    Interactions between the endocrine and immune systems in locusts

    The prophenoloxidase cascade in the haemolymph of mature adult Locusta migratoria migratorioides (R & F) is activated in response to injection of laminarin, a -1,3 glucan. Co-injection of adipokinetic hormone-I (Lom-AKH-I) and laminarin prolongs the activation of the enzyme in a dose-dependent manner. However, injections of bacterial lipopolysaccharide (LPS) do not activate prophenoloxidase unless AKH is co-injected, when there is a dose-dependent increase in the level of phenoloxidase that persists in the haemolymph for several hours. Even when AKH is co-injected, the highest levels of phenoloxidase activity are always greater after injection of laminarin than after LPS, and these two immunogens must activate the prophenoloxidase cascade by quite distinct pathways. In the present study, interactions between the endocrine and immune systems were examined with respect to activation of prophenoloxidase and the formation of nodules: injection of LPS induces nodule formation in adult locusts. With LPS from Pseudomonas aeruginosa, nodules form exclusively in dense accumulations in the anterior portion of the abdomen on either side of the dorsal blood vessel associated with the dorsal diaphragm. However, with LPS from Escherichia coli, fewer nodules are formed but with a similar distribution, except that occasionally some nodules are aligned additionally on either side of the ventral nerve cord. Co-injection of Lom-AKH-I with LPS from either bacteria stimulates greater numbers of nodules to be formed. This effect of coinjection of AKH on nodule formation is seen at low doses of hormone with only 0.3 or 0.4 pmol of Lom-AKH-1, respectively, increasing the number of nodules by 50%. Injections of octopamine or 5-hydroxytryptamine do not mimic either of the actions of Lom-AKH-I described here. Co-injection of an angiotensin-converting enzyme inhibitor, captopril, reduces nodule formation in response to injections of LPS but has no effect on the activation of phenoloxidase. Co-injection of an inhibitor of eicosanoid synthesis, dexamethasone, with LPS influences nodule formation (with or without AKH) in different ways according to the dose of dexamethasone used, but does not affect activation of prophenoloxidase. Eicosanoid synthesis is important for nodule formation, but not for the activation of the prophenoloxidase cascade in locust haemolymph

    Discrete approximations to vector spin models

    We strengthen a result of two of us on the existence of effective interactions for discretised continuous-spin models. We also point out that such an interaction cannot exist at very low temperatures. Moreover, we compare two ways of discretising continuous-spin models, and show that, except for very low temperatures, they behave similarly in two dimensions. We also discuss some possibilities in higher dimensions.Comment: 12 page

    Flow Assurance in Subsea Pipeline Design - A Case Study of Ghana’s Jubilee and TEN Fields

    The increasing exploration and production activities in the offshore Cape Three Point Blocks of Ghana have led to the discovery and development of gas condensate fields in addition to the oil fields which produce significant amount of condensate gas. These discoveries require pipelines to transport the fluids avoiding hydrates and wax formation. This paper focuses on subsea pipeline design using Pipesim software that addresses flow assurance problems associated with transporting condensate gas from the Jubilee and TEN Fields to the Atuabo Gas Processing Plant. It also considered an alternate design that eliminates the need for capacity increase of flowlines for the futuristic highest projected flow rates in 2030. The design comprises of two risers and two flowlines. Hydrate formation temperature was determined to be 72.5 ˚F at a pressure of 3 000 psig. The insulation thickness for flowlines 1 and 2 were determined to be 1.5 in. and 2 in. respectively. The pipe size for flowlines 1 and 2 were determined to be 12 in. and 14 in. respectively. The maximum designed flow rate was determined to be 150 MMSCFD. To meet the highest projected flow rate of 700 MMSCFD in the year 2030 at the processing plant, a 16 in. ID pipeline of 44 km length was placed parallel to the 12 in. ID flowline 1. This parallel pipeline increased the designed flow rate by approximately 4.7 times (705 MMSCFD).     Occurrence of entomopathogenic nematodes (Steinernematidae, heterorhabditidae) as potential biocontrol agents against Spodoptera frugiperda infesting Zea mays

