    Respiratory health effects of e-cigarette substitution for tobacco cigarettes: a systematic review

    Background: E-cigarettes (electronic nicotine delivery system, ENDS) have been presented as a harm reduction strategy for people who smoke tobacco cigarettes but who cannot achieve abstinence, or for those who wish to continue to enjoy nicotine and the habit of smoking. What are the health effects of the substitution of ENDS for tobacco cigarettes? This systematic review evaluates the evidence of human clinical tests on the respiratory effects of ENDS use in participants who smoke tobacco cigarettes. Methods: A registered and published protocol was developed conforming to PRISMA 2020 and AMSTAR2 standards. The literature search was conducted in PubMed, Scopus, and the CENTRAL Cochrane Library and updated to May 2022. Three supplementary searches and a grey literature search were performed. Studies were evaluated with the JBI quality tools and the Oxford Catalogue of Bias. Due to the heterogeneity (diversity) of the studies, a narrative data synthesis was performed on the test findings plus three sub-group analyses. Results: The review consists of sixteen studies and twenty publications. Spirometry tests comprised the majority of the data. In total, 66 respiratory test measurements were reported, out of which 43 (65%) were not significant. Statistically significant findings were mixed, with 9 tests showing improvements and 14 measuring declines, none of which was clinically relevant. Ten studies were rated at a high risk of bias, and six had some concerns primarily due to inadequate research designs and the conduct of the studies. Reporting bias was documented in thirteen studies. Conclusions: Most of the studies showed no difference in respiratory parameters. This indicates that ENDS substitution for smoking likely does not result in additional harm to respiratory health. Due to the low quality of the studies, confidence in the conclusions is rated as low. Robust studies with a longer duration and sufficient power are required to validate any potential benefits or possible harms of ENDS substitution.     Whole-genome sequencing reveals host factors underlying critical COVID-19

    Critical COVID-19 is caused by immune-mediated inflammatory lung injury. Host genetic variation influences the development of illness requiring critical care1 or hospitalization2,3,4 after infection with SARS-CoV-2. The GenOMICC (Genetics of Mortality in Critical Care) study enables the comparison of genomes from individuals who are critically ill with those of population controls to find underlying disease mechanisms. Here we use whole-genome sequencing in 7,491 critically ill individuals compared with 48,400 controls to discover and replicate 23 independent variants that significantly predispose to critical COVID-19. We identify 16 new independent associations, including variants within genes that are involved in interferon signalling (IL10RB and PLSCR1), leucocyte differentiation (BCL11A) and blood-type antigen secretor status (FUT2). Using transcriptome-wide association and colocalization to infer the effect of gene expression on disease severity, we find evidence that implicates multiple genes—including reduced expression of a membrane flippase (ATP11A), and increased expression of a mucin (MUC1)—in critical disease. Mendelian randomization provides evidence in support of causal roles for myeloid cell adhesion molecules (SELE, ICAM5 and CD209) and the coagulation factor F8, all of which are potentially druggable targets. Our results are broadly consistent with a multi-component model of COVID-19 pathophysiology, in which at least two distinct mechanisms can predispose to life-threatening disease: failure to control viral replication; or an enhanced tendency towards pulmonary inflammation and intravascular coagulation. We show that comparison between cases of critical illness and population controls is highly efficient for the detection of therapeutically relevant mechanisms of disease

    Detailed host records are listed for 39 species of Bactrocera and 2 species of Dacus fruit flies, infesting 98 species of commercial and edible fruits in the Pacific Island Countries and Territories, based on sampling and incubating in laboratory almost 13,000 field collected samples, or over 380,000 fruits. For each host-fly-country association, quantitative data are presented on the weight and number of fruits collected, the proportion of infested samples, the number of adult flies emerged per kg of fruits and, whenever available, the percentage of individual fruits infested. All the published records of each fly-host-country association are cited and erroneous or dubious published records are rectified or commented. Laboratory forced infestation data are also cited and reviewed

    Introduction to Communication Avoiding Algorithms for Direct Methods of Factorization in Linear Algebra

    International audienceModern, massively parallel computers play a fundamental role in a large and rapidly growing number of academic and industrial applications. However, extremely complex hardware architectures, which these computers feature, effectively prevent most of the existing algorithms to scale up to a large number of processors. Part of the reason behind this is the exponentially increasing divide between the time required to communicate a floating-point number between two processors and the time needed to perform a single floating point operation by one of the processors. Previous investigations have typically aimed at overlapping as much as possible communication with computation. While this is important, the improvement achieved by such an approach is not sufficient. The communication problem needs to be addressed also directly at the mathematical formulation and the algorithmic design level. This requires a shift in the way the numerical algorithms are devised, which now need to reduce, or even minimize when possible, the number of communication instances. Communication avoiding algorithms provide such a perspective on designing algorithms that minimize communication in numerical linear algebra. In this document we describe some of the novel numerical schemes employed by those communication avoiding algorithms, with a particular focus on direct methods of factorization
