    Boxfishes (Teleostei: Ostraciidae) as a model system for fishes swimming with many fins: kinematics

    Swimming movements in boxfishes were much more complex and varied than classical descriptions indicated. At low to moderate rectilinear swimming speeds (<5 TL s^(-1), where TL is total body length), they were entirely median- and paired-fin swimmers, apparently using their caudal fins for steering. The pectoral and median paired fins generate both the thrust needed for forward motion and the continuously varied, interacting forces required for the maintenance of rectilinearity. It was only at higher swimming speeds (above 5 TL s^(-1)), when burst-and-coast swimming was used, that they became primarily body and caudal-fin swimmers. Despite their unwieldy appearance and often asynchronous fin beats, boxfish swam in a stable manner. Swimming boxfish used three gaits. Fin-beat asymmetry and a relatively nonlinear swimming trajectory characterized the first gait (0–1 TL s^(-1)). The beginning of the second gait (1–3 TL s^(-1)) was characterized by varying fin-beat frequencies and amplitudes as well as synchrony in pectoral fin motions. The remainder of the second gait (3–5 TL s^(-1)) was characterized by constant fin-beat amplitudes, varying finbeat frequencies and increasing pectoral fin-beat asynchrony. The third gait (>5 TL s^(-1)) was characterized by the use of a caudal burst-and-coast variant. Adduction was always faster than abduction in the pectoral fins. There were no measurable refractory periods between successive phases of the fin movement cycles. Dorsal and anal fin movements were synchronized at speeds greater than 2.5 TL s^(-1), but were often out of phase with pectoral fin movements

    DNA double strand breaks but not interstrand crosslinks prevent progress through meiosis in fully grown mouse oocytes

    There is some interest in how mammalian oocytes respond to different types of DNA damage because of the increasing expectation of fertility preservation in women undergoing chemotherapy. Double strand breaks (DSBs) induced by ionizing radiation and agents such as neocarzinostatin (NCS), and interstrand crosslinks (ICLs) induced by alkylating agents such as mitomycin C (MMC), are toxic DNA lesions that need to be repaired for cell survival. Here we examined the effects of NCS and MMC treatment on oocytes collected from antral follicles in mice, because potentially such oocytes are readily collected from ovaries and do not need to be in vitro grown to achieve meiotic competency. We found that oocytes were sensitive to NCS, such that this ionizing radiation mimetic blocked meiosis I and caused fragmented DNA. In contrast, MMC had no impact on the completion of either meiosis I or II, even at extremely high doses. However, oocytes treated with MMC did show ?-H2AX foci and following their in vitro maturation and parthenogenetic activation the development of the subsequent embryos was severely compromised. Addition of MMC to 1-cell embryos caused a similarly poor level of development, demonstrating oocytes have eventual sensitivity to this ICL-inducing agent but this does not occur during their meiotic division. In oocytes, the association of Fanconi Anemia protein, FANCD2, with sites of ICL lesions was not apparent until entry into the embryonic cell cycle. In conclusion, meiotic maturation of oocytes is sensitive to DSBs but not ICLs. The ability of oocytes to tolerate severe ICL damage and yet complete meiosis, means that this type of DNA lesion goes unrepaired in oocytes but impacts on subsequent embryo quality

    A Modified Price-Sales Ratio: A Useful Tool For Investors?

    This study compares returns for stock portfolios using low price-to-earnings, price-to-book, price-to-sales, and price-to-sales ratios modified for profitability of sales and long-term debt. Non-financial companies with fiscal years ending on December 31 were included in a sample covering a 17-year period (1988-2004). The resulting sample was sub-divided into six groups by level of market capitalization.  A 25 stock portfolio was then compiled for each of six valuation ratios at each level of market capitalization. An index portfolio was also computed for each level of market capitalization.  First, the results of this study showed that additional expected returns based on firm size, measured by the market value of equity, is found only at the micro-cap level (less than 25millionmarketcapitalization).Theindexportfoliosofminicaps(25 million market capitalization). The index portfolios of mini-caps (25-100 mill.), small caps (100500mill.),andmidcaps(100-500 mill.), and mid-caps (500-$1 bill.) did not show any additional returns in comparison to the large-cap index portfolio. Second, no "value" portfolio for mid-caps provided significant returns above the index portfolio, suggesting a much greater level of market efficiency at this level of market capitalization.  Finally, the most significant findings of our study were that the portfolios constructed from low price-to-sales and modified price-to sales ratios performed as well or better than the more traditional price-to-earnings and price-to-book ratios. The price-to-sales ratio adjusted for profit margin (based on the prior three years) was statistically significant in three of six portfolios in comparison to the more traditional valuation ratios

