    Covariance and PCA for Categorical Variables

    Covariances from categorical variables are defined using a regular simplex expression for categories. The method follows the variance definition by Gini, and it gives the covariance as a solution of simultaneous equations. The calculated results give reasonable values for test data. A method of principal component analysis (RS-PCA) is also proposed using regular simplex expressions, which allows easy interpretation of the principal components. The proposed methods apply to variable selection problem of categorical data USCensus1990 data.     The cross-correlation search for a hot spot of gravitational waves : Numerical study for point spread function

    The cross-correlation search for gravitational wave, which is known as 'radiometry', has been previously applied to map of the gravitational wave stochastic background in the sky and also to target on gravitational wave from rotating neutron stars/pulsars. We consider the Virgo cluster where may be appear as `hot spot' spanning few pixels in the sky in radiometry analysis. Our results show that sufficient signal to noise ratio can be accumulated with integration times of the order of a year. We also construct numerical simulation of radiometry analysis, assuming current constructing/upgrading ground-based detectors. Point spread function of the injected sources are confirmed by numerical test. Typical resolution of radiometry analysis is a few square degree which corresponds to several thousand pixels of sky mapping.Comment: 9 pages, 9 figures, Amaldi 9 & NRD

    Imaging and mapping of mouse bone using MALDI-imaging mass spectrometry

    Matrix-assisted laser desorption/ionization-imaging mass spectrometry (MALDI-IMS) is an advanced method used globally to analyze the distribution of biomolecules on tissue cryosections without any probes. In bones, however, hydroxyapatite crystals make it difficult to determine the distribution of biomolecules using MALDI-IMS. Additionally, there is limited information regarding the use of this method to analyze bone tissues. To determine whether MALDI-IMS analysis of bone tissues can facilitate comprehensive mapping of biomolecules in mouse bone, we first dissected femurs and tibiae from 8-week-old male mice and characterized the quality of multiple fixation and decalcification methods for preparation of the samples. Cryosections were mounted on indium tin oxide-coated glass slides, dried, and then a matrix solution was sprayed on the tissue surface. Images were acquired using an iMScope at a mass-to-charge range of 100–1000. Hematoxylin-eosin, Alcian blue, Azan, and periodic acid-Schiff staining of adjacent sections was used to evaluate histological and histochemical features. Among the various fixation and decalcification conditions, sections from trichloroacetic acid-treated samples were most suitable to examine both histology and comprehensive MS images. However, histotypic MS signals were detected in all sections. In addition to the MS images, phosphocholine was identified as a candidate metabolite. These results indicate successful detection of biomolecules in bone using MALDI-IMS. Although analytical procedures and compositional adjustment regarding the performance of the device still require further development, IMS appears to be a powerful tool to determine the distribution of biomolecules in bone tissues

    Upper Limit on Gravitational Wave Backgrounds at 0.2 Hz with Torsion-bar Antenna

    We present the first upper limit on gravitational wave (GW) backgrounds at an unexplored frequency of 0.2 Hz using a torsion-bar antenna (TOBA). A TOBA was proposed to search for low-frequency GWs. We have developed a small-scaled TOBA and successfully found {\Omega}gw(f) < 4.3 \times 1017 at 0.2 Hz as demonstration of the TOBA's capabilities, where {\Omega}gw (f) is the GW energy density per logarithmic frequency interval in units of the closure density. Our result is the first nonintegrated limit to bridge the gap between the LIGO band (around 100 Hz) and the Cassini band (10-6 - 10-4 Hz).Comment: 4 pages, 5 figure

    Minimum oxygen cost of human walking with geometrically similar leg movements

    The mechanism by which the expenditure of oxygen to walk per unit distance at an intermediate speed is minimized, by definition optimal walking, was investigated to characterize optimal walking in humans with variations in individual walking speeds. Oxygen uptake and step rate(SR)were measured among 7 young male subjects walking at an increasing speed from 16.7 to 131.7 m min−1 with 5 m min−1 increments every 1 min on a level treadmill. Measurements of leg length(L)were also made and step length(SL)was calculated by dividing walking speed by SR. The hip joint angle(θ)was calculated as a function of both L and SL such that θ=2sin−[1 SL(/ 2L)] deduced from a mathematical geometrically similar model of pendulum−like legs. The minimum oxygen cost to walk per unit distance for each subject was observed over a wide range of speeds from 60 to 100 m min−1.However, the oxygen cost of walking for all the subjects was minimized during a step cycle through a hip−joint angle of about 46 deg in the astride position, regardless of L. The stifflegged model demonstrated that the pathway of the trunk during optimal walking with a swing leg angle of 46 deg was approximately maintained at an even level by the counteracting effects of the leg decline and the heel rise. These results suggest that the minimum oxygen cost of transport during optimal walking was achieved by the mechanism underlying the maximum interchange between the gravitational−potential and kinetic energy for the body with an even level of the trunk that reduces extra muscular work needed against internal and external resistance, as well as against gravit

    The cross-correlation search has been previously applied to map the gravitational wave (GW) stochastic background in the sky and also to target GW from rotating neutron stars/pulsars. Here we investigate how the cross-correlation method can be used to target a small region in the sky spanning at most a few pixels, where a pixel in the sky is determined by the diffraction limit which depends on the (i) baseline joining a pair of detectors and (ii) detector bandwidth. Here as one of the promising targets, we consider the Virgo cluster - a "hot spot" spanning few pixels - which could contain, as estimates suggest 1011\sim 10^{11} neutron stars, of which a small fraction would continuously emit GW in the bandwidth of the detectors. For the detector baselines, we consider advanced detector pairs among LCGT, LIGO, Virgo, ET etc. Our results show that sufficient signal to noise can be accumulated with integration times of the order of a year. The results improve for the multibaseline search. This analysis could as well be applied to other likely hot spots in the sky and other possible pairs of detectors.Comment: 7 pages, 2 figure

