    Neural mechanisms underlying target detection in a dragonfly centrifugal neuron

    © The Company of Biologists Ltd 2007Visual identification of targets is an important task for many animals searching for prey or conspecifics. Dragonflies utilize specialized optics in the dorsal acute zone, accompanied by higher-order visual neurons in the lobula complex, and descending neural pathways tuned to the motion of small targets. While recent studies describe the physiology of insect small target motion detector (STMD) neurons, little is known about the mechanisms that underlie their exquisite sensitivity to target motion. Lobula plate tangential cells (LPTCs), a group of neurons in dipteran flies selective for wide-field motion, have been shown to take input from local motion detectors consistent with the classic correlation model developed by Hassenstein and Reichardt in the 1950s. We have tested the hypothesis that similar mechanisms underlie the response of dragonfly STMDs. We show that an anatomically characterized centrifugal STMD neuron (CSTMD1) gives responses that depend strongly on target contrast, a clear prediction of the correlation model. Target stimuli are more complex in spatiotemporal terms than the sinusoidal grating patterns used to study LPTCs, so we used a correlation-based computer model to predict response tuning to velocity and width of moving targets. We show that increasing target width in the direction of travel causes a shift in response tuning to higher velocities, consistent with our model. Finally, we show how the morphology of CSTMD1 allows for impressive spatial interactions when more than one target is present in the visual field.Bart R. H. Geurten, Karin Nordström, Jordanna D. H.     Impact of Analog and Digital Pre-emphasis on the Signal-to-Noise Ratio of Bandwidth-limited Optical Transceivers

    The ever-growing machine-to-machine traffic in data centers has stimulated the increase of transceiver data rate from 25Gb/s/λ to 100Gb/s/λ and beyond. It is believed that advanced modulation formats and digital-to-analog converters (DACs) will be employed in next generation short-reach transceivers. Digital pre-emphasis techniques are widely employed in DAC-based transceivers to compensate for the high frequency roll-off due to the RF and optoelectronics components in optical transceivers. However, digital pre-emphasis essentially reduces the magnitude of the signal low frequency components for a flat frequency response, which unavoidably increases quantization error, reducing the overall signal-to-noise ratio. This trade-off between SNR and bandwidth conflicts with the high SNR requirement of advanced modulation formats such as the Nyquist-shaped pulse amplitude modulation (PAM). To mitigate the quantization error induced SNR degradation, we show that analog pre-emphasis filters can be used in conjunction with digital pre-emphasis for improved system performance. To understand the impact of the analog pre-emphasis filter on system performance, we analyze the relationship between the flatness of the system frequency response and the SNR degradation due to digital pre-emphasis, and demonstrate 1.1-dB increase of receiver sensitivities, for both 64-Gb/s and 128-Gb/s intensity-modulation direct detection (IM-DD) 20 PAM4 signals, respectively employing a directly modulated laser (DML) and an electroabsorption modulator (EAM)

    Sulfidation enhances stability and mobility of carboxymethyl cellulose stabilized nanoscale zero-valent iron in saturated porous media

    Sulfidation can enhance the reactivity and longevity of nanoscale zero-valent iron (nZVI), but little is known about its effect on the fate and transport of nZVI in saturated porous media. This work compared the stability and mobility of carboxymethyl cellulose (CMC) stabilized nZVI (CMC-nZVI) and sulfidated nZVI (CMC-S-nZVI) particles in saturated porous media. After sulfidation, the hydrodynamic size of CMC-S-nZVI was 100–150 nm larger than CMC-nZVI due to enhanced adsorption of CMC onto the S-nZVI surface, which was facilitated by the bidentate bridging interaction between CMC and the FeSx phase on S-nZVI. Of note is that they had a similar core size and zeta potential. In comparison to CMC-nZVI, CMC-S-nZVI exhibited less physical settling (0–5% vs. 5–73%) and chemical dissolution (2–10% vs. 3–27%) within 55 min under the same ionic conditions (Na+, K+ < 200 mM; Al3+ < 0.75 mM). Column breakthrough experiments showed that both CMC-S-nZVI and CMC-nZVI had relatively high mobility in saturated porous media. However, CMC-S-nZVI exhibited greater breakthrough (C/C0 = 0.57–1.0) and corresponding greater mass recovery rates than the corresponding CMC-nZVI (C/C0 = 0.44–1.0) under most of the experimental conditions (e.g., different ion type and concentration, flow rate, and input concentration). The fitted colloid filtration theory model was in good agreement with experiments. This work suggests that in addition to the significant reactivity and longevity improvements demonstrated in other studies, CMC-S-nZVI is also more mobile than CMC-nZVI suggesting that CMC-S-nZVI has many of the characteristics favorable for field application

    Suicide attempts and non-suicidal self-harm: national prevalence study of young adults