    Synthetic insecticides are used worldwide to manage invasive fall armyworm (Spodoptera frugiperda) in cereals, in spite of the chemical’s prohibitive cost and threats to environmental health. In Ghana, little attention has been given to entomopathogenic nematodes as insect pest biocontrol agents, due to the existence of dismal expertise in the field. This study evaluated maize (Zea mays L.) farms for endemic entomopathogenic nematodes as potential biological control agents against fall armyworms infesting maize in Ghana. Entomopathogenic nematode juveniles were extracted using an insect baiting technique, White traps. These Rhabditid nematodes (Heterorhabditis; Steinernema) were found in 75% of 200 soil samples; and 92.5% of 40 districts targeted by this study. The Greater Accra region recorded the greatest population density (1,820 juveniles per 5 fall armyworm larvae cadavers); while; the Central region recorded the lowest density (81 juveniles per 5 fall armyworm larvae cadavers). There was high presence of entomopathogenic nematodes across the study areas. Therefore, entomopathogenic nematodes offer a promising alternative strategy for managing fall armyworm infestation of maize in Ghana to minimise over-reliance on synthetic insecticides. &nbsp

    Decentralized Solid Waste Management in Rural Ghana: A Case Study of Assin Kushea Community in Assin North Municipality

    Management of solid waste continues to be a major developmental challenge for developing countries like Ghana. The current focus and attention have been on the collection and dumping of waste in urban communities where generation rate is high to the detriment of rural communities. In this study, a pilot waste management scheme was undertaken in Assin Kushea, a rural community in the Assin North Municipality, to determine the willingness of the community to sort their waste at source, the quantity of waste generated by the community, and the characteristics of the waste to inform the treatment or disposal options suitable for the community. The results of the study showed that the community generates approximately 20 to 40 kg waste per day which comprises about 77% biological municipal waste (BMW) and 23% of residual waste. Laboratory analysis of the waste showed that the moisture content of the waste was about 68%. The percentage volatile solid was about 85.45%, leaving an ash content of approximately 14.55% all by weight of the waste materials. More than 50% of the sampled population achieved 100% source separation efficiency. Given the composition and characteristics of the waste, and the willingness of the community to sort their waste at source, composting or anaerobic fermentation of the organic waste fraction is recommended as the best waste treatment option for the organic component of the waste for the community

    The Embryonic Transcriptome Of The Red-Eared Slider Turtle (Trachemys Scripta)

    The bony shell of the turtle is an evolutionary novelty not found in any other group of animals, however, research into its formation has suggested that it has evolved through modification of conserved developmental mechanisms. Although these mechanisms have been extensively characterized in model organisms, the tools for characterizing them in non-model organisms such as turtles have been limited by a lack of genomic resources. We have used a next generation sequencing approach to generate and assemble a transcriptome from stage 14 and 17 Trachemys scripta embryos, stages during which important events in shell development are known to take place. The transcriptome consists of 231,876 sequences with an N-50 of 1,166 bp. GO terms and EC codes were assigned to the 61,643 unique predicted proteins identified in the transcriptome sequences. All major GO categories and metabolic pathways are represented in the transcriptome. Transcriptome sequences were used to amplify several cDNA fragments designed for use as RNA in situ probes. One of these, BMP5, was hybridized to a T. scripta embryo and exhibits both conserved and novel expression patterns. The transcriptome sequences should be of broad use for understanding the evolution and development of the turtle shell and for annotating any future T. scripta genome sequences

    Repellency Potential, Chemical Constituents of Ocimum Plant Essential Oils, and Their Headspace Volatiles against Anopheles gambiae s. s., Malaria Vector

    African malaria mosquitoes (Anopheles gambiae sensu stricto) transmit a malaria parasite (Plasmodium falciparum) to humans. The current control strategies for the vector have mainly focussed on synthetic products, which negatively impact the environment and human health. Given the potential use of environmentally friendly plant-derived volatiles as a control, this work aims to examine and compare the repellency potential of essential oils and headspace volatiles from Ocimum gratissimum, Ocimum tenuiflorum, and Ocimum basilicum and their chemical compositions. The repellency potential and chemical composition of the plants were achieved by using the protected arm-in-cage method and gas chromatography-mass spectrometry (GC-MS) analysis. Among the three Ocimum species, both the essential oils and the headspace volatiles from O. tenuiflorum achieved the longest repellency time lengths of 90–120 minutes. One hundred and one (101) chemical constituents were identified in the headspace volatiles of the three Ocimum spp. Nonetheless, (−)-camphor, (E)-γ-bisabolene, terpinolene, β-chamigrene, cubedol, (E)-farnesol, germacrene D-4-ol, viridiflorol, γ-eudesmol, tetracyclo [6.3.2.0 (2,5).0(1,8)] tridecan-9-ol, 4,4-dimethyl, α-eudesmol, isolongifolol, and endo-borneol were unique only to O. tenuiflorum headspace volatiles. Either essential oils or headspace volatiles from O. tenuiflorum could offer longer protection time length to humans against An. gambiae. Though field studies are needed to assess the complementarity between the chemical constituents in the headspace volatiles of O. tenuiflorum, our observations provide a foundation for developing effective repellents against An. gambiae
