Voice and rural wireless mesh community networks: a framework to quantify scalability and manage end-user smartphone battery consumption
Philosophiae Doctor - PhDCommunity wireless mesh initiatives are a pioneering option to cheap βlast-mileβ access to network services for rural low-income regions primarily located in Sub-Saharan Africa and Developing Asia. However, researchers have criticized wireless mesh networks for their poor scalability; and scalability quantification research has mostly consisted of modularization of per-node throughput capacity behaviour. A scalability quantification model to design wireless mesh networks to provide adequate quality of service is lacking. However, scalability quantification of community mesh networks alone is inadequate because rural users need affordable devices for access; and they need to know how best to use them. Low-cost low-end smartphones offer handset affordability solutions but require smart management of their small capacity battery. Related work supports the usage of Wi-Fi for communication because it is shown to consume less battery than 2G, 3G or Bluetooth. However, a model to compare Wi-Fi battery consumption amongst different low-end smartphones is missing, as is a comparison of different over-the-top communication applications
Towards a scalability model for wireless mesh networks
Zenzeleni mesh network is a wireless ad-hoc mesh network that provides voice services using public analogue telephones to the Mankosi community in the Eastern Cape
Province. We would like to improve on the network infrastructure by upgrading the mesh routers and introducing low-end smartphones onto the network; and offer both data and voice over Internet protocol services. However, before deploying resources, it is imperative to identify the maximum number of mesh nodes, clients and simultaneous voice over internet protocol calls that can be supported by the mesh network while maintaining acceptable quality of service levels. Absence of such data might lead to financial risk and time depletion when setting up an optimal network. Bolstering the claim are investigations that report drop in quality levels as network density and hop count escalate. As current investigations mostly yield capacity models to predict per-node throughput with increasing hop count, we propose experiments to devise a scalability model to quantify scalability of mesh networks in this paper. Battery and data drain of over-the-top applications on low-end smartphones
Low-end smartphones with sub $50 price tags provide affordable device ownership to low-income populations. However, their limited capacity, when combined with the need for multimodal connectivity, raises usage concerns in rural off-grid regions. Some off-grid regions in sub-Saharan Africa provide recharge facilities using solar power and charge money for the service. Adding data bundle costs to frequent recharge costs, affordability of low-end smartphones becomes questionable in such areas. Community-controlled solar-powered wireless mesh network models with Session Initiation Protocol capability could alleviate the network usage cost conundrum and consume less power in low-end smartphones with the usage of WiFi. This paper reports on investigations that reveal usage of WiFi consumes less battery than 3G, 2G and Bluetooth. In addition, we feel that lowering recharge costs also requires battery consumption knowledge of the over-the-top applications. Using automated voice calls, this paper reports on battery and data consumption by multiple popular social media applications using one type of low-end smartphone. Data consumption was calculated with the objective of learning how to lower data bundle costs by selecting the application with least data consumption. Battery consumption due to CPU usage by the applications was also measured. Results show that WhatsApp consumes the least battery amongst instant messengers and also the least data over all apps measured. SipDroid consumes the least battery overall. Additionally, the reported experiments provide a framework for future experiments aimed at evaluating battery and data consumption by other smartphone applications.NRF, Openserve/Aria Centre of Excellenc
Evaluating energy consumption on low-end smartphones
The relationship between battery consumption in
smartphones and the usage statistics of a phone is direct. Modern
smartphones, even low-end, are equipped with multiple wireless
technologies, e.g. GSM, 3G, WiFi and Bluetooth. Each of these
technologies has a different energy consumption profile. A
wireless mesh project in the Mankosi community in rural South
Africa is about to introduce low-end smartphones onto the
network. The mesh network is powered with solar-charged
batteries because the community at present does not have
electricity. Local residents also use these batteries to recharge cell
phones at a nominal cost. Introduction of smartphones will
increase the recharge frequency as phone usage will increase;
thus draining a phone battery more quickly, as well as escalate
recharge costs. Thus, the smartphones must be chosen and used
effectively in order for batteries to last longer. Related work
identifies WiFi wireless technology as the most battery efficient
way of transfer when compared to GSM, 3G and Bluetooth. This
research proposes experiments to further investigate energy
efficiency of WiFi in low-end smartphones that we intend to use
for local and breakout voice over Internet protocol (VoIP) calls
and data services, on a rural wireless mesh network
Extra-Renal Elimination of Uric Acid via Intestinal Efflux Transporter BCRP/ABCG2
