    Multimodal Data at Signalized Intersections: Strategies for Archiving Existing and New Data Streams to Support Operations and Planning & Fusion and Integration of Arterial Performance Data

    There is a growing interest in arterial system management due to the increasing amount of travel on arterials and a growing emphasis on multimodal transportation. The benefits of archiving arterial-related data are numerous. This research report describes our efforts to assemble and develop a multimodal archive for the Portland-Vancouver region. There is coverage of data sources from all modes in the metropolitan region; however, the preliminary nature of the archiving process means that some of the data are incomplete and samples. The arterial data sources available in the Portland-Vancouver region and that are covered in this report include data for various local agencies (City of Portland, Clark County, WA, TriMet and C-TRAN) covering vehicle, transit, pedestrian, and bicycle modes. We provide detailed descriptions of each data source and a spatial and temporal classification. The report describes the conceptual framework for an archive and the data collection and archival process, including the process for extracting the data from the agency systems and transferring these data to our multimodal database. Data can be made more useful though the use of improved visualization techniques. Thus as part of the project, a number of novel, online visualizations were created and implemented. These graphs and displays are summarized in this report and example visualizations are shown. As with any automated sensor system, data quality and completeness is an important issue and the challenge of automating data quality is large. Preliminary efforts to validate and monitor data quality and automate data quality processing are explored. Finally, the report presents efforts to combine transit and travel time data and signal timing and vehicle count data to generate some sample congestion measures

    CD25+ CD4+ T Cells, Expanded with Dendritic Cells Presenting a Single Autoantigenic Peptide, Suppress Autoimmune Diabetes

    In the nonobese diabetic (NOD) mouse model of type 1 diabetes, the immune system recognizes many autoantigens expressed in pancreatic islet β cells. To silence autoimmunity, we used dendritic cells (DCs) from NOD mice to expand CD25+ CD4+ suppressor T cells from BDC2.5 mice, which are specific for a single islet autoantigen. The expanded T cells were more suppressive in vitro than their freshly isolated counterparts, indicating that DCs from autoimmune mice can increase the number and function of antigen-specific, CD25+ CD4+ regulatory T cells. Importantly, only 5,000 expanded CD25+ CD4+ BDC2.5 T cells could block autoimmunity caused by diabetogenic T cells in NOD mice, whereas 105 polyclonal, CD25+ CD4+ T cells from NOD mice were inactive. When islets were examined in treated mice, insulitis development was blocked at early (3 wk) but not later (11 wk) time points. The expanded CD25+ CD4+ BDC2.5 T cells were effective even if administered 14 d after the diabetogenic T cells. Our data indicate that DCs can generate CD25+ CD4+ T cells that suppress autoimmune disease in vivo. This might be harnessed as a new avenue for immunotherapy, especially because CD25+ CD4+ regulatory cells responsive to a single autoantigen can inhibit diabetes mediated by reactivity to multiple antigens

    COVID and Curriculum: Elementary Teachers Report on the Challenges of Teaching and Learning Mathematics Remotely

    This article reports on findings from a survey administered to 524 elementary teachers across 46 states that asked about their experiences with mathematics teaching, learning, and curriculum use before and during the COVID-19 pandemic. The purpose of this article is to report on the challenges teachers experienced with mathematics teaching, learning, and curriculum use during the pandemic and to explore educational inequities faced by students of families with lower income backgrounds. In particular, we discuss differences across high- and low-income schools regarding teachers’ perceived preparedness for online teaching, teachers’ use and decisions about mathematics curriculum, and their students’ remote resources (i.e., internet, computer, and workspace). We also share statements from teachers that illustrate challenges they experienced in engaging students, assessing learning outcomes, and building relationships. We found that students and teachers in schools from all socioeconomic levels faced hardship. However, lower income schools generally faced greater challenges in terms of access to computers, internet, and adequate workspaces. This often resulted in decreased access to teachers and decreased engagement. The exacerbation of educational inequities experienced by students from low income backgrounds during the pandemic suggests that we may see further widening of gaps in learning growth between students of low- and high-socioeconomic backgrounds. This has implications for policy makers, teacher educators, and other stakeholders seeking to advance equity and justice for less wealthy students