    The Identification of a Small Molecule Compound That Reduces HIV-1 Nef-Mediated Viral Infectivity Enhancement

    Nef is a multifunctional HIV-1 protein that accelerates progression to AIDS, and enhances the infectivity of progeny viruses through a mechanism that is not yet understood. Here, we show that the small molecule compound 2c reduces Nef-mediated viral infectivity enhancement. When added to viral producer cells, 2c did not affect the efficiency of viral production itself. However, the infectivity of the viruses produced in the presence of 2c was significantly lower than that of control viruses. Importantly, an inhibitory effect was observed with Nef+ wild-type viruses, but not with viruses produced in the absence of Nef or in the presence of proline-rich PxxP motif-disrupted Nef, both of which displayed significantly reduced intrinsic infectivity. Meanwhile, the overexpression of the SH3 domain of the tyrosine kinase Hck, which binds to a PxxP motif in Nef, also reduced viral infectivity. Importantly, 2c inhibited Hck SH3-Nef binding, which was more marked when Nef was pre-incubated with 2c prior to its incubation with Hck, indicating that both Hck SH3 and 2c directly bind to Nef and that their binding sites overlap. These results imply that both 2c and the Hck SH3 domain inhibit the interaction of Nef with an unidentified host protein and thereby reduce Nef-mediated infectivity enhancement. The first inhibitory compound 2c is therefore a valuable chemical probe for revealing the underlying molecular mechanism by which Nef enhances the infectivity of HIV-1

    Insertable inductively coupled volumetric coils for MR microscopy in a human 7T MR system

    PURPOSE: To demonstrate the capability of insertable inductively coupled volumetric coils for MR microscopy in a human 7T MR system. METHODS: Insertable inductively coupled volume coils with diameters of 26 and 64 mm (D26 and D64 coils) targeted for monkey and mouse brain specimen sizes were designed and fabricated. These coils were placed inside the imaging volume of a transmit/receive knee coil without wired connections to the main system. Signal-to-noise ratio (SNR) evaluations were conducted with and without the insertable coils, and the g-factor maps of parallel imaging (PI) were also calculated for the D64 coil. Brain specimens were imaged using 3D T 2 ∗ -weighted images with spatial resolution of isotropic 50 and 160 μm using D26 and D64 coils, respectively. RESULTS: Relative average (SD) SNRs compared with knee coil alone were 12.54 (0.30) and 2.37 (0.05) at the center for the D26 and D64 coils, respectively. The mean g-factors of PI with the D64 coil for the factor of 2 were less than 1.1 in the left-right and anterior-posterior directions, and around 1.5 in the superior-inferior direction or when the PI factor of 3 was used. Acceleration in two directions showed lower g-factors but suffered from intrinsic low SNR. Representative T 2 ∗ -weighted images of the specimen showed structural details. CONCLUSION: Inductively coupled small diameter coils insertable to the knee coil demonstrated high SNR and modest PI capability. The concept was successfully used to visualize fine structures of the brain specimen. The insertable coils are easy to handle and enable MR microscopy in a human whole-body 7T MRI system

    Protocol of a Prospective Observational Study on the Relationship Between Glucose Fluctuation and Cardiovascular Events in Patients with Type 2 Diabetes

    IntroductionA recent study demonstrated that large glucose fluctuations were associated with an increased incidence of cardiovascular disease (CVD) in patients with type 2 diabetes mellitus (T2DM) and acute myocardial infarction. However, it is unknown whether glucose fluctuations are related to the incidence of CVD or the progression of atherosclerosis in patients with T2DM with no apparent history of CVD. In this protocol, we will be investigating the relationships of glucose fluctuations evaluated by continuous glucose monitoring (CGM) to the incidence of composite cardiovascular events and the progression of atherosclerosis in patients with T2DM who had no apparent history of CVD.MethodsThis is a prospective, multicenter, 5-year follow-up observational study. Between April 2018 and October 2019, 1000 participants are expected to be recruited at 34 medical institutions. CGM using FreeStyle Libre Pro is useful for evaluating glucose fluctuations by continuously monitoring glucose levels in interstitial fluid for up to 14 days. The primary study outcome is the relationship between fluctuations in glucose levels evaluated by CGM and the incidence of composite cardiovascular events. Secondary outcomes include the relationships of fluctuations in glucose levels evaluated by CGM to changes in carotid intima media thickness evaluated by echography or grayscale median (an index of tissue characteristics of the carotid wall), brachial–ankle pulse wave velocity, development or progression of diabetic retinopathy or nephropathy, quality-of-life-related diabetes therapy, quality of sleep, development of dementia, and autonomic nerve function.Planned OutcomeThis protocol is designed to investigate the relationship between glucose fluctuations and the incidence of composite cardiovascular events. We completed the registration of 1000 participants in March 2019. Thus, results will be available in 2024.     The geomorphology, color, and thermal properties of Ryugu: Implications for parent-body processes

    The near-Earth carbonaceous asteroid 162173 Ryugu is thought to have been produced from a parent body that contained water ice and organic molecules. The Hayabusa2 spacecraft has obtained global multi-color images of Ryugu. Geomorphological features present include a circum-equatorial ridge, east/west dichotomy, high boulder abundances across the entire surface, and impact craters. Age estimates from the craters indicate a resurfacing age of ≲ 106 years for the top 1-meter layer. Ryugu is among the darkest known bodies in the Solar System. The high abundance and spectral properties of boulders are consistent with moderately dehydrated materials, analogous to thermally metamorphosed meteorites found on Earth.
