    Depth of focus increase by multiplexing programmable diffractive lenses

    A combination of several diffractive lenses written onto a single programmable liquid crystal display (LCD) is proposed for increasing the Depth of Focus (DOF) of the imaging system as a whole. The lenses are spatially multiplexed in a random scheme onto the LCD. The axial irradiance distribution produced by each lens overlaps with the next one producing an extended focal depth. To compare the image quality of the multiplexed lenses, the Modulation Transfer Function (MTF) is calculated. Finally we obtain the experimental Point Spread Functions (PSF) for these multiplexed lenses and experimental results in which an extended object is illuminated under spatially incoherent monochromatic light. We compare the images obtained in the focal plane and in some defocused planes with the single lens and with three multiplexed lenses.     Mineralization of human premolar occlusal fissures: a quantitative histochemical microanalysis

    The mechanisms of cariogenesis in occlusal fissures remain elusive because of limited information about fissure structure and wall mineralization. The purpose of the present study was to determine the correlation between morphological patterns in occlusal fissures in human premolars and quantitative histochemical patterns of mineralization in the walls of these formations. We used scanning electron microscopy and quantitative X-ray microanalysis with the peak-tolocal background ratio method and microcrystalline calcium salts as standards. We distinguished three morphological patterns of fissures in scanning electron microscopic images. The wall of the fissures was less mineralized than the control enamel in all three types of fissures. Because the fissure walls are hypomineralized, we suggest that practicing dentists should take into account the degree of mineralization when they are preparing the fissures for the application of sealant.This work was partially supported by the Ministerio de Educación y Cultura (PB97-0840) and the Agencia Española de Cooperación Internacional (AECI)

    Effects of lng Mutations on LngA Expression, Processing, and CS21 Assembly in Enterotoxigenic Escherichia coli E9034A

    Enterotoxigenic Escherichia coli (ETEC) is a major cause of morbidity in children under 5 years of age in low- and middle-income countries and a leading cause of traveler's diarrhea worldwide. The ability of ETEC to colonize the intestinal epithelium is mediated by fimbrial adhesins, such as CS21 (Longus). This adhesin is a type IVb pilus involved in adherence to intestinal cells in vitro and bacterial self-aggregation. Fourteen open reading frames have been proposed to be involved in CS21 assembly, hitherto only the lngA and lngB genes, coding for the major (LngA) and minor (LngB) structural subunit, have been characterized. In this study, we investigated the role of the LngA, LngB, LngC, LngD, LngH, and LngP proteins in the assembly of CS21 in ETEC strain E9034A. The deletion of the lngA, lngB, lngC, lngD, lngH, or lngP genes, abolished CS21 assembly in ETEC strain E9034A and adherence to HT-29 cells was reduced 90%, compared to wild-type strain. Subcellular localization prediction of CS21 proteins was similar to other well-known type IV pili homologues. We showed that LngP is the prepilin peptidase of LngA, and that ETEC strain E9034A has another peptidase capable of processing LngA, although with less efficiency. Additionally, we present immuno-electron microscopy imagens to show that the LngB protein could be localized at the tip of CS21, and probably helps to control CS21 length. In conclusion, our results demonstrate that the LngA, LngB, LngC, LngD, LngH, and LngP proteins are essential for CS21 assembly, as well as for bacterial aggregation and adherence to HT-29 cells

    Fluorescent "turn-Off" detection of fluoride and cyanide ions using zwitterionic spirocyclic meisenheimer compounds

    Stable zwitterionic spirocyclic Meisenheimer compounds were synthesized using a one-step reaction between picric acid and diisopropyl (ZW1) or dicyclohexyl (ZW3) carbodiimide. A solution of these compounds displays intense orange fluorescence upon UV or visible light excitation, which can be quenched or "turned-off" by adding a mole equivalent amount of F⁻ or CN⁻ ions in acetonitrile.Fluorescence is not quenched in the presence of other ions such as Cl⁻, Br⁻, I⁻, NO₂⁻ , NO₃⁻ , or H₂PO₄⁻ These compounds can therefore be utilized as practical colorimetric and fluorescent probes for monitoring the presence of F⁻ or CN⁻ anions

    The regionalization of framing issues for producers Livestock Group of Validation and Technology Transfer (GGAVATT) from San Jose Miahuatlán Veracruz, Mexico, under the current production of native grass prairie with mainly during the months of higher temperature have a poor performance and thus milk production declines which promotes a great use of concentrated feeds which causes an increase in production cost and less use to them. For this purpose we made a diagnosis of current conditions GGAVATT production, yielding information that helps us source and compared to determine precisely what the effect of applying the proposed alternative. We propose an alternative implementation of pastures with improved pastures needed to produce a greater amount of forage per unit area especially in the colder months.La regionalización de la problemática encuadrando a los productores del Grupo Ganadero de Validación y Transferencia de Tecnología (GGAVATT) de San José Miahuatlán Veracruz, México, bajo la actual situación de producción de praderas básicamente con gramas nativas que durante los meses de menor frío tienen un pobre desempeño y por ende la producción de leche se deteriora lo cual promueve una gran utilización de alimentos concentrados lo que provoca un incremento en el costo de producción y una menor utilidad para estos. Para dicho propósito se realizó un diagnóstico de las condiciones actuales de producción del GGAVATT, arrojando información que nos sirve de partida y de comparación para determinar con precisión cuál es el efecto de aplicar la alternativa propuesta. Se plantea como alternativa la implantación de praderas con pastos mejorados que permitan producir una mayor cantidad de forrajes por unidad de superficie sobre todo en los meses de menor temperatura

    Histone H2A (H2A.X and H2A.Z) Variants in Molluscs: Molecular Characterization and Potential Implications For Chromatin Dynamics

    Histone variants are used by the cell to build specialized nucleosomes, replacing canonical histones and generating functionally specialized chromatin domains. Among many other processes, the specialization imparted by histone H2A (H2A.X and H2A.Z) variants to the nucleosome core particle constitutes the earliest response to DNA damage in the cell. Consequently, chromatin-based genotoxicity tests have been developed in those cases where enough information pertaining chromatin structure and dynamics is available (i.e., human and mouse). However, detailed chromatin knowledge is almost absent in most organisms, specially protostome animals. Molluscs (which represent sentinel organisms for the study of pollution) are not an exception to this lack of knowledge. In the present work we first identified the existence of functionally differentiated histone H2A.X and H2A.Z variants in the mussel Mytilus galloprovincialis (MgH2A.X and MgH2A.Z), a marine organism widely used in biomonitoring programs. Our results support the functional specialization of these variants based on: a) their active expression in different tissues, as revealed by the isolation of native MgH2A.X and MgH2A.Z proteins in gonad and hepatopancreas; b) the evolutionary conservation of different residues encompassing functional relevance; and c) their ability to confer specialization to nucleosomes, as revealed by nucleosome reconstitution experiments using recombinant MgH2A.X and MgH2A.Z histones. Given the seminal role of these variants in maintaining genomic integrity and regulating gene expression, their preliminary characterization opens up new potential applications for the future development of chromatin-based genotoxicity tests in pollution biomonitoring programs