    Motility induced changes in viscosity of suspensions of swimming microbes in extensional flows

    Suspensions of motile cells are model systems for understanding the unique mechanical properties of living materials which often consist of ensembles of self-propelled particles. We present here a quantitative comparison of theory against experiment for the rheology of such suspensions. The influence of motility on viscosities of cell suspensions is studied using a novel acoustically-driven microfluidic capillary-breakup extensional rheometer. Motility increases the extensional viscosity of suspensions of algal pullers, but decreases it in the case of bacterial or sperm pushers. A recent model [Saintillan, Phys. Rev. E, 2010, 81:56307] for dilute active suspensions is extended to obtain predictions for higher concentrations, after independently obtaining parameters such as swimming speeds and diffusivities.     “My Uni Experience Wasn’t Completely Ruined”: The Impacts of COVID-19 on the First-Year Experience

    The first year at university is always challenging, but particularly in 2020 when COVID-19 triggered lockdowns and a rapid shift to online learning. This mixed methods study tracked the wellbeing and engagement of 60 new students in an undergraduate teacher education program at an Australian university throughout the first trimester of 2020. Follow-up focus groups with 14 students used interview and photo elicitation to explore how COVID-19 influenced wellbeing and engagement. Quantitative results demonstrate both student wellbeing and student engagement dipped strongly at the start of lockdown but recovered towards the end of the trimester. Focus group findings illustrate the diversity of experience in terms of student access to time and space to study, their ability to sustain relationships online, and the cumulative stress of COVID-19. The findings lead to recommendations for supporting this cohort and for future research

    The title merocyanine-type mol­ecule, C21H22N2O5·2H2O, crystallizes in a zwitterionic form and has an E configuration at the styryl C=C bond. The styryl part of the mol­ecule and the indolium ring are slightly twisted and form a dihedral angle of 13.4 (1)°. The 1.274 (3) Å C—O bond length in the phenolate fragment is the longest among similar mol­ecules. Hydrogen bonds between solvent water mol­ecules, two hydroxyl groups and the phenolate O atom dictate the packing arrangement of mol­ecules in the crystal and join the mol­ecules into a two-dimensional polymeric network which propagates parallel to (001). Four water mol­ecules and four hydr­oxy groups form a centrosymmetric homodromic cyclic motif of O—H⋯O hydrogen bonds. Another cyclic centrosymmetric motif is generated by four water mol­ecules and two phenolate O atoms

    Leopards provide public health benefits in Mumbai, India

    Populations of large carnivores are often suppressed in human-dominated landscapes because they can kill or injure people and domestic animals. However, carnivores can also provide beneficial services to human societies, even in urban environments. We examined the services provided by leopards (Panthera pardus) to the residents of Mumbai, India, one of the world's largest cities. We suggest that by preying on stray dogs, leopards reduce the number of people bitten by dogs, the risk of rabies transmission, and the costs associated with dog sterilization and management. Under one set of assumptions, the presence of leopards in this highly urbanized area could save up to 90 human lives per year. A further indirect benefit of leopard presence may be an increase in local abundance of other wildlife species that would otherwise be predated by dogs. The effective conservation of carnivores in human-dominated landscapes involves difficult trade-offs between human safety and conservation concerns. Quantitative assessments of how large carnivores negatively and positively affect urban ecosystems are critical, along with improved education of local communities about large carnivores and their impacts

