    This paper was commissioned by the Association of Directors of Children’s Services (ADCS) National Performance and Information Management Group (NPIMG) to support the work of ADCS to evaluate and assess the impact of Sector Led Improvement of Children and Young People’s Services, and to explore the possibility of designing and developing (with ADCS and other partners) an ‘Early Warning’ model. The objective is to identify when and where the performance of Children and Young People’s Services is moving in the wrong direction. We undertake pre-post testing and difference in difference analysis, the latter allows us to use control data to ensure that external factors affecting all regions are considered

    Orbital ATK, in partnership with Mark ONeill LLC (MOLLC) and SolAero Technologies Corp., has developed a novel solar array platform, PFC-CTA, which provides a significant advance in performance and cost reduction compared to all currently available space solar systems. PFC refers to the Point Focus Concentration of light provided by MOLLCs thin, flat Fresnel optics. These lenses focus light to a point of approximately 100 times the intensity of the ambient light, onto a solar cell of approximately 1/25th the size of the lens. CTA stands for Compact Telescoping Array1, which is the solar array blanket structural platform originally devised by NASA and currently being advanced by Orbital ATK and partners under NASA and AFRL funding to a projected TRL 5+ by late-2018. The NASA Game Changing Development Extreme Environment Solar Power (EESP) Option 1 Phase study has enabled Orbital ATK to generate and refine component designs, perform component level and system performance analyses, and test prototype hardware of the key elements of PFC-CTA, and increased the TRL of PFC-specific technology elements to TRL ~5. Key performance metrics currently projected are as follows: Scalability from 300 kW per wing (AM0); Specific Power > 250 W/kg (BoL, AM0); Stowage Efficiency > 60 kW/m3; 5:1 margin on pointing tolerance vs. capability; >50% launched cost savings; Wide range of operability between Venus and Saturn by active and/or passive thermal management

    Modulation of pathogen-induced CCL20 secretion from HT-29 human intestinal epithelial cells by commensal bacteria

    BACKGROUND: Human intestinal epithelial cells (IECs) secrete the chemokine CCL20 in response to infection by various enteropathogenic bacteria or exposure to bacterial flagellin. CCL20 recruits immature dendritic cells and lymphocytes to target sites. Here we investigated IEC responses to various pathogenic and commensal bacteria as well as the modulatory effects of commensal bacteria on pathogen-induced CCL20 secretion. HT-29 human IECs were incubated with commensal bacteria (Bifidobacterium infantis or Lactobacillus salivarius), or with Salmonella typhimurium, its flagellin, Clostridium difficile, Mycobacterium paratuberculosis, or Mycobacterium smegmatis for varying times. In some studies, HT-29 cells were pre-treated with a commensal strain for 2 hr prior to infection or flagellin stimulation. CCL20 and interleukin (IL)-8 secretion and nuclear factor (NF)-kappaB activation were measured using enzyme-linked immunosorbent assays. RESULTS: Compared to untreated cells, S. typhimurium, C. difficile, M. paratuberculosis, and flagellin activated NF-kappaB and stimulated significant secretion of CCL20 and IL-8 by HT-29 cells. Conversely, B. infantis, L. salivarius or M. smegmatis did not activate NF-kappaB or augment CCL20 or IL-8 production. Treatment with B. infantis, but not L. salivarius, dose-dependently inhibited the baseline secretion of CCL20. In cells pre-treated with B. infantis, C. difficile-, S. typhimurium-, and flagellin-induced CCL20 were significantly attenuated. B. infantis did not limit M. Paratuberculosis-induced CCL20 secretion. CONCLUSION: This study is the first to demonstrate that a commensal strain can attenuate CCL20 secretion in HT-29 IECs. Collectively, the data indicate that M. paratuberculosis may mediate mucosal damage and that B. infantis can exert immunomodulatory effects on IECs that mediate host responses to flagellin and flagellated enteric pathogens

    Advanced Paramedic Delivered Finger Thoracostomy

    Introduction & AimsTension pneumothorax is a potentially fatal but reversible injury encountered in major trauma and traumatic cardiac arrest. Needle decompression has been the standard treatment approach pre hospital in Ireland and internationally. However, concerns exist regarding the effectiveness of this approach due to anatomy and body habitus. We aim to describe the training, introduction and experience of finger thoracostomy by advanced paramedics within a pre hospital service in Ireland.MethodsFinger thoracostomy has been advocated as an alternative pre hospital treatment which is both diagnostic and therapeutic. Paramedic delivered thoracostomy is commonplace in pre hospital critical care services internationally. The MCI Medical Team (as part of Motorsport Rescue Services) is a PHECC-registered multidisciplinary team which provides medical cover at motorcycle road racing events in Ireland. The MCI Medical Team has significant experience of major trauma and routinely performs pre hospital anaesthesia for trauma patients. We introduced a training module on finger thoracostomy, comprising: theory, practical instruction and assessment for advanced paramedic members of the team.Results & ConclusionsAdvanced paramedic members of the team we trained to deliver finger thoracostomy in predefined circumstances when operating as part of the MCI medical team. To date, advanced paramedic delivered finger thoracostomy has been utilised on three occasions. Introduction of advanced paramedic delivered thoracostomy is a feasible and effective technique for the treatment of tension pneumothorax within a closely governed system

    SeedGerm: a cost‐effective phenotyping platform for automated seed imaging and machine‐learning based phenotypic analysis of crop seed germination

    Efficient seed germination and establishment are important traits for field and glasshouse crops. Large-scale germination experiments are laborious and prone to observer errors, leading to the necessity for automated methods. We experimented with five crop species, including tomato, pepper, Brassica, barley, and maize, and concluded an approach for large-scale germination scoring. Here, we present the SeedGerm system, which combines cost-effective hardware and open-source software for seed germination experiments, automated seed imaging, and machine-learning based phenotypic analysis. The software can process multiple image series simultaneously and produce reliable analysis of germination- and establishment-related traits, in both comma-separated values (CSV) and processed images (PNG) formats. In this article, we describe the hardware and software design in detail. We also demonstrate that SeedGerm could match specialists’ scoring of radicle emergence. Germination curves were produced based on seed-level germination timing and rates rather than a fitted curve. In particular, by scoring germination across a diverse panel of Brassica napus varieties, SeedGerm implicates a gene important in abscisic acid (ABA) signalling in seeds. We compared SeedGerm with existing methods and concluded that it could have wide utilities in large-scale seed phenotyping and testing, for both research and routine seed technology applications
