    Cytotoxicity, Antiproliferative Assays, and Expresion of P53 and Bcl2 of Ethanolic Fraction From Tea (Camellia Sinensis (L.) O.k.) Leaves Infuse to Hela Cells

    Tea (Camellia sinensis (L.) O.K.) is one of medical plant traditionally used by society as anticancer. The aim of this research is to evaluate the citotoxic and antiproliferative effect of ethanolic fraction of tea (Camellia sinensis (L.) O.K.) and its effect on P53 and bcl-2. Tea were collected and extracted by infundation and fractioned with ethanol as solvent. Citotoxicity test was done by incubating HeLa cell at a density of 2.104 with treatment using extract in concentration of 250; 125 ; 62,5; 31,25; 15,63; and 7,81 µg/mL during 24 hours. Antiproliferative test were done by calculating doubling time which was equaling a life cell on treatment sample concentration 31,25 µg/mL; 15,63 µg/mL; 7,81 µg/mL; and 3,91 µg/mL with cell control at 24, 48 and 72 hours. Imunohistochemistry assay was used in LC50 with p53 and bcl-2 antibody. P53 and bcl-2 expression is compared to control. The result indicated that ethanol fraction of tea (Camellia sinensis (L.) O.K.) leaf infuse has citotoxicity effect on HeLa cell with LC50 of 24,45 µg/mL. The result of doubling time test indicated that doubling time value for a solvent control is 74,11 hours and a cell control is 78,22 hours. While, with treatment 31,25 µg/mL; 15,63 µg/mL; 7,81 µg/mL; and 3,91 µg/mL resulted on negative slope then did not produce doubling time.     Enhancement of Icariin Aphrodisiac Effect by Solid-SNEDDS Method Using Shark Liver Oil Phase

    The Epimedium brevicornu Maxim plant contains icariin, a flavonoid compound known for its aphrodisiac effects. However, icariin has low solubility and bioavailability. This study aims to find the best formula for S-SNEDDS icariin, its physical properties, characteristics, and aphrodisiac activity. Using the S-SNEDDS (Solid-Self Nanoemulsifying Drug Delivery System) method with shark liver oil phase is expected to increase the solubility and bioavailability of icariin. The optimum formula used is tween 80 (72.5%): PEG 400 (13.75%) and shark liver oil (13.75%). The optimal formula of S-SNEDDS icariin with the adsorption method to solid carriers requires aerosol 200 as much as 783±28.86mg per 1mL. S-SNEDDS icariin has characteristics with an average emulsification time of 12.88±0.26 seconds, transmittance value of 98.08±0.94%, droplet size 171.8±8.9 nm, zeta potential -35.2±1.1 mV, flow speed less than 10 seconds, resting angle 35.159°. The dissolution test of S-SNEDDS icariin is better than icariin instead of S-SNEDDS. S-SNEDDS icariin dose 50 mg/KgBW has a better aphrodisiac effect than pure icariin 100 mg/kgBW

    Hubungan Antara Konsentrasi Minyak Atsiri Kayu Manis (Cinnamomum Burmannii Nees Ex Bl.) Dalam Lotion Dengan Sifat Fisik Dan Tingkat Kesukaan Konsumen

    Essential oil of Cinnamomum burmannii contains cinnamaldehyde as active substance as antibacterial and repellant. The study about development of dosage forms was needed in order be useful to people. The purpose of this study was to determine the concentration\u27s effect of cinnamon to the physical characteristic and results hedonic test of lotion. This study was done in Laboratory of Pharmacy UAD in 2014 that was begun with distillation of essential oil. Then, it was formulated as lotion with concentration 1% (F1), 3% (F II), 5% (F III), 7% (F IV), respectively. The lotion was evaluated its physical characteristic (homogeneity, viscosity, spreadability, adhesivity, pH) and hedonic test to know the level of acceptability based on colour and flavor of lotion. Data was collected by quistionnaire. Results of study showed that all of the lotion was homogenous. The increasing concentrations of cinnamon caused decreasing of viscosity (p <0.05) and adhesivity (P <0.05), an increasing of spreadability (P <0.05), did not affect the pH value (p> 0.05), and increasing hedonic level until 5% concentration. It can be concluded that concentration of essential oil of cinnamon at 5% fullfilled the requirement of spreadability, adhesivity, pH and the most prefered by consumers. Minyak atsiri kayu manis dengan kandungan bahan aktif cinnamaldehyde telah terbukti berkhasiat sebagai antibakteri dan repelan. Penelitian terkait pengembangan bentuk sediaan diperlukan agar dapat dimanfaatkan oleh masyarakat. Tujuan penelitian ini adalah mengetahui pengaruh konsentrasi minyak atsiri kayu manis dalam lotion terhadap sifat fisik dan tingkat kesukaan konsumen. Penelitian ini dilaksanakan di Laboratorium Farmasi UAD tahun 2014 dengan rancangan eksperimental yang diawali dengan destilasi minyak atsiri kayu manis. Selanjutnya minyak atsiri diformulasikan dalam bentuk lotion dengan konsentrasi 1% (F1), 3% (F II), 5% (F III), 7% (F IV). Sediaan lotion diuji sifat fisik (homogenitas, viskositas, daya sebar, daya lekat, pH) dan uji hedonik untuk mengetahui tingkat kesukaan konsumen berdasarkan warna dan aroma lotion. Data tersebut diperoleh dengan memberikan kuisioner kepada 30 orang peserta. Hasil uji menunjukkan bahwa lotion homogen pada semua konsentrasi. Peningkatan konsentrasi minyak atsiri kayu manis menyebabkan penurunan viskositas (p<0,05) dan daya lekat (P<0,05), peningkatan daya sebar (P<0,05), tidak mempengaruhi nilai pH (p>0,05) serta peningkatan skor hedonik sampai pada konsentrasi 5%. Berdasarkan hasil uji maka dapat disimpulkan bahwa konsentrasi minyak atsiri kayu manis sebesar 5% memenuhi persyaratan daya sebar, daya lekat, pH dan paling disukai oleh konsumen