Urinary excretion accounts for two-thirds of total elimination of uric acid and the remainder is excreted in feces. However, the mechanism of extra-renal elimination is poorly understood. In the present study, we aimed to clarify the mechanism and the extent of elimination of uric acid through liver and intestine using oxonate-treated rats and Caco-2 cells as a model of human intestinal epithelium. In oxonate-treated rats, significant amounts of externally administered and endogenous uric acid were recovered in the intestinal lumen, while biliary excretion was minimal. Accordingly, direct intestinal secretion was thought to be a substantial contributor to extra-renal elimination of uric acid. Since human efflux transporter BCRP/ABCG2 accepts uric acid as a substrate and genetic polymorphism causing a decrease of BCRP activity is known to be associated with hyperuricemia and gout, the contribution of rBcrp to intestinal secretion was examined. rBcrp was confirmed to transport uric acid in a membrane vesicle study, and intestinal regional differences of expression of rBcrp mRNA were well correlated with uric acid secretory activity into the intestinal lumen. Bcrp1 knockout mice exhibited significantly decreased intestinal secretion and an increased plasma concentration of uric acid. Furthermore, a Bcrp inhibitor, elacridar, caused a decrease of intestinal secretion of uric acid. In Caco-2 cells, uric acid showed a polarized flux from the basolateral to apical side, and this flux was almost abolished in the presence of elacridar. These results demonstrate that BCRP contributes at least in part to the intestinal excretion of uric acid as extra-renal elimination pathway in humans and rats
A Common Role for Various Human Truncated Adenomatous Polyposis Coli Isoforms in the Control of Beta-Catenin Activity and Cell Proliferation
The tumour suppressor gene adenomatous polyposis coli (APC) is mutated in most colorectal cancer cases, leading to the synthesis of truncated APC products and the stabilization of Ξ²-catenin. Truncated APC is almost always retained in tumour cells, suggesting that it serves an essential function. Here, RNA interference has been used to down-regulate truncated APC in several colorectal cancer cell lines expressing truncated APCs of different lengths, thereby performing an analysis covering most of the mutation cluster region (MCR). The consequences on proliferation in vitro, tumour formation in vivo and the level and transcriptional activity of Ξ²-catenin have been investigated. Down-regulation of truncated APC results in an inhibition of tumour cell population expansion in vitro in 6 cell lines out of 6 and inhibition of tumour outgrowth in vivo as analysed in one of these cell lines, HT29. This provides a general rule explaining the retention of truncated APC in colorectal tumours and defines it as a suitable target for therapeutic intervention. Actually, we also show that it is possible to design a shRNA that targets a specific truncated isoform of APC without altering the expression of wild-type APC. Down-regulation of truncated APC is accompanied by an up-regulation of the transcriptional activity of Ξ²-catenin in 5 out of 6 cell lines. Surprisingly, the increased signalling is associated in most cases (4 out of 5) with an up-regulation of Ξ²-catenin levels, indicating that truncated APC can still modulate wnt signalling through controlling the level of Ξ²-catenin. This control can happen even when truncated APC lacks the Ξ²-catenin inhibiting domain (CiD) involved in targeting Ξ²-catenin for proteasomal degradation. Thus, truncated APC is an essential component of colorectal cancer cells, required for cell proliferation, possibly by adjusting Ξ²-catenin signalling to the βjust rightβ level
Roles of Major Facilitator Superfamily Transporters in Phosphate Response in Drosophila
The major facilitator superfamily (MFS) transporter Pho84 and the type III transporter Pho89 are responsible for metabolic effects of inorganic phosphate in yeast. While the Pho89 ortholog Pit1 was also shown to be involved in phosphate-activated MAPK in mammalian cells, it is currently unknown, whether orthologs of Pho84 have a role in phosphate-sensing in metazoan species. We show here that the activation of MAPK by phosphate observed in mammals is conserved in Drosophila cells, and used this assay to characterize the roles of putative phosphate transporters. Surprisingly, while we found that RNAi-mediated knockdown of the fly Pho89 ortholog dPit had little effect on the activation of MAPK in Drosophila S2R+ cells by phosphate, two Pho84/SLC17A1β9 MFS orthologs (MFS10 and MFS13) specifically inhibited this response. Further, using a Xenopus oocyte assay, we show that MSF13 mediates uptake of [Β³Β³P]-orthophosphate in a sodium-dependent fashion. Consistent with a role in phosphate physiology, MSF13 is expressed highest in the Drosophila crop, midgut, Malpighian tubule, and hindgut. Altogether, our findings provide the first evidence that Pho84 orthologs mediate cellular effects of phosphate in metazoan cells. Transient Ureteral Obstruction Prevents against Kidney Ischemia/Reperfusion Injury via Hypoxia-Inducible Factor (HIF)-2Ξ± Activation
Although the protective effect of transient ureteral obstruction (UO) prior to ischemia on subsequent renal ischemia/reperfusion (I/R) injury has been documented, the underlying molecular mechanism remains to be understood. We showed in the current study that 24 h of UO led to renal tubular hypoxia in the ipsilateral kidney in mice, with the accumulation of hypoxia-inducible factor (HIF)-2Ξ±, which lasted for a week after the release of UO. To address the functions of HIF-2Ξ± in UO-mediated protection of renal IRI, we utilized the Mx-Cre/loxP recombination system to knock out target genes. Inactivation of HIF-2Ξ±, but not HIF-1Ξ± blunted the renal protective effects of UO, as demonstrated by much higher serum creatinine level and severer histological damage. UO failed to prevent postischemic neutrophil infiltration and apoptosis induction in HIF-2Ξ± knockout mice, which also diminished the postobstructive up-regulation of the protective molecule, heat shock protein (HSP)-27. The renal protective effects of UO were associated with the improvement of the postischemic recovery of intra-renal microvascular blood flow, which was also dependent on the activation of HIF-2Ξ±. Our results demonstrated that UO protected the kidney via activation of HIF-2Ξ±, which reduced tubular damages via preservation of adequate renal microvascular perfusion after ischemia. Thus, preconditional HIF-2Ξ± activation might serve as a novel therapeutic strategy for the treatment of ischemic acute renal failure
Investigation of a dual-band dual radio indoor mesh testbed
Scalability of community wireless mesh networks in terms of mesh nodes and mobile end-users along with their anticipated usage profile remains unexplored territory. However, conducting such experiments on a live community network could cause network breakdown leading to disrupted network services, and discomfort to end-users