    Background There are few prevalence studies of suicide attempts and non-suicidal self-harm (NSSH). Aims We aimed to estimate the prevalence of thoughts of NSSH, suicidal thoughts, NSSH and suicide attempts among 18- to 34-year-olds in Scotland. Method We interviewed a representative sample of young adults from across Scotland. Results We interviewed 3508 young people; 11.3 and 16.2% reported a lifetime history of suicide attempts and NSSH, respectively. The first episode of NSSH tended to precede the first suicide attempt by about 2 years. Age at onset of NSSH and suicide attempt was younger in females. Earlier age at onset was associated with more frequent NSSH/suicide attempts. Women are significantly more likely to report NSSH and suicide attempts compared with men. Conclusions One in nine young people has attempted suicide and one in six has engaged in NSSH. Clinicians should be vigilant, as suicide attempts and NSSH are relatively common

    Addressing complex pharmacy consultations: methods used to develop a person-centred intervention to highlight alcohol within pharmacist reviews of medications

    Background: Alcohol is challenging to discuss, and patients may be reluctant to disclose drinking partly because of concern about being judged. This report presents an overview of the development of a medications review intervention co-produced with the pharmacy profession and with patients, which breaks new ground by seeking to give appropriate attention to alcohol within these consultations. // Methods: This intervention was developed in a series of stages and refined through conceptual discussion, literature review, observational and interview studies, and consultations with advisory groups. In this study we reflect on this process, paying particular attention to the methods used, where lessons may inform innovations in other complex clinical consultations. // Results: Early work with patients and pharmacists infused the entire process with a heightened sense of the complexity of consultations in everyday practice, prompting careful deliberation on the implications for intervention development. This required the research team to be highly responsive to both co-production inputs and data gathered in formally conducted studies, and to be committed to working through the implications for intervention design. The intervention thus evolved significantly over time, with the greatest transformations resulting from patient and pharmacist co-design workshops in the second stage of the process, where pharmacists elaborated on the nature of the need for training in particular. The original research plans provided a helpful structure, and unanticipated issues for investigation emerged throughout the process. This underscored the need to engage dynamically with changing contexts and contents and to avoid rigid adherence to any early prescribed plan. // Conclusions: Alcohol interventions are complex and require careful developmental research. This can be a messy enterprise, which can nonetheless shed new insights into the challenges involved in optimising interventions, and how to meet them, if embraced with an attitude of openness to learning. We found that exposing our own research plans to scrutiny resulted in changes to the intervention design that gained the confidence of different stakeholders. Our understanding of the methods used, and their consequences, may be bounded by the person-centred nature of this particular intervention

    Modulator-free quadrature amplitude modulation signal synthesis

    The ability to generate high-speed on–off-keyed telecommunication signals by directly modulating a semiconductor laser’s drive current was one of the most exciting prospective applications of the nascent field of laser technology throughout the 1960s. Three decades of progress led to the commercialization of 2.5 Gbit s−1-per-channel submarine fibre optic systems that drove the growth of the internet as a global phenomenon. However, the detrimental frequency chirp associated with direct modulation forced industry to use external electro-optic modulators to deliver the next generation of on–off-keyed 10 Gbit s−1 systems and is absolutely prohibitive for today’s (>)100 Gbit s−1 coherent systems, which use complex modulation formats (for example, quadrature amplitude modulation). Here we use optical injection locking of directly modulated semiconductor lasers to generate complex modulation format signals showing distinct advantages over current and other currently researched solutions

    Therapeutic limitations in tumor-specific CD8+ memory T cell engraftment

    BACKGROUND: Adoptive immunotherapy with cytotoxic T lymphocytes (CTL) represents an alternative approach to treating solid tumors. Ideally, this would confer long-term protection against tumor. We previously demonstrated that in vitro-generated tumor-specific CTL from the ovalbumin (OVA)-specific OT-I T cell receptor transgenic mouse persisted long after adoptive transfer as memory T cells. When recipient mice were challenged with the OVA-expressing E.G7 thymoma, tumor growth was delayed and sometimes prevented. The reasons for therapeutic failures were not clear. METHODS: OT-I CTL were adoptively transferred to C57BL/6 mice 21 – 28 days prior to tumor challenge. At this time, the donor cells had the phenotypical and functional characteristics of memory CD8+ T cells. Recipients which developed tumor despite adoptive immunotherapy were analyzed to evaluate the reason(s) for therapeutic failure. RESULTS: Dose-response studies demonstrated that the degree of tumor protection was directly proportional to the number of OT-I CTL adoptively transferred. At a low dose of OT-I CTL, therapeutic failure was attributed to insufficient numbers of OT-I T cells that persisted in vivo, rather than mechanisms that actively suppressed or anergized the OT-I T cells. In recipients of high numbers of OT-I CTL, the E.G7 tumor that developed was shown to be resistant to fresh OT-I CTL when examined ex vivo. Furthermore, these same tumor cells no longer secreted a detectable level of OVA. In this case, resistance to immunotherapy was secondary to selection of clones of E.G7 that expressed a lower level of tumor antigen. CONCLUSIONS: Memory engraftment with tumor-specific CTL provides long-term protection against tumor. However, there are several limitations to this immunotherapeutic strategy, especially when targeting a single antigen. This study illustrates the importance of administering large numbers of effectors to engraft sufficiently efficacious immunologic memory. It also demonstrates the importance of targeting several antigens when developing vaccine strategies for cancer