    The unequal burden of human-wildlife conflict

    The costs of human-wildlife conflict are measurably more impactful in areas of the developing world where the loss of cattle can mean the entire livelihood of a family being erased.Human-wildlife conflict is one of the most pressing sustainable development challenges globally. This is particularly the case where ecologically and economically important wildlife impact the livelihoods of humans. Large carnivores are one such group and their co-occurrence with low-income rural communities often results in real or perceived livestock losses that place increased costs on already impoverished households. Here we show the disparities associated with the vulnerability to conflict arising from large carnivores on cattle (Bos taurus) globally. Across the distribution of 18 large carnivores, we find that the economic vulnerability to predation losses (as measured by impacts to annual per capita income) is between two and eight times higher for households in transitioning and developing economies when compared to developed ones. This potential burden is exacerbated further in developing economies because cattle keepers in these areas produce on average 31% less cattle meat per animal than in developed economies. In the lowest-income areas, our estimates suggest that the loss of a single cow or bull equates to nearly a year and a half of lost calories consumed by a child. Finally, our results show that 82% of carnivore range falls outside protected areas, and five threatened carnivores have over one third of their range located in the most economically sensitive conflict areas. This unequal burden of human-carnivore conflict sheds light on the importance of grappling with multiple and conflicting sustainable development goals: protecting life on land and eliminating poverty and hunger

    Conceptual design of a fifth generation unmanned strike fighter

    Unmanned aircraft have significantly transformed aerial warfare through a combination of new technologies, extended operational capabilities, and reduced risks and costs. Similarly, computational modelling techniques have accelerated the rate of development for aircraft by being able to explore a large number of design options from the earliest design stages, further reducing time, risks, and costs. The near future will see the proliferation of unmanned combat aerial vehicles under a variety of roles such as unmanned tankers, strike aircraft, and even air - to - air fighters. In this paper the GENUS aircraft design framework is used to develop an unmanned weapons carrying platform able to partially match the performance of 5th generation fighters such as the Joint Strike Fighter F-35A. The vision of future joint operations is for a single lead manned fighter to command and designate targets to its various loyal wingmen unmanned aircraft, extending the combat capabilities and significantly multiplying force and air superiority

    The testis and epididymis are productively infected by SIV and SHIV in juvenile macaques during the post-acute stage of infection

    BACKGROUND: Little is known about the progression and pathogenesis of HIV-1 infection within the male genital tract (MGT), particularly during the early stages of infection. RESULTS: To study HIV pathogenesis in the testis and epididymis, 12 juvenile monkeys (Macacca nemestrina, 4–4.5 years old) were infected with Simian Immunodeficiency Virus mac 251 (SIV(mac251)) (n = 6) or Simian/Human Immunodeficiency Virus (SHIV(mn229)) (n = 6). Testes and epididymides were collected and examined by light microscopy and electron microscopy, at weeks 11–13 (SHIV) and 23 (SIV) following infection. Differences were found in the maturation status of the MGT of the monkeys, ranging from prepubertal (lacking post-meiotic germ cells) to post-pubertal (having mature sperm in the epididymal duct). Variable levels of viral RNA were identified in the lymph node, epididymis and testis following infection with both SHIV(mn229 )and SIV(mac251). Viral protein was detected via immunofluorescence histochemistry using specific antibodies to SIV (anti-gp41) and HIV-1 (capsid/p24) protein. SIV and SHIV infected macrophages, potentially dendritic cells and T cells in the testicular interstitial tissue were identified by co-localisation studies using antibodies to CD68, DC-SIGN, αβTCR. Infection of spermatogonia, but not more mature spermatogenic cells, was also observed. Leukocytic infiltrates were observed within the epididymal stroma of the infected animals. CONCLUSION: These data show that the testis and epididymis of juvenile macaques are a target for SIV and SHIV during the post-acute stage of infection and represent a potential model for studying HIV-1 pathogenesis and its effect on spermatogenesis and the MGT in general
