes and HIV-1

    High-Accuracy Measurements of Total Column Water Vapor From the Orbiting Carbon Observatory-2

    Accurate knowledge of the distribution of water vapor in Earth's atmosphere is of critical importance to both weather and climate studies. Here we report on measurements of total column water vapor (TCWV) from hyperspectral observations of near-infrared reflected sunlight over land and ocean surfaces from the Orbiting Carbon Observatory-2 (OCO-2). These measurements are an ancillary product of the retrieval algorithm used to measure atmospheric carbon dioxide concentrations, with information coming from three highly resolved spectral bands. Comparisons to high-accuracy validation data, including ground-based GPS and microwave radiometer data, demonstrate that OCO-2 TCWV measurements have maximum root-mean-square deviations of 0.9-1.3mm. Our results indicate that OCO-2 is the first space-based sensor to accurately and precisely measure the two most important greenhouse gases, water vapor and carbon dioxide, at high spatial resolution [1.3 x 2.3 km(exp. 2)] and that OCO-2 TCWV measurements may be useful in improving numerical weather predictions and reanalysis products

    Ubiquitination of HIV-1 and MuLV Gag

    AbstractOur previous biochemical studies of HIV-1 and MuLV virions isolated and identified mature Gag products, HIV-1 p6Gag and MuLV p12Gag, that were conjugated to a single ubiquitin. To study the importance of the monoubiquitination of Gag, a series of lysine to arginine mutants were constructed that eliminated ubiquitination at one or both of the lysines in HIV-1NL4-3 p6Gag and both lysines in Moloney MuLV p12Gag. HPLC and immunoblot analysis of the HIV-1 mutants demonstrated that either of the lysines in p6Gag, K27 or K33, could be monoubiquitinated. However, infectivity assays showed that monoubiquitination of HIV-1 p6Gag or MuLV p12Gag is not required for viral replication in vitro. Pulse-chase radiolabeling of HIV-1-producing cells revealed that monoubiquitination of p6Gag does not affect the short-term release of virus from the cell, the maturation of Pr55Gag, or the sensitivity of these processes to proteasome inhibitors. Experiments with protease-deficient HIV-1 showed that Pr55Gag can be monoubiquitinated, suggesting that p6Gag is first modified as a domain within Gag. Examination of the proteins inside an HIV-1 mutant found that free ubiquitin was incorporated into the virions in the absence of the lysines in p6Gag, showing that the ubiquitin inside the virus is not initially brought in as a p6Gag conjugate. Although our results establish that monoubiquitination of p6Gag and p12Gag is not required for viral replication in vitro, this modification may be a by-product of interactions between Gag and cellular proteins during assembly and budding

    Diffuse HI Disks in Isolated Galaxies

    In order to investigate the contribution of diffuse components to their total HI emission, we have obtained high precision HI line flux densities with the 100m Green Bank Telescope for a sample of 100 isolated spiral and irregular galaxies which we have previously observed with the 43m telescope. A comparison of the observed HI line fluxes obtained with the two different telescopes, characterized by half-power beam widths of 9 arcmin and 21 arcmin respectively, exploits a ``beam matching'' technique to yield a statistical determination of the occurrence of diffuse HI components in their disks. A simple model of the HI distribution within a galaxy well describes ~75 % of the sample and accounts for all of the HI line flux density. The remaining galaxies are approximately evenly divided into two categories: ones which appear to possess a significantly more extensive HI distribution than the model predicts, and ones for which the HI distribution is more centrally concentrated than predicted. Examples of both extremes can be found in the literature but little attention has been paid to the centrally concentrated HI systems. Our sample has demonstrated that galaxies do not commonly possess extended regions of low surface brightness HI gas which is not accounted for by our current understanding of the structure of HI disks. Eight HI-rich companions to the target objects are identified, and a set of extragalactic HI line flux density calibrators is presented.Comment: 26 page

    Metabolic reprogramming of human CD8+ memory T cells through loss of SIRT1.

