    Pivotal Advance: Eosinophil infiltration of solid tumors is an early and persistent inflammatory host response

    Tumor-associated eosinophilia has been observed in numerous human cancers and several tumor models in animals; however, the details surrounding this eosinophilia remain largely undefined and anecdotal. We used a B16-F10 melanoma cell injection model to demonstrate that eosinophil infiltration of tumors occurred from the earliest palpable stages with significant accumulations only in the necrotic and capsule regions. Furthermore, the presence of diffuse extracellular matrix staining for eosinophil major basic protein was restricted to the necrotic areas of tumors, indicating that eosinophil degranulation was limited to this region. Antibody-mediated depletion of CD4+ T cells and adoptive transfer of eosinophils suggested, respectively, that the accumulation of eosinophils is not associated with T helper cell type 2-dependent immune responses and that recruitment is a dynamic, ongoing process, occurring throughout tumor growth. Ex vivo migration studies have identified what appears to be a novel chemotactic factor(s) released by stressed/dying melanoma cells, suggesting that the accumulation of eosinophils in tumors occurs, in part, through a unique mechanism dependent on a signal(s) released from areas of necrosis. Collectively, these studies demonstrate that the infiltration of tumors by eosinophils is an early and persistent response that is spatial-restricted. It is more important that these data also show that the mechanism(s) that elicit this host response occur, independent of immune surveillance, suggesting that eosinophils are part of an early inflammatory reaction at the site of tumorigenesis. © Society for Leukocyte Biology

    This case reports the unique association of eosinophilic gastrointestinal disease with eosinophilic bronchitis, asthma and chronic rhinosinusitis and some features of lymphocytic hypereosinophilic syndrome, describes a diagnostic protocol for patients with asthma and persistent eosinophilic bronchitis, and suggests that the use of a novel EPX-mAb provides a reliable method to identify eosinophilic inflammation

    Defining a link with asthma in mice congenitally deficient in eosinophils

    Eosinophils are often dominant inflammatory cells present in the lungs of asthma patients. Nonetheless, the role of these leukocytes remains poorly understood. We have created a transgenic line of mice (PHIL) that are specifically devoid of eosinophils, but otherwise have a full complement of hematopoietically derived cells. Allergen challenge of PHIL mice demonstrated that eosinophils were required for pulmonary mucus accumulation and the airway hyperresponsiveness associated with asthma. The development of an eosinophi-less mouse now permits an unambiguous assessment of a number of human diseases that have been linked to this granulocyte, including allergic diseases, parasite infections, and tumorigenesis

    A causative relationship exists between eosinophils and the development of allergic pulmonary pathologies in the mouse

    Asthma and mouse models of allergic respiratory inflammation are invariably associated with a pulmonary eosinophilia; however, this association has remained correlative. In this report, a causative relationship between eosinophils and allergen-provoked pathologies was established using eosinophil adoptive transfer. Eosinophils were transferred directly into the lungs of either naive or OVA-treated IL-5-/- mice. This strategy resulted in a pulmonary eosinophilia equivalent to that observed in OVA-treated wild-type animals. A concomitant consequence of this eosinophil transfer was an increase in Th2 bronchoalveolar lavage cytokine levels and the restoration of intracellular epithelial mucus in OVA-treated IL-5-/- mice equivalent to OVA-treated wild-type levels. Moreover, the transfer also resulted in the development of airway hyperresponsiveness. These pulmonary changes did not occur when eosinophils were transferred into naive IL-5-/- mice, eliminating nonspecific consequences of the eosinophil transfer as a possible explanation. Significantly, administration of OVA-treated IL-5-/- mice with GK1.5 (anti-CD4) Abs abolished the increases in mucus accumulation and airway hyperresponsiveness following adoptive transfer of eosinophils. Thus, CD4+ T cell-mediated inflammatory signals as well as signals derived from eosinophils are each necessary, yet alone insufficient, for the development of allergic pulmonary pathology. These data support an expanded view of T cell and eosinophil activities and suggest that eosinophil effector functions impinge directly on lung function

    Estudio psicológico acerca de una pedagogía de la sexualidad para los adolescentes. En primer lugar se relaciona educación y adolescencia y se señala que es necesario unificar la base de la educación y tomar como punto de arranque de la misma un concepto del hombre más humanista. El hombre domina la técnica, pero se olvida de la técnica de sí mismo. Los educadores han de estar alerta para que, cuando el adolescente se asome a la vida, no se encuentre con un horizonte cerrado que le precipite en la desesperanza y el conflicto. En cuanto a la educación en la pubertad, uno de los momentos de desarrollo más difíciles que atraviesa el ser humano, su importancia resulta capital. El joven camina por un lugar hostil, con escasas fuerzas, mal equipado en muchos casos, absorbido por la ansiedad y oprimido por la angustia, expuesto a desviarse. La prioridad del factor psíquico debe ser absoluta. La psicología del adolescente ha intentado explicar las transformaciones psíquicas de esta edad por las transformaciones corporales, poniendo de relieve el factor sexual. Pero lo importante, desde el punto de vista psicológico, es la vivencia de este hecho, cómo vive el adolescente en general y cada adolescente en particular esta transformación. La tarea educativa debe analizar lo que ocurre con la sexualidad y con 1a inhibición de este impulso. Las normas sociales, los criterios éticos, religiosos, educativos, son fuerzas inhibidoras y represoras de gran carga pero no las únicas. Existe también un temor innato al misterio que emana de la sexualidad.
