    We used 199 Corynebacterium diphtheriae isolated from 1995 to 1997 in Russia to evaluate the ability of random amplified polymorphic DNA (RAPD) to identify the unique clonal group that emerged there in 1990.     Recombinant Pseudomonas Vaccine: Technological Aspects of Obtaining and Evaluating Quality Indicators

    For an aim to prevent Pseudomonas aeruginosa, a candidate recombinant vaccine has been developed. This vaccine – (RPV) was based on two protective proteins of P. aeruginosa: the outer membrane protein F (OprF) and the recombinant truncated form of the Exotoxin A (toxoid) that were adsorbed on the aluminum hydroxide. The optimal immunization schedule for mice included two intraperitoneal administrations with a two-week interval. RPV promoted to increase survival rates in challenged immunized mice and stimulated humoral and innate immune responses. During preclinical studies, we confirmed the immunogenicity of the vaccine that had not pyrogenicity, acute and chronic toxicity, allergenicity and immunotoxicity.     Improving the level of food security of the region: cluster approach

    Scientific approaches to the study of food security at the regional level of the Russian Federation are considered. The main indicators characterizing the level of food security of the Vologda region have been analysed. The main problems in providing the population of the region with food products of their own production have been identified. The problem of the low level of production of meat and meat products in the region has been identified as a problem that requires the most careful attention of regional government bodies. It has been concluded that the strengthen state support for meat livestock enterprises of the region is necessary. The analysis of the opinions of domestic scientists on the application of the cluster approach to the development of the agro-industrial complex has been carried out. The expediency of creating a meat cluster of the Vologda region as a promising tool for solving the problem of the low level of provision of the population of the region with meat is justified. The results of the SWOT analysis of the meat cluster of the Vologda region have been presented. The general concept of the meat cluster of the region is proposed

    COMPARISON OF COLORIMETRIC AND CHEMILUMINESCENT ELISA TESTS FOR DETECTION OF IgG ANTIBODIES TO HUMAN EPO IN THE SERA OF EXPERIMENTAL ANIMALS

    Production of antibodies to erythropoietin-stimulating drugs is an important problem of therapy with recombinant human erythropoietin (EPO). It leads to changes in the pharmacokinetic profile and decreased therapeutic efficiency. Upon long-term treatment with EPO preparations, neutralizing antibodies can result in rare cases, thus leading to complete pure red cell aplasia. Hence, detection of antibodies to EPO is an important stage in the assessment of the drug immunogenicity in preclinical and clinical studies, as well as during the treatment with EPO. We have compared colorimetric and chemiluminescent ELISA tests for detection of IgG antibodies to human EPO with 3,3’,5,5’-tetramethylbenzidine – hydrogen peroxide and luminol -hydrogen peroxide detection systems, respectively. Аntibodies to human EPO were determined in blood serum samples of experimental animals, i.e., rabbits and guinea pigs following their immunization withdifferent doses of pegylated human recombinant EPO-beta subcutaneously or intravenously. The affinitypurified rabbit polyclonal antibodies to human EPO were used as a reference material. The effects of hydrogen peroxide and luminol concentrations upon sensitivity of a chemiluminescent method were also studied. We have shown a 1.5-2-fold increase in sensitivity when using 4-iodophenol for amplification of chemiluminescence. A comparison of the chemiluminescence intensity with time has demonstrated a better stability for the substrate mixture prepared on borate buffer. A decrease in chemiluminescence signal with time was proportional to the decrease in background signal, thus rendering stability of the signal/background ratio for 3 to 30 minutes. Due to optimizing the substrate mixture composition and conditions of chemiluminescence recording, the reached detection limits for colorimetric and chemiluminescent ELISA’s were, respectively, 0.6 ng/ml and 0.08 ng/ml. The measurement range was extended by more than 20 times for chemiluminescent ELISA. The chemiluminescent ELISA for anti-erythropoietin antibody detection showed a 1.9 to 2.6-fold increase in sensitivity for rabbit serum, and 1.8 to 8.9-fold for guinea pigs serum. Good correlation of results was found for quantitative detection of antibodies in rabbit sera using the two methods (R = 0.981).     The Middle Urals Distributed Power Generation Formation Based on CCCPP

    Irbit and Kamyshlov (as the examples of little towns in the Middle Urals) power systems state is analyzed in this paper and conclusions about the probable way of problems overcoming by means of distributed power generation formation in this region by medium sized high-performance CHP (Central Heating and Power) plants locating in these towns are drawn on the basis of perceived disadvantages of the systems. Highly efficient combined-cycle cogeneration power plant (CCCPP) able to provide high fuel combustion efficiency, flexibility and ecological compatibility are considered as energy providers in the proposed conception of the local power systems formation in the Middle Urals. The significance of this conception for the social and economic development of Irbit and Kamyshlov towns and Sverdlovsk region generally is described.В статье проанализировано состояние энергосистем малых городов Среднего Урала на примере городов Ирбит и Камышлов, на основе выявленных недостатков систем сделаны выводы о возможном способе преодоления проблем с помощью формирования распределённой генерации в регионе путём размещения в этих городах приближенных к потребителям высокоэкономичных ТЭЦ. В качестве генераторов энергии в предложенной концепции создания локальных энергетических структур на Среднем Урале выступают высокоэффективные когенерационные парогазовые установки ПГУ-ТЭЦ, способные обеспечить высокую эффективность использования топлива, маневренность и экологичность. Описано значение данной концепции для социально-экономического развития городов Ирбит, Камышлов и Свердловской области в целом

    Leaf Mesophyll Structure and Photosynthetic Activity in Calla palustris L. from Natural Habitats with Different Level of Technogenic Pollution

    A study of leaf structure and CO2 assimilation rate was conducted on the emergent plant Calla palustris L. from natural habitats with different levels of technogenic pollution (the Sak-Elga River, upstream and downstream of the Karabash copper smelter, Chelyabinsk region, Russia). It was found that both chlorophyll a content andthe rate of CO2 assimilation decreased twofold in plants from the downstream site. No significant changes were observed in leaf mesophyll structure and the volume of aerenchyma in the leaf. It was shown that in plants from strongly contaminated site, the decrease in spongy mesophyll cell volume was compensated by the increase in their number, whereas the decrease in the number of chloroplasts per cell was accompanied by a growth in volume. It is concluded that the changes in the numerical and dimensional characteristics of mesophyll cells and chloroplasts provide for the viability of C. palustris under prolonged technogenic impact and demonstrate the plasticity of the photosynthetic apparatus.