    Summary of CPAS Gen II Parachute Analysis

    The Orion spacecraft is currently under development by NASA and Lockheed Martin. Like Apollo, Orion will use a series of parachutes to slow its descent and splashdown safely. The Orion parachute system, known as the CEV Parachute Assembly System (CPAS), is being designed by NASA, the Engineering and Science Contract Group (ESCG), and Airborne Systems. The first generation (Gen I) of CPAS testing consisted of thirteen tests and was executed in the 2007-2008 timeframe. The Gen I tests provided an initial understanding of the CPAS parachutes. Knowledge gained from Gen I testing was used to plan the second generation of testing (Gen II). Gen II consisted of six tests: three singleparachute tests, designated as Main Development Tests, and three Cluster Development Tests. Gen II required a more thorough investigation into parachute performance than Gen I. Higher fidelity instrumentation, enhanced analysis methods and tools, and advanced test techniques were developed. The results of the Gen II test series are being incorporated into the CPAS design. Further testing and refinement of the design and model of parachute performance will occur during the upcoming third generation of testing (Gen III). This paper will provide an overview of the developments in CPAS analysis following the end of Gen I, including descriptions of new tools and techniques as well as overviews of the Gen II tests

    Simulating New Drop Test Vehicles and Test Techniques for the Orion CEV Parachute Assembly System

    The Crew Exploration Vehicle Parachute Assembly System (CPAS) project is engaged in a multi-year design and test campaign to qualify a parachute recovery system for human use on the Orion Spacecraft. Test and simulation techniques have evolved concurrently to keep up with the demands of a challenging and complex system. The primary simulations used for preflight predictions and post-test data reconstructions are Decelerator System Simulation (DSS), Decelerator System Simulation Application (DSSA), and Drop Test Vehicle Simulation (DTV-SIM). The goal of this paper is to provide a roadmap to future programs on the test technique challenges and obstacles involved in executing a large-scale, multi-year parachute test program. A focus on flight simulation modeling and correlation to test techniques executed to obtain parachute performance parameters are presented

    Structure of a Protozoan Virus from the Human Genitourinary Parasite Trichomonas vaginalis

    The flagellated protozoan Trichomonas vaginalis is an obligate human genitourinary parasite and the most frequent cause of sexually transmitted disease worldwide. Most clinical isolates of T. vaginalis are persistently infected with one or more double-stranded RNA (dsRNA) viruses from the genus Trichomonasvirus, family Totiviridae, which appear to influence not only protozoan biology but also human disease. Here we describe the three-dimensional structure of Trichomonas vaginalis virus 1 (TVV1) virions, as determined by electron cryomicroscopy and icosahedral image reconstruction. The structure reveals a T = 1 capsid comprising 120 subunits, 60 in each of two nonequivalent positions, designated A and B, as previously observed for fungal Totiviridae family members. The putative protomer is identified as an asymmetric AB dimer consistent with either decamer or tetramer assembly intermediates. The capsid surface is notable for raised plateaus around the icosahedral 5-fold axes, with canyons connecting the 2- and 3-fold axes. Capsid-spanning channels at the 5-fold axes are unusually wide and may facilitate release of the viral genome, promoting dsRNA-dependent immunoinflammatory responses, as recently shown upon the exposure of human cervicovaginal epithelial cells to either TVV-infected T. vaginalis or purified TVV1 virions. Despite extensive sequence divergence, conservative features of the capsid reveal a helix-rich fold probably derived from an ancestor shared with fungal Totiviridae family members. Also notable are mass spectrometry results assessing the virion proteins as a complement to structure determination, which suggest that translation of the TVV1 RNA-dependent RNA polymerase in fusion with its capsid protein involves −2, and not +1, ribosomal frameshifting, an uncommonly found mechanism to date
