    Diffusion-weighted imaging: determination of the best pair ofb-values to discriminate breast lesions

    In breast diffusion-weighted imaging (DWI), the apparent diffusion coefficient (ADC) is used to discriminate between malignant and benign lesions. As ADC estimates can be affected by the weighting factors, our goal was to determine the optimal pair of b-values for discriminating breast lesions at 3.0 T. METHODS: 152 females with 157 lesions (89 malignant and 68 benign) underwent breast MRI, including a DWI sequence sampling six b-values 50, 200, 400, 600, 800 and 1000 s mm−2. ADC values were computed from different pairs of b-values and compared with ADC obtained by fitting the six b-values using a mono-exponential diffusion model (ADCall). Cut-off ADC values were determined and diagnostic performance evaluated by receiver operating characteristic analysis using Youden statistics. Mean ADCs were determined for normal tissue and lesions. Differences were evaluated by lesion and histological types. RESULTS: Considering the cut-off values 1.46 and 1.49 × 103mm2 s−1, the pairs 50, 1000 and 200, 800 s mm−2 showed the highest accuracy, 77.5% and 75.4% with areas under the curve 84.4% and 84.2%, respectively. The best pair for ADC quantification was 50, 1000 s mm−2 with 38/49 true-negative and 69/89 true-positive cases respectively; mean ADCs were 1.86 ± 0.46, 1.77 ± 0.37 and 1.15 ± 0.46 × 10−3 mm2 s−1 for normal, benign and malignant lesions. There were no significant differences in these ADC values when compared with ADCall (ADC calculated from the full set of b - values) [difference = 0.0075 × 10−3 mm2 s−1; confidence interval 95%: (−0.0036; 0.0186); p = 0.18].     Piezoelectric and optical response of uniaxially stretched (VDF/TrFE) (75/25) copolymer films

    The phase diagram of the poly(vinylidene fluoride-trifluorethylene) (P(VDF-TrFE)) copolymer system shows for VDF contents of 50...85 mol% a ferroelectric (FE)-paraelectric (PE) phase transition below melting temperature. Investigations on P(VDF-TrFE) 75/25 samples revealed a slight anisotropic behaviour, which leads to a strongly anisotropic stretching effect both on the phase transition and on the amount and nature of the FE phase in samples subjected to mechanical stretching along the main directions of the film. In this work, both the refractive index n1,2 and the piezoelectric coefficient d33 of mechanically stretched P(VDF-TrFE) have been measured for samples with different levels of permanent deformation. These parameters are found to reflect the anisotropy of the permanently deformed samples. The stretching effect is most pronounced (n1,2) or limited (d33) to the vicinity of the yielding point of the material.     Influence of Genetic Polymorphisms in Prostaglandin E2 Pathway (COX-2/HPGD/SLCO2A1/ABCC4) on the Risk for Colorectal Adenoma Development and Recurrence after Polypectomy

    OBJECTIVES: Deregulation of prostaglandin E2 (PGE2) levels reported in colorectal carcinogenesis contributes to key steps of cancer development. Our aim was to evaluate the influence of the genetic variability in COX-2/HPGD/SLCO2A1/ABCC4 PGE2 pathway genes on the development and recurrence of colorectal adenomas. METHODS: A case-control study was conducted gathering 480 unscreened individuals and 195 patients with personal history of adenomas. A total of 43 tagSNPs were characterized using the Sequenom platform or real-time PCR. RESULTS: Ten tagSNPs were identified as susceptibility biomarkers for the development of adenomas. The top three most meaningful tagSNPs include the rs689466 in COX-2 (odds ratio (OR)=3.23; 95% confidence interval (CI): 1.52-6.86), rs6439448 in SLCO2A1 (OR=0.38; 95% CI: 0.22-0.65) and rs1751051 in ABCC4 genes (OR=2.75; 95% CI: 1.58-4.80). The best four-locus gene-gene interaction model included the rs1346271, rs1863642 and rs12500316 single nucleotide polymorphisms in HPGD and rs1678405 in ABCC4 genes and was associated with a 13-fold increased susceptibility (95% CI: 3.84-46.3, P<0.0001, cross-validation (CV) accuracy: 0.78 and CV consistency: 8/10). Interesting, in low-risk patients the ABCC4 rs9524821AA genotype was associated not only with a higher hazard ratio (HR=2.93; 95% CI: 1.07-8.03), but half of these patients had adenoma recurrence at 60 months, considerably higher than the 21% noticed in low-risk patients. CONCLUSIONS: Genetic polymorphisms in COX-2/PGE2 pathway appear to contribute to the development of colorectal adenomas and influence the interval time to adenomas recurrence. The definition of risk models through the inclusion of genetic biomarkers might improve the adherence and optimization of current screening and surveillance guidelines for colorectal cancer prevention.Financial support: This study was supported by a research grant from the Instituto Português de Oncologia do Porto— Centro de Investigação.     AÇÃO ANTIFÚNGICA DE UM JATO DE PLASMA NÃO-TERMICO DE HÉLIO/AR COMPRIMIDO SOBRE BIOFILMES DE CANDIDA ALBICANS

    Os biofilmes fúngicos são uma predominante causa de infecções crônicas associadas à utilização de cateteres e próteses, conferindo resistência aos antibióticos e fatores imunológicos do hospedeiro, sendo as leveduras, do gênero Candida spp, as mais frequentemente isoladas. O plasma não-térmico, operado à pressão atmosférica, vem ganhando destaque como uma nova estratégia antimicrobiana, inclusive para erradicação de biofilmes. Este trabalho teve como objetivo avaliar a eficácia da inativação de biofilmes de Candida albicans ATCC sobre substrato de poliuretano, utilizando jatos de plasma atmosférico de 6L/min de hélio e 4L/min de ar comprimido, alternando entre sistema contínuo e pulsado, com frequência de 60Hz e distâncias de 10 a 30mm entre o bocal e substrato. Após tratamento, realizou-se a contagem das unidades formadoras de colônia e a análise morfológica da superfície do biofilme por Microscopia Eletrônica de Varredura. O melhor grupo foi o plasma de modo pulsado com distância de 30mm com redução de 92% das unidades formadoras de colônia, demonstrando ser uma tecnologia promissora para o controle de biofilmes de C. albicans