    Effects of Co-chemotherapy Ethyl Acetate Fraction of Eurycoma Longifolia Jack Roots and Doxorubicin Against Apoptosis Through Expression P53 Mutant and Bcl-2

    Background : It was found mutations of p53 gene in Breast cancer. Mutant p53 protein caused a decrease in cell apoptosis mechanisms through increased expression of Bcl-2. Breast cancer therapy is commonly used chemotherapy using Doxorubicin. However, effectiveness of the use of this chemotherapeutic agent is limited due to the emergence of side effects and toxic to normal cells. Therefore, it is necessary to develop new drugs for combination of chemotherapy. Eurycoma longifolia Jack roots has the potential as co-chemotherapy of breast cancer and it is not toxic to normal cells. Method : Rats were divided into 5 groups. Each group consisted of four female white Sprague Dawley rats. Group 1 (Normal), group 2 (DMBA 20 mg/kgB.W), group 3 (DMBA +Doxorubicin 1.12 mg/kgB.W), group 4 (DMBA +fractions 100 mg/kgB.W), group 5 (DMBA+Doxorubicin +fractions). All the rats were sacrificed at weeks 16 and to be taken their breast tissue. Immunohistochemistry was performed using a mouse monoclonal antibody mutant (BioGenex) and Bcl-2 (BIOSS). Results : expression of mutant p53 percentage obtained for group I (9.35 ± 0.32)%, II (21.65 ± 1.60)%, III (10.72 ± 2.52)%, IV (11.63 ± 3.39)%, V (12.72 ± 3.44)%, While the percentage of Bcl-2 expression obtained for I (20.62 ± 10.09)%, II (52.83 ± 3.61)%, III (24.38 ± 3.54)%, IV (38.01 ± 6.25)%, V (27.99 ± 4.27)%. The data was statistically tested by Kruskal Wallis test (p< 0.005). Conclussion : Co-chemotherapy of E. longifolia Jack roots and Doxorubicin can stimulate apoptosis through decreased in the expression of mutant p53 protein and Bcl-2 in breast tissue of rats induced by DMBA

    Capsule Formulation of Ethanolic Extract of Pasak Bumi (Eurycoma Longifolia Jack.,) and Its Effect on Human Health Vital Signs

    Pasak bumi (Eurycoma longifolia) has the potential to be developed as antihypertensive, antipyretic, aphrodisiacs and health supplements. The use of E. longifolia as a traditional medicine needs to be pursued in the form of more effective and appropriate formulation. The capsule preparations are easy to make and cancover the bitter taste of E. longifolia. Clinical trials in this study use design pre-post treatment in healthy humans. Subjects used were male - healthy men and healthy women who met inclusion criteria and were subjected with formulated capsule for 14 days. The study resulted capsule formula comprising of the ethanolic extract of E. longifolia 300 mg, vivapur 101 300 mg, 58 mg maydis starch, aerosil 3%, talc 2%, and Mg stearate 1%. The results showed that the capsule of E. longifolia did not affect the value of heart rate, respiration rate, body temperature and weight (P > 0.05), based on paired t-test, but they causes a decrease in blood pressure of healthy human. The ethanol extract of E. longifolia caused vasodilation of blood vessels that can be used in antihypertensive therapy