    The expansion of CD8+CD28- T cells, a population of terminally differentiated memory T cells, is one of the most consistent immunological changes in humans during aging. CD8+CD28- T cells are highly cytotoxic, and their frequency is linked to many age-related diseases. As they do not accumulate in mice, many of the molecular mechanisms regulating their fate and function remain unclear. In this paper, we find that human CD8+CD28- T cells, under resting conditions, have an enhanced capacity to use glycolysis, a function linked to decreased expression of the NAD+-dependent protein deacetylase SIRT1. Global gene expression profiling identified the transcription factor FoxO1 as a SIRT1 target involved in transcriptional reprogramming of CD8+CD28- T cells. FoxO1 is proteasomally degraded in SIRT1-deficient CD8+CD28- T cells, and inhibiting its activity in resting CD8+CD28+ T cells enhanced glycolytic capacity and granzyme B production as in CD8+CD28- T cells. These data identify the evolutionarily conserved SIRT1-FoxO1 axis as a regulator of resting CD8+ memory T cell metabolism and activity in humans

    Bifurcations of periodic and chaotic attractors in pinball billiards with focusing boundaries

    We study the dynamics of billiard models with a modified collision rule: the outgoing angle from a collision is a uniform contraction, by a factor lambda, of the incident angle. These pinball billiards interpolate between a one-dimensional map when lambda=0 and the classical Hamiltonian case of elastic collisions when lambda=1. For all lambda<1, the dynamics is dissipative, and thus gives rise to attractors, which may be periodic or chaotic. Motivated by recent rigorous results of Markarian, Pujals and Sambarino, we numerically investigate and characterise the bifurcations of the resulting attractors as the contraction parameter is varied. Some billiards exhibit only periodic attractors, some only chaotic attractors, and others have coexistence of the two types.Comment: 30 pages, 17 figures. v2: Minor changes after referee comments.     Evidence of a Cloud-Cloud Collision from Overshooting Gas in the Galactic Center

    The Milky Way is a barred spiral galaxy with "bar lanes" that bring gas towards the Galactic Center. Gas flowing along these bar lanes often overshoots, and instead of accreting onto the Central Molecular Zone, it collides with the bar lane on the opposite side of the Galaxy. We observed G5, a cloud which we believe is the site of one such collision, near the Galactic Center at (l,b) = (+5.4, -0.4) with the ALMA/ACA. We took measurements of the spectral lines 12^{12}CO J=2-1, 13^{13}CO J=2-1, C18^{18}O J=2-1, H2_2CO J=303_{03}-202_{02}, H2_{2}CO J=322_{22}-221_{21}, CH3_{3}OH J=422_{22}-312_{12}, OCS J=18-17 and SiO J=5-4. We observed a velocity bridge between two clouds at ∼\sim50 km/s and ∼\sim150 km/sin our position-velocity diagram, which is direct evidence of a cloud-cloud collision. We measured an average gas temperature of ∼\sim60 K in G5 using H2_2CO integrated intensity line ratios. We observed that the 12^{12}C/13^{13}C ratio in G5 is consistent with optically thin, or at most marginally optically thick 12^{12}CO. We measured 1.5 x 109^9 cmβˆ’2^{-2}(K km/s)βˆ’1^{-1} for the local XCO_{CO}, 10-20x less than the average Galactic value. G5 is strong direct observational evidence of gas overshooting the Central Molecular Zone (CMZ) and colliding with a bar lane on the opposite side of the Galactic center.Comment: Accepted to ApJ. 27 pages, 19 figure

    Real-Time Visualization of HIV-1 GAG Trafficking in Infected Macrophages

    HIV-1 particle production is driven by the Gag precursor protein Pr55Gag. Despite significant progress in defining both the viral and cellular determinants of HIV-1 assembly and release, the trafficking pathway used by Gag to reach its site of assembly in the infected cell remains to be elucidated. The Gag trafficking itinerary in primary monocyte-derived macrophages is especially poorly understood. To define the site of assembly and characterize the Gag trafficking pathway in this physiologically relevant cell type, we have made use of the biarsenical-tetracysteine system. A small tetracysteine tag was introduced near the C-terminus of the matrix domain of Gag. The insertion of the tag at this position did not interfere with Gag trafficking, virus assembly or release, particle infectivity, or the kinetics of virus replication. By using this in vivo detection system to visualize Gag trafficking in living macrophages, Gag was observed to accumulate both at the plasma membrane and in an apparently internal compartment that bears markers characteristic of late endosomes or multivesicular bodies. Significantly, the internal Gag rapidly translocated to the junction between the infected macrophages and uninfected T cells following macrophage/T-cell synapse formation. These data indicate that a population of Gag in infected macrophages remains sequestered internally and is presented to uninfected target cells at a virological synapse
